非抗凝猪血提取血红素的工艺优化研究

         

摘要

Objective:To introduce and optimize the extraction technology of heme from the whole blood of swine free of anticoagulation. Methods:Optimization of the extraction technology of heme was performed using L9 (34) orthogonal design in reference to homogenate duration, amount of double-distilled water input, volume of hydrochloric acid acetone solution to hemolysate and the mixing time before extracting. Results: The average production was 10.6 g for per kilogram of the blood, and the purity arrived at 90.8%. The final optimum parameter included homogenate duration of 5 min, equal amount of double-distilled water, ratio of hydrochloric acid acetone solution to hemolysate(5∶1 ) and 10 min of mixture before extraction. Conclusion: It is feasible and simple to extract the heme from the non-anticoagulated whole blood of swine.%目的:建立并优化非抗凝猪血提取血红素的工艺.方法:采用L9(34)正交试验法,从匀浆时间、匀浆后双蒸水加入量、盐酸丙酮溶液与溶血液的体积比(V盐酸丙酮:V溶血液)、搅拌抽提时间4个主要影响因素对非抗凝猪血提取血红素的工艺进行优化.结果:提取血红素的平均产量为10.6 g/kg非抗凝血,纯度为90.8%;确定的最佳提取工艺参数是:匀浆5 min,匀浆后加入等体积双蒸水,V盐酸丙酮:V溶血液=5:1,搅拌抽提10 min.结论:利用非抗凝猪血提取血红素切实可行,工艺简单.
