Rescue of interspecific hybrids of groundnut infected with peanut stripe virus through meristem culture

Meristem culture, either alone or coupled with chemotherapy, finds a special application in the regeneration of virus-free stock of commercial value. The shoot apices of ten interspecific hybrids, apparently showing PStV symptoms, were used for the study. MS medium containing 1 mg.l~(-1) each of NAA and BA supported maximum callusing of the hybrids J 11 X A.sp. PI 30008 as well as J 11 X A.sp. Manfredi # 5 (86.3 and 82.6%, respectively). MS medium with 3.5 mg.l~(-1) NAA and 0.2 mg.l~(-1) BA, the meristems of J 11 X A. stenosperrna as well as J 11 X A. otavioi gave good response (79.7 and 77.2%). The regeneration frequencies ranged from 16.7 to 76.9% and was more than 30% in all hybrids except for J 11 X PI 30085. Five cm long shoots regenerated fromthe virus-free (EL1SA tested) calli rooted well on MS basal medium supplemented with 0.4% activated charcoal and 200 mg.l~(-1) casein hydrolysate. However, long shoots without roots, when transferred gradually to soil during rainy season and protected from sun by shading, got established in the soil, making the hardening procedure easy and fast
