参芎化瘀胶囊对血管性痴呆大鼠海马CA1区BDNF表达的影响

         

摘要

Objective To study the effects of Shenxionghuayu capsule on the brain-derived neurotrophic factor ( BDNF) in CA1 area of hippocamps of vascular dementia rats. Methods Established vascular dementia rats models in common carotid artery repeatedly occlusion and sodium nitroprusside intraperitoneal injection. The learning and memory abilities of rats were detected by Morris water maze. The changes of cell morphology were observed by HE stain and the express of BDNF was detected by immunohistochemistry. Results The learning and memory abilities of rats in VD model group were significantly declined, and the expressions of BDNF in the hippocampus significantly increased compared with those in the sham-operation group and blank control group (all P <0.01 ). The learning and memory abilities of rats in Shenxionghuayu capsule intervention group were significantly improved, and the expressions of BDNF in the hippocampus significantly increased compared with those in the VD model group (P<0.05 or <0.01). Conclusions Shenxionghuayu capsule can improve the abilitiea of learning and memory of the VD rats by increasing the express of BDNF.%目的 探讨参芎化瘀胶囊对血管性痴呆(VD)模型大鼠海马CA1区脑源性神经营养因子(BDNF)表达的影响.方法 将40只大鼠随机分为正常组、假手术组、模型组及参芎化瘀胶囊治疗组.采用反复夹闭双侧颈总动脉加腹腔注射硝普钠法,制备VD大鼠模型.应用Morris水迷宫试验检测VD大鼠的学习、记忆能力,HE染色观察大鼠海马CA1区神经元形态变化,免疫组化法检测BDNF表达.结果 与正常组及假手术组比较,模型组出现明显学习、记忆障碍,海马区BDNF阳性细胞增多(P均<0.01);与模型组比较,治疗组学习记忆障碍明显改善、海马区BDNF阳性细胞增多(P<0.05或<0.01).结论 参芎化瘀胶囊能改善VD大鼠的学习、记忆能力,其机制可能与参芎化瘀胶囊上调BDNF表达有关.
