Objective To observation the expression of Toll-like receptor mRNA, composition of intestinal flora, bacterial translocation and their relationship in rats with antibiotic induced diarrhea. Methods The dynamic changes of expression of Toll-like receptor mRNA were detected by reverse transcription polymerase chain reaction ( RT-PCR ) in mesenteric lymph nodes, liver, spleen and intestinal tissues. The number of intestinal Bifidobacterium, Lactobacillus, Enterobacteriaceae, Enterococci, and the number of translocating bacterial in mesenteric lymph nodes, liver and spleen were determined by bacterial culture. Results In the early stage of diarrhea induced by antibiotics, the expression of Tolllike receptor mRNA was inhibited, the imbalance and translocation of intestinal flora was observed. The bacterial translocations were different in mesenteric lymph nodes, liver and spleen. With the interference of Lactobacillus, the inhibited expression of Toll-like receptor mRNA and bacterial translocation were recovered. Meanwhile with the attack of Enterococci, inhibited expression of Toll-like receptor mRNA and bacterial translocation were further deterioration. Conclusions Antibiotic could induce diarrhea, inhibit expression of Toll-like receptor mRNA, lead to gut microflora imbalance and bacterial translocation. Early intervention with Lactobacillus may provide protective effect.%目的 观察抗生素所致腹泻大鼠体内Toll受体mRNA转录水平、肠菌群构成和细菌移位现象的变化,阐明相互间关系.方法 60只SD大鼠分为A组(盐酸林可霉素组)、B组(盐酸林可霉素加服大肠杆菌组)、C组(盐酸林可霉素加服乳酸杆菌)和D组(对照组).采用反转录聚合酶链反应(RT-PCR)动态测定大鼠肠系膜淋巴结、肝脏、脾脏和肠黏膜组织中Toll样受体mRNA转录水平;采用细菌培养法动态测定抗生素所致腹泻大鼠肠道双歧杆菌、乳酸杆菌、肠杆菌和肠球菌及肠系膜淋巴结、肝脏和脾脏组织移位细菌量.结果 应用抗生素大鼠早期发生腹泻,体内Toll样受体mRNA转录水平受抑制,出现肠道菌群失调和肠细菌移位,肠系膜淋巴结、肠黏膜、肝脏和脾脏组织细菌移位状况不同.乳酸杆菌干预可减轻由抗生素引起的Toll样受体mRNA转录受抑及细菌移位程度;大肠杆菌攻击可上调Toll样受体mRNA转录水平,加剧细菌移位程度.结论 抗生素可致大鼠腹泻,抑制体内Toll样受体mRNA表达,导致肠道微生态失调和肠细菌移位.乳酸杆菌可保护天然免疫识别机制,维护肠道微生态平衡和降低细菌移位发生程度.
