    • 摘要: Aimof this study was to identify potential microRNAs associated with the progression of nasopharyngeal carcinoma.GSE36682 was downloaded from Gene Expression Omnibus database(GEO),and weighted gene co-expression network analysis(WGCNA)was constructed.The microRNA co-expression module was extracted from the network by WGCNA R software package.Hub microRNAs of module were Screened.miRNet was used to construct microNA-mRNA network and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis was performed with the target genes in the network.917 microRNAs were included and five co-expression modules were identified.Blue and turquoise modules were associated with metastasis of nasopharyngeal carcinoma.KEGG results showed that blue and tturquoise modules were involved in six pathways:Pathways in cancer,p53 signaling pathway,Glioma,Chronic myeloid leukemia,Prostate cancer and Neurotrophin signaling pathway.TP53 occupies an important position in the network.

    • 摘要: Background: In low incomes country, management and follow up of prostate cancer were hard due to difficult to earn drug and difficult access of new test imaging. We explore the epidemiological and diagnostic aspects of prostate cancer at regional university hospital of Parakou from 1st January 2017 to 1st January 2019. Objective: To study diagnostic and clinical aspects of prostate cancer at regional university hospital of Parakou. Methodology: This study was descriptive with prospective data collection (1st January 2017 to 1st January 2019). Patients who were admitted to the department were included. Demographic, clinical and anatomopathological variables were studied. Result: Eighty-one patients were included;the hospital prevalence is 1.6%. The mean year old of patients was sixty-nine years old ± 10 years old (farthest 45 years old and 95 years old). More than half patients were seventy years old. The medical past history of family cancer were found in 6.1% of cases. The complaints were voiding difficult in 54.3% of cases, urinary frequency in 46.9% of cases, complete urine retention in 28.4% of cases and bones pain in 14.8% of cases. The average time of consultation was 24.3 months (farthest 2 months and 48 months). The mean of prostate specific antigen (PSA) was 93.1 ng/ml (farthest 4 ng/ml and 1000 ng/ml). Adenocarcinoma was found in 84.1% of cases. Classic X-ray imaging was used in most cases to detect metastasis. Five patients had done thoracic-abdominal-pelvis tomography. Sixty-six patients (81.5% of cases) were prostate cancer metastasis. Conclusion: Prostate cancer was frequently disease in seventy years old patients. Diagnostic was late in this study. Awareness campaign of patients for early consult was recommended.

    • 摘要: Background: Nanotechnology symbolizes a broad discipline with enormous potential in cancer treatment bridging one of the bottlenecks of traditional approaches in cancer therapy which is an inability to deliver adequate quantities of anti-cancer drugs to the tumor area. Studies on nanoparticles indicate their importance in cancer angiogenesis and metastasis. Aim: The present study assessed anti-angiogenesis and anti-metastatic effects of biogenic silver nanoparticles (AgNPs) synthesized from neem plant (Azadirachta indica). Methods: Chicken chorioallantoic membrane (CAM) and two-dimensional (2D) wound healing assays were used to study anti-angiogenic and anti-metastatic effects of the AgNPs respectively. Twenty-four fertilized eggs were divided into four groups: two biogenic AgNPs treatments at 100 μg/ml and 200 μg/ml;negative control (1% DMSO) and positive control (cyclophosphamide). On day 8 of incubation, filter discs impregnated with different concentration levels of the treatments were placed on the CAM. On day 12 of incubation, the CAMs were imaged using a stereomicroscope, scaled using ImageJ, and different morphometric and spatial parameters computed using AngioTool software. Vessel area, vessel percent area, total number of junctions, total vessel length, average vessel length, mean lacunarity, and junction density were measured. The crown-rump length (CRL) and fetal weight were also recorded on day 16 of incubation. In order to determine relative gene expression profiles of iNOS and VEGF, total RNA was extracted from the CAM, and qRT-PCR was performed with β-Actin as a reference gene. For the 2D wound healing assay, DU145 human prostate cells were grown in Dulbecco’s Modified Eagle’s Medium supplemented with 10% Fetal Bovine Serum. Results: Biogenic AgNPs demonstrated anti-angiogenic effects in a dose-dependent manner in the parameters generated from the CAM images. Also, qRT-PCR revealed down-regulation of iNOS and VEGF genes. The 2-dimensional wound healing assay showed inhibition of migration and motility of the DU145 cells for the 72-hours of assessment. Conclusion: The present study postulates that the biogenic AgNPs can prevent angiogenesis by inactivation of VEGF-NO and VEGF/VEGF-R pathways while inhibiting cell migration and metastasis.

