西黄丸对肾母细胞瘤增殖、凋亡的影响及机制探讨

         

摘要

目的 观察西黄丸对肾母细胞瘤细胞增殖、凋亡的影响,并探讨其作用机制.方法 收集临床确诊为肾母细胞瘤患者的术中癌组织切块,进行原代培养,取处于对数生长期的肾母细胞瘤细胞,分为高、中、低浓度组及对照组.高、中、低浓度组分别加入4、2、1 mg/mL的西黄丸浸提液,对照组加入相同剂量的培养基.采用MTS法测定各组细胞增殖抑制率,流式细胞仪测定各组细胞瘤凋亡率,Western blotting法检测各组细胞中B细胞淋巴瘤因子-2(Bcl-2)和Bcl-2相关X蛋白(Bax)的表达.结果 高、中、低浓度组细胞增殖抑制率均高于对照组,且浓度较高组的细胞增殖抑制率较高(P均<0.05).高、中、低浓度组细胞凋亡率均高于对照组,且浓度较高组的细胞凋亡率较高(P均<0.05).高、中、低浓度组Bax蛋白相对表达量均高于对照组,且浓度较高组的Bax蛋白相对表达量较高(P均<0.05).高、中、低浓度组Bcl-2蛋白相对表达量均低于对照组,且浓度较高组的Bcl-2蛋白相对表达量较低(P均<0.05).结论 西黄丸可以抑制肾母细胞瘤的增殖并诱导其凋亡,其作用机制可能与促进Bax蛋白表达、抑制Bcl-2蛋白表达有关.%Objective To observe the effects of Xihuang pills on the proliferation and apoptosis of Wilms tumor cells, and to explore its mechanism. Methods We collected the cancer tissues from the diagnosed cancer patients to perform primitive culture. Then,we selected the cells in the logarithmic growth phase to divide them into the high-dose,medium-dose and low-dose groups and the control group which were separately added with 4,2,and 1 mg/ mL water extracts of Xi-huang pills,and the same dose of medium. The proliferation inhibition rate was detected by MTS,the rate of apoptosis was detected by flow cytometry,and the expression of Bax and Bcl-2 proteins was detected by Western blotting. Results The proliferation inhibition rates of the high-dose group,medium-dose group,and low-dose group were higher than that of the control group,and the proliferation inhibition rate of the higher dose group was much higher than that of lower dose group (all P < 0. 05). The apoptosis rates of the high-dose group,medium-dose group,and low-dose group were higher than that of the control group,and the apoptosis rate of the higher dose group was much higher than that of lower dose group (all P< 0. 05). Western blotting showed that the relative expression of Bax was in the following order:high-dose group > medi-um-dose group > low-dose group > control group,all P < 0. 05;and the relative expression of Bcl-2 was in the opposite or-der:high-dose group < medium-dose group < low-dose group < control group,all P < 0. 05. Conclusion Xihuang pills can inhibit the proliferation of Wilms tumor cells and induce its apoptosis,and the mechanism may be related with the up-regulated expression of Bax and down-regulated expression of Bcl-2.
