    • 摘要: 目的 探讨研究p27、nm23蛋白在甲状腺癌中的表达与临床意义.方法 收集2017年3月至2018年9月在我院就诊的68例甲状腺癌患者及28例甲状腺良性病变患者临床资料,进行免疫组化SP法检测p27、nm23蛋白在组织中的表达情况.结果 甲状腺良性病变组中p27蛋白的阳性表达率显著高于甲状腺癌组,差异有统计学意义(P<0.05);甲状腺癌组织中p27、nm23蛋白的表达均与其分化程度和被膜侵犯程度有关,p27蛋白表达还与淋巴结转移有一定关联,nm23蛋白表达则与淋巴结转移无关.结论 p27与nm23蛋白表达在甲状腺癌的发病和进展过程中具有一定作用,联合检测可用于早起诊断甲状腺癌以及判断预后.

    • 摘要: 目的:分析散发性结直肠癌癌组织中错配修复蛋白(MMRP)和nm23蛋白表达与临床病理特征的关系以及两者相关性,旨在为治疗散发性结直肠癌患者提供参考.方法:选取2014年8月至2017年1月本院存档的680例手术切除散发性结直肠癌标本.采用免疫组化法以及组织芯片检测MMRP以及mm23蛋白的表达情况,将4种MMRP中的1种以上表达缺失定为微卫星不稳定组(MSI组),全部阳性表达设为微卫星稳定(MSS组)组.分析MMRP与mm23的表达与临床病理特征的相关性以及两种蛋白之间的相关性.结果:在本组研究中,MMRP阳性者有605例,表达缺失者有75例,缺失率为11.03%;其中MLH1缺失11例(14.67%)、PMS2缺失5例(6.67%)、MSH2缺失6例(8.00%)及MSH6表达缺失3例(4.00%),MLH1联合PMS2表达缺失19例(25.33%),MSH2联合MSH6表达缺失12例(16.00%),MLH1联合MSH2表达缺失5例(6.67%),MLH1联合MSH6表达缺失3例(4.00%),MSH2联合PMS2表达缺失3例(4.00%),MLH1联合MSH2、MSH6表达缺失2例(2.67%),MSH2联合MSH6、PMS2表达缺失3例(4.00%),4种表达均缺失3例(4.00%);nm23阴性表达278例(40.88%),阳性表达402例(59.12%);MSI表达情况在不同年龄、肿瘤部位、肿瘤直径、临床分期、淋巴结转移、组织类型等患者间存在差异(P<0.05),nm23表达情况在不同组织类型、远处转移、浸润深度、大体分型等患者间存在差异(P<0.05);经Spearman分析得出,MSI与nm23呈正相关(r=0.563,P<0.001).结论:(1)在散发性结直肠癌患者中存在MMRP缺失,并且MLH1、PMS2缺失表达较MSH2和MSH6多见;(2)MSI及nm23与散发性结直肠临床病理特征关系密切,对散发性结直肠癌的预测以及恶性程度的评估具有参考价值,并且两者呈正相关,可能参与散发性结直肠癌的发生发展过程.

