    • 摘要: Due to the high heterogeneity and complexity of water flow movement for multiple karst water-bearing mediums,the evaluation,effective development,and utilization of karst water resources are significantly limited.Matrix flow is usually laminar,whereas conduit flow is usually turbulent.The driving mechanisms of water exchange that occur between the karst conduit and its adjacent matrix are not well understood.This paper investigates the hydrodynamic characteristics and the mechanism of flow exchange in dual water-bearing mediums(conduit and matrix)of karst aquifers through laboratory experimentation and numerical simulation.A karst aquifer consisting of a matrix network and a conduit was proposed,and the relationship between the water exchange flux and hydraulic head differences generated from the laboratory experiments was analyzed.Two modes of experimental tests were performed with different fixed water level boundaries in the laboratory karst aquifer.The results indicate that the water exchange capacity was proportional to the square root of hydraulic head differences.The linear exchange term in the conduit flow process(CFP)source program was modified according to experimental results.The modified CFP and the original CFP model experimental data results were compared,and it was found that the modified CFP model had better fitting effects.These results showed that the water exchange mechanism between conduit and matrix is very important for solid-liquid interface reaction,water resource evaluation,and understanding of karst hydrodynamic behavior.

    • 摘要: Inflammation-associated proteinase functions are key determinants of inflammatory stromal tissues deconstruction.As a specialized inflammatory pathological process,dental internal resorption(IR)includes both soft and hard tissues deconstruction within the dentin-pulp complex,which has been one of the main reasons for inflammatory tooth loss.Mechanisms of inflammatory matrix degradation and tissue resorption in IR are largely unclear.In this study,we used a combination of Cre-loxP reporter,flow cytometry,cell transplantation,and enzyme activities assay to mechanistically investigate the role of regenerative cells,odontoblasts(ODs),in inflammatory mineral resorption and matrices degradation.We report that inflamed ODs have strong capabilities of matrix degradation and tissue resorption.Traditionally,ODs are regarded as hard-tissue regenerative cells;however,our data unexpectedly present ODs as a crucial population that participates in IR-associated tissue deconstruction.Specifically,we uncovered that nuclear factor-kappa b(NF-κB)signaling orchestrated Tumor necrosis factorα(TNF-α)-induced matrix metalloproteinases(Mmps)and Cathepsin K(Ctsk)functions in ODs to enhance matrix degradation and tissue resorption.Furthermore,TNF-αincreases Rankl/Opg ratio in ODs via NF-κB signaling by impairing Opg expression but increasing Rankl level,which utterly makes ODs cell line 17IIA11(A11)become Trap^(+) and Ctsk^(+) multinucleated cells to perform resorptive actions.Blocking of NF-κB signaling significantly rescues matrix degradation and resorptive functions of inflamed ODs via repressing vital inflammatory proteinases Mmps and Ctsk.Utterly,via utilizing NF-κB specific small molecule inhibitors we satisfactorily attenuated inflammatory ODs-associated human dental IR in vivo.Our data reveal the underlying mechanisms of inflammatory matrix degradation and resorption via proteinase activities in IR-related pathological conditions.

    • 摘要: Recent animal studies showed that isoflurane exposure may lead to the disturbance of hippocampal neurogenesis and later cognitive impairment.However,much less is known about the effect of isoflurane exposure on the neurons generated form tertiary dentate matrix of dentate gyrus,even though a great increase in granule cell population during the infantile period is principally derived from this site.To label the nascent cells from the tertiary dentate matrix of dentate gyrus,the mice were injected with bromodeoxyuridine(BrdU)on postnatal day 6(P6)and were exposed to isoflurane for 4 hat 1,8,21 day(s)and 42 day(s)after BrdU injection and the brains were collected 24 h later.The loss of different developmental stage nascent cells was assessed with BrdU,BrdU+DCX,BrdU+NeuN or BrdU+Prox-1 staining respectively.We found that the isoflurane significantly decreased the newly born(1-day old)cells and mature cells(42-day old),but not maturing cells(8-and 21-day old),suggesting isoflurane exposure exerts a neurotoxic effects on tertiary dentate matrix-originated nascent cells with an age defined pattern in mice.

    • 摘要: Decellularized extracellular matrix(dECM)is widely used in regenerative medicine as a scaffold material due to its unique biological activity and good biocompatility.Hydrogel is a three dimensional network structure polymer with high water content and high swelling that can simulate the water environment of human tissues,has good biocompatibility,and can exchange nutrients,oxygen,and waste with cells.At present,hydrogel is the ideal biological material for tissue engineering.In recent years,rapid development of the hydrogel theory and technology and progress in the use of dECM to form hydrogels have attracted considerable attention to dECM hydrogels as an innovative method for tissue engineering and regenerative medicine.

    • 摘要: Diffusion-ordered nuclear magnetic resonance spectroscopy(DOSY),dubbed chromato-graphic NMR spectroscopy,can be used to simultaneously distinguish and identify the structures of components in a mixture according to their different diffusion coefficients.In order to improve the resolution of DOSY on the diffusion dimension,a lot of matrices have been developed to expand the application of this technique in mixture analysis.However,there is no matrix to detect the mixture of ionic liquids(ILs).Herein,we introduced a new matrix,poly(sodium-p-styrenesulfonate)(PSSNa),which can be used to fully separate the signals of a mixture of different ILs.The mixture of three imidazolium ILs of 1-butyl-3-methylimidazolium bromide,1-allyl-3-vinylimidazolium bromide and 1-arboxymethy-3-methylimidazolium chloride could be fully distinguished by virtue of their different interactions with PSSNa.We also investigated the influences of PSSNa amount,IL con-centration and solution pH value on the signal resolution of mixtures.This work provides a scientific reference for the analysis of the other IL analytes.
