Glycosylation is a major protein modification that modulates a variety of biological functions. It is therefore important to understand their roles in health and disease. Proteins modified by oligosaccharides on asparagine residues (N-linked glycosylation) are most expressed on cell membrane or secreted to the extracellular space. Glycoproteins of specific tissues carry tissuespecific information that can be used as a gauge to survey the status of diseased tissues. However, characterization of glycoproteins faces enormous challenges due to glycan diversity arising from complex nontemplate-based biosynthesis on multiple glycosylation sites of glycoproteins.
