Fibulin-2,a member of Fibulin gene family,is encoded by the FBLN2 in the human body.Fibulin-2 is an extracellular matrix protein of which is associated with basement membranes and elastic extracellular matrix fibres.Fibulin-2 plays an essential role in cell adhesion and migration through its association with other ECM protein and cell surface receptors such as integrins and syndecans.Mutations in Fibulins result in human disease.Observations with data in human cancer tissue suggest that the Fibulin-2 plays dual roles with the promotion and inhibition of tumorigenesis and development.Fibulin-2 inhibits tumor migration and invasion in nasopharyngeal carcinoma,breast cancer,childhood acute lymphoblastic leukemia and Kaposi's sarcoma.However,Fibulin-2 is a driver of malignant progression in lung adenocarcinoma cell metastatic.This review summerizes the progress of Fibulin-2'function and mechanism of action in tumor.%Fibulin-2是FBLN2基因编码的细胞外基质蛋白Fibulin家族一员,是细胞外基质、基底膜和弹性基底纤维的重要组成部分.Fibulin-2与细胞基质蛋白及整合素等表面受体相互作用而在细胞吸附和迁移中扮演重要作用.Fibulin-2基因发生变异往往会导致人类疾病.根据在肿瘤组织中大量研究数据发现Fibulin-2在肿瘤的发生发展中扮演了促肿瘤或抑肿瘤双刃剑的作用,Fibulin-2在鼻咽癌、乳腺癌、儿童急性淋巴细胞白血病和卡波西肉瘤中能够抑制肿瘤的迁移和侵袭;相反,Fibulin-2却能够促进肺腺癌中细胞的迁移和侵袭.本文就Fibulin-2在肿瘤中的表达、功能及作用机制等研究进展作一综述.
