异硫氰酸苄酯对脑胶质瘤 U-87 MG细胞周期的影响及其机制

         

摘要

Objective To investigate the effect and mechanism of benzyl isothiocyanate ( BITC) on arresting cell cy-cle of human glioma cell line U-87 MG.Methods U-87 MG cells in the logarithmic phase were treated with 2 and 5μmol/L BITC.The cell cycle distribution was measured by flow cytometry , the expression of cell cycle related proteins Cy-clin B1, Cdc2, p21 and Cdc25C, and the phosphorylation of Akt was measured by Western blotting and RT-PCR, the tran-scriptional activity of NF-κB, HIF and eIF4E was determined by reporter gene system .Results After BITC treatment for 24 h, the proportion of cells in the G2/M phase of cell cycle was increased, and the protein expression of Cyclin B1, Cdc2, Cdc25C and p-Akt was down-regulated, but in contrast p21 was elevated; the mRNA expression of Cyclin B1, Cdc2, Cdc25C and p21 was down-regulated, and the transcriptional activities of NF-κB, HIF and eIF4E were significantly de-creased (all P<0.05).Conclusions BITC is able to arrest cell cycle of human glioma cell line U-87 MG.The mecha-nism may be related to the inhibition of Akt/NF-κB pathway and thus regulating cell cycle related gene expression , which provides a new idea for the target treatment of glioma cell cycle .%目的:观察异硫氰酸苄酯( BITC)对人脑胶质瘤细胞系U-87 MG细胞周期的阻滞作用,并探讨其机制。方法取对数期U-87 MG细胞分别以2、5μmol/L的BITC处理,应用流式细胞仪检测细胞周期分布,Western blot-ting法检测细胞周期相关蛋白Cyclin B1、Cdc2、p21、Cdc25C及磷酸化Akt(p-Akt)蛋白表达,RT-PCR法检测Cyclin B1、Cdc2、p21和Cdc25C的mRNA表达,报告基因技术检测NF-κB、缺氧诱导因子( HIF)和真核细胞翻译起始因子4E(eIF4E)的转录活性。结果 BITC作用24 h后,U-87 MG细胞周期G2/M期比例显著升高,Cyclin B1、Cdc2、Cdc25C、p-Akt蛋白表达下调,p21蛋白表达上升,Cyclin B1、Cdc2、Cdc25C、p21 mRNA表达均显著下降,NF-κB、HIF、eIF4E转录活性均显著下降(P均<0.05)。结论 BITC对U-87 MG细胞周期有阻滞作用,机制可能与抑制Akt/NF-κB信号转导路径,进而调节细胞周期相关基因表达有关;此为胶质瘤的细胞周期靶点治疗提供了新思路。