    • 摘要: 目的 研究甲状腺微小乳头状癌(PTMC)组织中B淋巴细胞瘤-2相关X蛋白(Bax)、肿瘤转移抑制基因23(nm23)蛋白的表达及临床意义.方法 选择2016年5月~2017年5月在南通大学附属海安医院接受手术治疗的PTMC患者的组织石蜡标本103例记为观察组,将术中切除的距离肿瘤组织≥5 cm的癌旁组织石蜡标本103例记为对照组,另选同期在医院接受手术治疗的甲状腺腺瘤(TA)患者100例记为TA组,检测并对比各组Bax及nm23蛋白的表达,分析观察组Bax及nm23蛋白的表达与其病理参数的关系,以及Bax与nm23蛋白的相关性.结果 观察组和TA组Bax蛋白的阳性表达率均分别明显高于对照组,差异均有统计学意义(均P<0.05).观察组nm23蛋白的阳性表达率均明显高于TA组和对照组,差异均有统计学意义(均P<0.05).观察组有淋巴结转移的患者Bax及nm23蛋白的阳性表达率均分别明显低于无淋巴结转移者,差异均有统计学意义(均P<0.05).根据Spearman法对相关性分析后发现,Bax与nm23蛋白的表达呈正相关(r=0.617,P<0.05).结论 PTMC组织中Bax和nm23蛋白表达均明显升高,且与淋巴结转移有关,此二者关系紧密,可作为监测靶点,有助于评价PTMC患者的病情及预后.%Objective To study the expression and clinical significance of B lymphocyte tumor associated-2 related X protein (Bax),tumor metastasis suppressor gene 23 (nm23) protein in papillary thyroid microcarcinoma (PTMC) tissues.Methods One hundred and three paraffin specimens of PTMC patients who was given surgery treatment in Haian Affiliated Hospital to Nantong University from May 2016 to May 2017 were selected as observation group,the resection of the tumor was ≥ 5 cm far from paraffin embedded specimens of 103 cases of tumor adjacent tissues selected was as control group,another 100 patients with thyroid adenoma (TA) who was given surgery at the same time were chosen and enrolled as TA group,the expression of Bax and nm23 protein in each group was detected and compared,the relationship between the expression of Bax and nm23 protein and pathological parameters in the observation group was analyzed,and the correlation between Bax and nm23 protein was analyzed.Results The positive expression rates of Bax protein in the observation group and the TA group were significantly higher than the control group,and the differences were statistically significant (P< 0.05).The positive expression rate of nm23 protein in the observation group was significantly higher than the TA group and the control group,and the differences were statistically significant (P < 0.05).The positive expression rates of Bax and nm23 protein in the observation group with lymph node metastasis were significantly lower than without lymph node metastasis,and the differences were statistically significant (P < 0.05).According to the correlation analysis of Spearman,it was found that the expression of Bax was positively correlated with the expression of nm23 protein (r =0.617,P < 0.05).Conclusion The expression of Bax and nm23 protein in PTMC tissues increased significantly,it is closely related to lymph node metastasis and both two indexes can be used as a monitoring target to evaluate the condition and prognosis of PTMC patients.

    • 摘要: 目的 检测恶性淋巴瘤组织中nm23、Survivin蛋白表达情况,并分析其临床意义.方法 选择58例恶性淋巴瘤患者和27例反应增生性淋巴结炎患者,应用免疫组化染色法检测恶性淋巴瘤组织(观察组)和反应增生性淋巴结组织(对照组)中nm23、Survivin蛋白表达情况.结果 观察组nm23、Survivin呈过表达,对照组nm23、Survivin呈低表达(P均<0.01).nm23过表达与B组症状、临床分期高、恶性程度高、结外累计部位数多有关(P均<0.05),Survivin过表达与临床分期高、恶性程度高及乳酸脱氢酶增高有关(P均<0.05).nm23与Survivin表达呈正相关(r=0.323,P<0.05).结论 恶性淋巴瘤组织中nm23蛋白与Survivin蛋白均过表达,且nm23蛋白、Survivin蛋白过表达与恶性淋巴瘤的发生、发展过程密切相关.

