    • 摘要: Accumulating evidence indicates that ataxia-telangiectasia mutated kinase is critical for maintaining cellular homeostasis and that it has both nuclear and cytoplasmic functions.However,the functions of ataxia-telangiectasia mutated that when lost lead to cerebellar degeneration are still unknown.In this review,we first describe the role of ataxia-telangiectasia mutated in cerebellar pathology.In addition to its canonical nuclear functions in DNA damage response circuits,ataxia-telangiectasia mutated functions in various cytoplasmic and mitochondrial processes that are critically important for cellular homeostasis.We discuss these functions with a focus on the role of ataxia-telangiectasia mutated in maintaining the homeostatic redox state.Finally,we describe the unique functions of ataxia-telangiectasia mutated in various types of neuronal and glial cells including cerebellar granule neurons,astrocytes,and microglial cells.

    • 摘要: Objective We aimed to observe the radiosensitization effect of mir-30a-5p in a nude mouse model with subcutaneous lung-cancer xenograft and to explore the underlying mechanism.Methods A549 cell lines with either stable upregulation or downregulation of mir-30a-5p,and their negative control,were transfected with lentivirus vectors.These cell lines were used to establish a nude mouse model with subcutaneous lung-cancer xenograft.Each group was randomly divided into irradiated and non-irradiated groups.The radiosensitization effect of mir-30a-5p in vivo was studied by observing xenograft growth trends and tumor weight.The mechanisms involved in this radiosensitization were investigated by detecting expressed radiosensitization-related proteins,using immunohistochemistry and Western blotting.Results The expression level of mir-30a-5p in the lenti-mir-30a-5p group was higher than that in the negative control(lenti-GFP)group and lower in the lenti-inhibitor group(P<0.05).Subcutaneous lung-cancer xenografts in the irradiation group and lenti-mir-30a-5p increased in size slowly;tumors were lighter and tumor inhibition rates were higher than those in the non-irradiation and lenti-GFP groups.In contrast,the opposite of these effects was observed in the lenti-inhibitor group.Immunohistochemistry and Western blotting indicated that ATM protein expression level was lower in the lenti-mir-30a-5p group,with or without irradiation,compared to that in the lenti-GFP group.ATM protein levels were higher in the lenti-inhibitor groups.The phosphorylation level of ATM at residue 1981 was low in the groups without irradiation and increased significantly after irradiation(P<0.05).Moreover,the phosphorylation level was lower in the lenti-mir-30a-5p group and higher in the lenti-inhibitor group than that in the lenti-GFP group after irradiation(P<0.05).Conclusion Mir-30a-5p enhanced the radiosensitivity of nude mice with subcutaneous lung-cancer xenografts by inhibiting ATM phosphorylation.

    • 摘要: 目的分析驱动蛋白家族4A(KIF4A)在子宫内膜癌组织中的表达情况,探索KIF4A参与子宫内膜癌的潜在机制。方法利用网络数据库中子宫内膜癌相关芯片数据及测序数据分析KIF4A的表达水平。通过WebGestalt基因富集分析在线工具预测KIF4A相关差异基因在子宫内膜癌中参与的生物过程及通路,探索KIF4A参与子宫内膜癌发生、发展的机制。纳入60例子宫内膜样腺癌及30例正常子宫内膜石蜡组织标本,利用免疫组织化学实验分析上述两类组织中KIF4A表达水平,并分析KIF4A表达与子宫内膜癌临床病理参数的关系。结果京都基因与基因组百科全书(KEGG)、Wiki通路富集分析结果显示,在子宫内膜癌组织中,KIF4A共表达基因主要参与的生物过程或通路包括ATM信号通路、ATR介导的DNA损伤与细胞反应、范可尼贫血通路。子宫内膜癌组织中的KIF4A阳性表达率显著高于正常子宫内膜组织(P=0.001)。KIF4A表达与子宫内膜癌分级相关(P=0.037)。结论高表达的KIF4A可能通过依赖ATM/ATR通路介导的磷酸化修饰参与细胞周期调控,在子宫内膜癌的发生、发展中发挥重要作用。

