应用组织芯片技术检测HIF-1α、NOS2、VEGF在胃肠道间质瘤中的表达及临床意义

         

摘要

目的:检测HIF-1α、NOS2、VEGF在胃肠道间质瘤(GIST)中的表达及其与临床病理行为的关系.方法:收集124例GIST标本,用组织芯片和免疫组织化学法检测HIF-1α、NOS2、VEGF表达.结果:在124例GIST中,HIF-1α阳性表达79例(63.7%);NOS2阳性表达80例(64.5%);VEGF阳性表达72例(58.1%),其表达与GIST侵袭转移、NIH分级、黏膜受侵有相关性(P<0.05),三者之间呈正相关(P<0.05).结论:HIF-1α、NOS2、VEGF在GIST组织中高表达,这为揭示GIST的发生发展提供了新的理论依据,并为GIST有效治疗提供新思路.应用组织芯片大规模高效检测临床样本是可行的,具有快速、准确、方便、经济的特点.%Objective :To investigate the expression and significance of HIF - 1α, NOS2 , VEGF in human GIST. Methods : Total of 124 GIST specimens and their normal counterparts were detected for protein expression of HIF 1α,NOS2,VEGF. Results: The positive expression of HIF - 1αin 124 cases of GIST was found in 79 cases ( 63. 7% ), positive expression of NOS2 was found 80 cases ( 64. 5% ), positive expression of VEGF was found in 72 cases ( 58. 1% ). The expression was closely related to NIH classification, mucosal invasion and metastasis. The positive correlation was found between the 3 genes. Conclusion : The expression of HIF - 1α , NOS2 ,VEGF is up - regulated in GIST and they may play an important role in the genesis and development of GIST and find effective the rapeutic method. It is feasible to use tissue micro - array for a rapid screen of massive tissue specimens clinically and it is an high efficacy, economic and accurate method for detecting clinical specimens.