    • 摘要: 本研究对白斑综合征病毒(White Spot Syndrome Virus,WSSV)与传染性皮下及造血组织坏死病毒(Infectious Hypodermal and Haematopoietic Necrosis Virus,IHHNV)能否竞争虾鳃细胞膜上的NM23蛋白受体进行探索.先用蔗糖梯度离心法提纯WSSV的全蛋白,利用病毒覆盖蛋白印迹技术(VOPBA)与对虾鳃细胞膜NM23蛋白作用,将疑似蛋白条带进行LC-MS/MS分析,初步筛选出3种WSSV蛋白,分别为WSSV013、Wsv497和Wsv035,再构建这3种蛋白的原核表达载体,通过VOPBA和免疫共沉淀(co-immunoprecipitation)验证了Wsv497、Wsv035能与NM23蛋白相互作用,WSSV013不能与NM23蛋白相互作用.初步推测WSSV与IHHNV可能竞争对虾鳃细胞膜上的NM23蛋白受体,该结果为今后研究两种病毒竞争细胞膜受体和虾病毒蛋白作用机制提供理论基础.%White spot syndrome and infectious hypodermal and haematopoietic necrosis are primary viral diseases of prawns. Outbreaks of white spot syndrome virus (WSSV) and infectious hypodermal and haematopoietic ne-crosis virus (IHHNV) arise worldwide and have caused serious economic losses in recent years, including in China. Some researchers found that, given the same conditions, WSSV-infected prawn display lower levels of mortality than IHHNV-infected prawn. Our laboratory previously demonstrated that IHHNV capsid protein (CP) has the ability to combine with NM23 protein in prawn gill membrane. NM23, a kind of nucleoside diphosphate kinase (NDPK), has various biological functions and is located mainly in the cytoplasm and nucleus, but is also evident in the cell membrane. We conducted an experiment to investigate whether WSSV and IHHNV compete with NM23 protein receptor sites on the cell membrane of shrimp gill tissue. First, we purified the total WSSV proteins using sucrose gradient centrifugation, and then we used a virus overlay protein binding assay (VOPBA) to detect pro-tein-protein interaction between the total proteins and NM23. Next, three suspected positive proteins were selected by the combined LC-MS/MS technique: WSSV013, Wsv497 and Wsv035. Wsv035, also known as VP110, is one of the capsule membrane proteins of WSSV and is encoded byORF035. It contains a membrane structure domain and a RGD locus (namely, the Arg-Gly-Asp motif). Some scholars, using fluorescence assay and competitive in-hibition tests, found Wsv035 could stick to the host cell: Wsv035 RGD can form RGDT peptides at the cell surface and it plays a role in WSSV infection. There have been few studies of Wsv497 and WSSV013 proteins, however, and thus their distribution, structure and functions are not yet clear. Therefore, we constructed prokaryotic expres-sion vectors of these three proteins, for purifying WSSV013, Wsv497 and Wsv035 in order to interact with NM23 protein, using VOPBA and co-immunoprecipitation, respectively. The results showed that Wsv497 and Wsv035 have the ability to interact with NM23, while WSSV013 showed an invalid effect on NM23. Accordingly, we sur-mise that WSSV and IHHNV have the ability to compete with NM23 protein receptors on the cell membrane of prawn gill tissue. This study adds to theories about the mechanism of competition between WSSV and IHHNV for receptor sites on the cell membrane of prawn gill tissue, and lays a foundation for further studies of protein inter-actions in the context of WSSV and prawn culture.

    • 摘要: 目的:探讨nm23蛋白(nm23-H1)在胚胎组织和肺癌组织中的表达.方法:对SD大鼠、人体肺癌组织、人体胚胎组织实施nm23 (nm23-H1)蛋白免疫组化检测.结果:胚胎发育期器官形成以及各种上皮组织的分化与nm23蛋白(nm23-H1)积聚相互一致,对于正常的胚胎组织而言,只有具备了一定的器官结构,发育到了一定的时期,nm23蛋白(nm23-H1)才会稳定积聚.原发瘤灶与转移癌灶nm23蛋白(nm23-H1)阳性率比较P>0.05,且在肺癌原发病灶中,nm23蛋白(nm23-H1)阳性组与nm23蛋白(nm23-H1)阴性组的淋巴结转移率比较P>0.05.结论:肺癌组织、胚胎组织器官样化结构的分化与nm23蛋白(nm23-H1)在肺癌组织和胚胎组织中的积聚具有密切关系,nm23蛋白(nm23-H1)不能直接抑制肺癌细胞的转移,并且nm23蛋白(nm23-H1)对上皮组织结构的稳定具有调节作用.

    • 摘要: 目的 探讨C-erbB-2、P53和nm23蛋白在乳腺癌中的表达及其与乳腺癌预后的关系.方法 收集女性乳腺癌手术标本70例,采用HE切片光镜观察和免疫组化标记分析C-erbB-2、P53和nm23蛋白在乳腺癌中的表达及其意义.结果 在70例乳腺癌中C-erbB-2及P53阳性率有随乳腺癌分级升高而上升的趋势,差异有显著性意义(P<0.05),而nm23蛋白则相反;且P53在淋巴结转移者阳性率高,在无淋巴结转移者阳性率低,两者有显著性差异(P<0.01),而nm23蛋白则与之相反.结论 C-erbB-2与P53为判断乳腺癌预后不良的重要参考指标,而nm23蛋白为乳腺癌预后好的一个重要参考指标.

