目的:探讨热疗联合紫杉醇(Paclitaxel)对卵巢癌 A2780细胞增殖、侵袭及 AKT 磷酸化的影响。方法:将 A2780细胞分为4组:热疗组(41℃加热90min)、紫杉醇组、热疗联合紫杉醇组及对照组(常规培养细胞)。用 MTT 法检测细胞增殖情况,测定紫杉醇联合热疗对 A2780细胞体外增殖抑制的影响;Transwell 侵袭实验检测细胞的侵袭能力的变化,测定紫杉醇联合热疗对 A2780细胞体外侵袭能力的影响;Western blot 检测各组细胞中 AKT 磷酸化的变化。结果:热疗联合紫杉醇组细胞增殖率与其他各组比较均明显降低,差异有统计学意义(P <0.05);热疗联合紫杉醇组细胞的侵袭能力同样明显减弱,差异有统计学意义(P <0.05);同时,热疗联合紫杉醇组细胞中 AKT 磷酸化水平与其他各组相比亦显著降低,差异有统计学意义(P <0.05)。结论:热疗联合紫杉醇可通过降低 A2780细胞中 AKT 的磷酸化水平从而明显抑制细胞的增殖及侵袭能力。%Objective:To examine the effect of hyperthermia and Paclitaxel combination on the proliferation,inva-sion and AKT phosphorylation levels in ovarian cancer A2780 cell line.Methods:The A2780 cells were divided into 4 groups:Hyperthermia group(41℃,90min),Paclitaxel group,hyperthermia and Paclitaxel group,control group.The cell proliferation rate of A2780 cell lines was detected by MTT assay to evaluate the proliferation of A2780 cell line. Transwell experimental was used to detect the invasion ability of A2780 cell line.Western blot was used to detect AKT phosphorylation levels in ovarian cancer A2780 cell line.Results:Compared with other group,the cell proliferation of Paclitaxel and hyperthermia group decreased significantly(P <0.05).The invasion ability of A2780 cell in Paclitaxel and hyperthermia group decreased significantly compared with other groups(P <0.05),and the AKT phosphorylation level of Paclitaxel and hyperthermia group was inhibited significantly.Conclusion:Paclitaxel and hyperthermia group has obvious inhibitive effect on A2780 cell line AKT phosphorylation to inhibit cells proliferation and invasion ability.
