热休克蛋白在人舌癌细胞株和癌前病变细胞株的差异表达

         

摘要

目的 检测热休克蛋白(HSPs)27、70、90α在舌癌细胞株及癌前细胞株的表达情况.方法 体外培养人舌癌细胞株SCC-15,SCC-6和癌前细胞株DOK.采用RT-PCR测定人舌癌细胞株SCC-15、SCC-6及癌前细胞株DOK中HSP27、HSP70、HSP90α的mRNA表达情况,并作对比分析.采用Western Blot方法检测人舌癌细胞株SCC-15、SCC-6和癌前病变细胞株DOK中的HSP27、HSP70、HSP90α的蛋白表达情况,并作对比分析.结果 HSP27、HSP70、HSP90α在舌癌细胞株中的mRNA表达水平和蛋白表达量均明显高于癌前细胞株,差异具有统计学意义(P<0.05).结论 热休克蛋白在舌癌细胞株中的表达明显高于癌前细胞株,热休克蛋白的高表达可能与舌癌的发生发展有关.%Objective To examine the expression of heat shock proteins ( HSP) 27,70 and 90a in tongue carcinoma cell lines and precancerous cell lines. Methods Human tongue carcinoma cell lines SCC-15 and SCC-6 and precancerous cell lines DOK were cultured in vitro. The rnRNA expression of HSP27, 70 and 90a in tongue carcinoma cell lines and precancerous cell lines was detected by RT-PCR . The protein expression of HSP27, 70 and 90a in tongue carcinoma cell lines and precancerous cell lines was measured by Western blotting. Results The mRNA and the protein expression levels of HSP27,70 and 90a in tongue carcinoma cell lines were higher than those in precancerous cell lines(P <0. 05). Conclusion The expression of HSP 27, 70 and 90a in tongue carcinoma cell lines was significantly up-regulated compared to their expression in precancerous cell lines. The heat shock proteins may play an important role in careinogenesis of tongue cancer.