    • 摘要: 目的:探讨胃癌组织中p27、nm23蛋白的表达及其临床意义.方法:采用免疫组织化学法检测20例正常胃黏膜、58例胃癌组织中p27、nm23的表达,分析p27、nm23蛋白表达与患者年龄、组织学分级、临床分期、淋巴结转移的关系.结果:p27、nm23蛋白在胃癌组织中的表达水平低于胃正常黏膜中的表达,p27蛋白在正常胃黏膜、胃癌组织中阳性表达率分别为85.0%、37.9%,差异有统计学意义(P<0.05);nm23蛋白在正常胃黏膜、胃癌组织中阳性表达率分别为80.0%、34.5%,差异有统计学意义(P<0.05).p27、nm23蛋白的表达与患者年龄无关(P>0.05),与胃癌组织分化程度和浸润深度相关(P<0.05);p27蛋白的表达还与淋巴结转移相关(P<0.05).结论:p27、nm23蛋白表达水平与胃癌的进展及恶性程度相关,p27蛋白表达水平降低可能出现淋巴结转移.p27、nm23蛋白有望成为判断胃癌恶性程度及评价预后的指标.%Objective: To investigate the expression and the clinical significance of p27 protein and nm23 protein in human stomach cancer (GC). Methods: p27 protein and nm23 protein from 20 cases of normal gastric mucosa tissue and 58 cases stomach cancer were detected by immunohistochemical assay. The relationship between p27, nm23 protein expression and patient age, histological grade, clinical stage or lymph node metastasis was analyzed. Results: (1) The positive expression rates of p27, nm23 protein in stomach cancer tissue was lower than that in normal gastric mucosa. The positive expression rates of p27 were 85.0% and 379% in stomach cancer and normal gastric mucosa, respectively; The rates of nm23 were 80.0% and 34.5%. The difference was statistically significant (P<0 05), (2)The expression of p27. nm23 protein in stomach cancer was correlated with hist ological grade, clincal stage (P<0.05). But it was independent with patient age(P>0.05);meanwhile, the expression of p27 protein in stomach cancer was correlated with lymph node metastasis (P<0.05) Conclusion: The level of expression in p27、nm23 protein is related with the level of malignance and progression of stomach cancer. The expression of p27 protein is associated with lymph node metastasis in stomach cancer.

    • 摘要: 目的 探讨nm23与PCNA在鼻咽癌中的表达与鼻咽癌转移的关系.方法 应用免疫组织化学S-P法检测66例鼻咽癌标本中nm23蛋白和PCNA在鼻咽癌中的表达.结果 鼻咽癌患者中的nm23蛋白和PCNA的表达高于慢性鼻咽炎的.nm23低表达与PCNA的高表达均和鼻咽癌转移有关(P均<0.05).结论 nm23低表达和PCNA 高表达具有促进鼻咽癌转移的作用.

    • 摘要: 目的:研究nm23在非小细胞肺癌患者的具体表达意义.方法:对患者的手术切除标本使用S-P免疫组化检查,检查两组患者中nm23蛋白表达水平.结果:nm23蛋白在NSCLC组织阳性表达与患者年龄、性别、组织学类型均无关(P>0.05).nm23的表达与肿瘤的组织分化程度无关(P>0.05),与肿瘤的临床分期、淋巴结转移和患者的生存期有明显相关性(P<0.05).结论:nm23在非小细胞肺癌(NSCLC)组织中均异常表达,与NSCLC的发生发展密切相关,可作为NSCLC诊断的标志物.nm23在NSCLC的发生发展中起协同作用,检测nm23表达对于临床选择高危复发病例,给予积极治疗,提高预后水平具有重要意义,并可作为建立NSCLC预后模型的重要指标.
