CD137L突变体在咽喉癌及癌旁组织的表达及意义

摘要

目的：探讨咽喉癌CD137L两种突变体的表达水平和临床意义。方法：对70例咽喉癌组织芯片，通过免疫组织化学的方法，采用不同抗体，测定其CD137L N端和C端表达情况。Image-Pro Plus软件中的Pa-thology批处理程序计算各病例的IOD值。结果：咽喉癌病理组织中，CD137L N端表达阳性占87．7％，C端表达阳性占78．5％。喉癌及癌旁组织之间相比均有显著差异（P＜0．01）。其表达情况与年龄、性别及肿瘤生长部位无显著相关性，与病理类型和肿瘤分级有一定相关性。在不同的咽喉癌患者，两种抗体检测的IOD值差值显著不同。结论：CD137L突变体的表达在不同患者呈现不同特点，其发生剪接突变的比率和机制不同。%Objective:Two distinct antibodies were used to detect CD137L expression in laryngeal carcinoma and its clinical significance.Methods:Immunohistochemistry was performed in tissue microarrays composed of 80 cases of laryngeal carcinoma using two anti-CD137L antibodies raised against the C-terminus and N-terminus of CD137L, respectively.Integrated optical density (IOD)of immunoreactivity was quantified using Image-Pro plus software.Re-sults:The CD137L N -terminal positive rate was higher than that of adjacent normal tissues (P<0.01).It was 87.7% and 78.5%,respectively in laryngeal carcinoma.There was no significant correlation was observed between the CD137L expression and the age,sex or tumor location.However,there was some relevance in tumor histological type and tumor grade.Conclusion:The mutation of CD137L expression showed different characteristics in different patients.The ratio and mechanism of CD137L mutation is different.
