顺铂联合1,25(OH)2D3对肺癌细胞凋亡及VEGF、IL-6表达水平的影响

摘要

目的:探讨顺铂联合1,25(OH)2D3对肺癌细胞凋亡及VEGF、IL-6表达水平的影响.方法:体外培养恶性胸水中肺癌细胞,经药物作用后CCK-8法测定细胞抑制率,流式细胞仪检测细胞周期和凋亡,应用化学发光仪检测细胞培养液中VEGF、IL-6表达水平.结果:不同浓度的1,25(OH)2D3、顺铂单独作用于肺癌细胞均有明显的抑制作用,两药联合表现为协同作用,与单药相比具有显著性差异(P＜0. 05).流式细胞分析显示,经药物联合处理后的肺癌细胞的 G0/G1期细胞数增多,诱导细胞凋亡(P ＜0. 05 ).顺铂、1,25 (OH)2D3对VEGF的表达均有抑制作用,顺铂能够上调IL-6水平,1,25(OH)2D3对IL-6表达无显著作用.两药联合作用时能抑制 VEGF、IL -6的表达,且较单药显著增强(P ＜0. 05).结论:顺铂联合1,25 (OH)2D3能够抑制肺癌细胞生长,诱导细胞凋亡,降低了VEGF、IL-6的表达水平,可为肺癌提供新的治疗策略.%Objective:To investigate the effect of cisplatin combined with 1,25(OH)2D3on the apoptosis and the expression of VEGF and IL-6 in lung cancer cells. Methods:Lung cancer cells from malignant pleural effusion were cultured in vitro. The inhibition rate of tumor cells was measured by CCK-8 assay. The cell cycle and apoptosis were detected by flow cytometry. The expression level of VEGF and IL-6 in cell culture medium was detected by chemilu-minescence. Results:Different concentrations of 1,25(OH)2D3and cisplatin alone had obvious inhibitory effects on lung cancer cells,which increased with time and concentration increased (P＜0. 05). The combination of the two drugs showed synergistic effect,which was significantly different from that of single drug (P＜0. 05). Cell cycle anal-ysis showed that the number of G0/G1cells increased in the lung cancer cells treated by drug treatment and decreased in S and G2/M stages. DDP,1,25(OH)2D3inhibited the expression of VEGF,DDP could decrease the level of IL-6,and 1,25(OH)2D3had no significant effect on IL-6 expression. The combination of DDP and 1,25(OH)2D3sig-nificantly induced the expression of VEGF and IL-6,and significantly enhanced (P＜0. 05)compared with the sin-gle drug. Conclusion:DDP combined with 1,25(OH)2D3can inhibit the growth of lung cancer cells,induce apoptosis and reduce the expression level of VEGF and IL-6,can provide a new therapeutic strategy for lung cancer.