    • 摘要: Poly Adenylate Binding Protein Interacting protein 1(PAIP1) plays a critical role in translation initiation and is associated with the several cancer types. However, its function and clinical significance have not yet been described in oral squamous cell carcinoma(OSCC) and its associated features like lymph node metastasis(LNM). Here, we used the data available from Gene Expression Omnibus(GEO), The Cancer Genome Atlas(TCGA), and Clinical Proteomic Tumor Analysis Consortium(CPTAC) to analyze PAIP1 expression in oral cancer. The publicly available data suggests that PAIP1 mRNA and protein levels were increased in OSCC. The high PAIP1 expression was more evident in samples with advanced stage, LNM, and worse pattern of invasion. Moreover, the in vitro experiments revealed that PAIP1 knockdown attenuated colony forming, the aggressiveness of OSCC cell lines, decreasing MMP9 activity and SRC phosphorylation. Importantly, we found a correlation between PAIP1 and pSRC through the analysis of the IHC scores and CPTAC data in patient samples. Our findings suggest that PAIP1 could be an independent prognostic factor in OSCC with LNM and a suitable therapeutic target to improve OSCC patient outcomes.

    • 摘要: Most hematological cancer-related relapses and deaths are caused by metastasis;thus,the importance of this process as a target of therapy should be considered.Hematological cancer is a type of cancer in which metabolism plays an essential role in progression.Therefore,we are required to block fundamental metastatic processes and develop specific preclinical and clinical strategies against those biomarkers involved in the metabolic regulation of hematological cancer cells,which do not rely on primary tumor responses.To understand progress in this field,we provide a summary of recent developments in the understanding of metabolism in hematological cancer and a general understanding of biomarkers currently used and under investigation for clinical and preclinical applications involving drug development.The signaling pathways involved in cancer cell metabolism are highlighted and shed light on how we could identify novel biomarkers involved in cancer development and treatment.This review provides new insights into biomolecular carriers that could be targeted as anticancer biomarkers.

    • 摘要: BACKGROUND Renal cell carcinoma(RCC)is an aggressive tumor,with an incidental discovery in most patients.Classic presentation is rare,and it has a high frequency of local and distant metastasis at the time of detection.CASE SUMMARY We present a rare case of a 58-year-old man with a ball-shaped thrombus in the right atrium at the time of first incidental identification of RCC in the emergency department.Cardiac metastasis,especially thrombus in the right atrium,is rare.It could either be a bland thrombus or a tumor thrombus,and physicians should consider this potentially fatal complication of RCC early at the time of initial presentation.CONCLUSION Ball-shaped lesions in the right atrium are rare,and bland thrombus should be differentiated from tumor thrombus secondary to intracardiac metastasis.

    • 摘要: Bone metastases occur in patients with advanced-stage prostate cancer(PCa). The cell-cell interaction between PCa and the bone microenvironment forms a vicious cycle that modulates the bone microenvironment, increases bone deformities, and drives tumor growth in the bone. However, the molecular mechanisms of PCa-mediated modulation of the bone microenvironment are complex and remain poorly defined. Here, we evaluated growth differentiation factor-15(GDF15) function using in vivo preclinical PCa-bone metastasis mouse models and an in vitro bone cell coculture system. Our results suggest that PCa-secreted GDF15 promotes bone metastases and induces bone microarchitectural alterations in a preclinical xenograft model. Mechanistic studies revealed that GDF15 increases osteoblast function and facilitates the growth of PCa in bone by activating osteoclastogenesis through osteoblastic production of CCL2 and RANKL and recruitment of osteomacs. Altogether, our findings demonstrate the critical role of GDF15 in the modulation of the bone microenvironment and subsequent development of PCa bone metastasis.

    • 摘要: BACKGROUND Secondary thyroid malignancies are rarely reported,especially thyroid metastasis after resection of hepatocellular carcinoma(HCC).We report a patient with thyroid metastasis after resection of HCC.CASE SUMMARY A 42-year-old female underwent partial hepatectomy for HCC three years ago.She attended hospital because of neck discomfort.After various examinations,she was diagnosed with metastatic HCC.She survived after surgical resection of the affected side of the thyroid.CONCLUSION Although secondary malignant tumor of the thyroid is classified as distant metastasis,surgical resection is still necessary according to the patient's condition.