    • 摘要: 目的 探讨长链非编码RNA(lncRNA)ANRIL负反馈调控ATM-E2F1信号通路在胆囊癌上皮间质转化(EMT)中的作用及机制.方法 选择人正常胆囊上皮细胞和胆囊癌GBC-SD、NOZ细胞,构建过表达ANRIL质粒、过表达ATM载体及oe-ATM+oe-ANRIL共转染GBC-SD细胞;RT-qPCR检测ANRIL及各组ATM-E2F1信号通路中ATM、E2F1、INK4A、INK4B和ARF的mRNA表达;Western blotting检测各组EMT标志物(E-cadherin、N-cadherin、Vimentin)及ATM-E2F1信号通路蛋白(ATM、E2F1、INK4A、INK4B、ARF)的表达;采用小分子药物抑制ATM-E2F1信号通路调控的胆囊癌细胞EMT过程;Transwell实验检测细胞侵袭能力;划痕实验检测细胞迁移能力;MTT检测细胞增殖活力.结果 胆囊癌细胞中ANRIL的表达水平显著高于正常胆囊上皮细胞(P<0.05).过表达ANRIL可抑制胆囊癌细胞中INK4B、INK4A、ARF的表达,导致上皮性标志物E-cadherin表达下调,而间质性标志物N-cadherin、Vimentin表达上调,差异均有统计学意义(P<0.05).过表达ATM可促进抑癌基因INK4B、INK4A、ARF的表达,导致E-cadherin表达上调,N-cadherin、Vimentin表达下调,差异均有统计学意义(P<0.05).使用ATM激酶抑制剂CGK733处理ATM过表达的胆囊癌细胞后,E-cadherin表达下调,N-cadherin、Vimentin表达上调,差异均有统计学意义(P<0.05).过表达ANRIL或抑制ATM-E2F1信号通路能够促进胆囊癌细胞的增殖、迁移和侵袭.结论 lncRNA ARNIL在胆囊癌中呈高表达,并依赖于ATM-E2F1信号通路促进胆囊癌细胞发生EMT,提示在治疗胆囊癌时可通过靶向沉默ANRIL或激活ATM-E2F1信号通路来抑制胆囊癌细胞发生EMT,从而提高其临床疗效.

    • 摘要: 目的 探讨鼻咽癌细胞HONE1中抑制DNA修复的潜在机制.方法 使用ETP处理鼻咽癌细胞HONE1造成DNA损伤后,通过高通量测序检测鼻咽癌细胞HONE1中miRNA的表达谱.过表达差异miRNA后,通过基于I-PpoI的可诱导DSB系统检测DNA双链断裂处(DNA double-strand breaks,DSB)的DNA-PKcs含量.通过miRDB在线分析miRNA的潜在靶标.通过荧光素酶报告系统检测miRNA靶向mRNA.结果 ETP处理后hsa-miR-9983-3p、hsa-miR-4731-5p、hsa-miR-3675-5p、hsa-miR-516b-5p、hsa-miR-4437、hsa-miR-7114-5p、hsa-miR-637、hsa-miR-5008-5p的表达水平上升至少8倍.miR-516b-5p mimics导致DSB的DNA-PKcs含量减少(P<0.05),而miR-516b-5p inhibitor导致DSB的DNA-PKcs含量增加(P<0.05).在鼻咽癌细胞HONE1中过表达miR-516b-5p后,ATM mRNA的表达水平和蛋白表达水平下降,DNA-PKcs的磷酸化水平下降;而抑制miR-516b-5p表达后,ATM mRNA的表达水平和蛋白表达水平上升,DNA-PKcs的磷酸化水平上升.同时,miR-516b-5p靶向ATM的3端非编码区.过表达ATM后,DSB的DNA-PKcs含量上升(P<0.05),而敲低ATM后,DSB的DNA-PKcs含量下降(P<0.05).结论 miR-516b-5p通过靶向ATM mRNA的3端非编码区,降解了ATM mRNA,抑制了ATM的翻译,随后DNA-PKcs的磷酸化水平下降,DNA-PKcs与DSB的结合减少,最终抑制了鼻咽癌细胞HONE1的DNA修复.
