Parkinson's disease(PD)is the most common movement disorder,with resting tremor,rigidity,bradykinesia and postural instability being major symptoms(1).Neuropathologically,it is characterised by the presence of abundant filamentous inclusions ofα-synuclein in the form of Lewy bodies and Lewy neurites in some brain cells,including dopaminergic nerve cells of the substantia nigra(2).PD is increasingly recognised as a multisystem disorder,with cognitive decline being one of its most common non-motor symptoms.Many patients with PD develop dementia more than 10 years after diagnosis(3).PD dementia(PDD)is clinically and neuropathologically similar to dementia with Lewy bodies(DLB),which is diagnosed when cognitive impairment precedes parkinsonian motor signs or begins within one year from their onset(4).In PDD,cognitive impairment develops in the setting of well-established PD.Besides PD and DLB,multiple system atrophy(MSA)is the third major synucleinopathy(5).It is characterised by the presence of abundant filamentousα-synuclein inclusions in brain cells,especially oligodendrocytes(PappLantos bodies).We previously reported the electron cryo-microscopy(cryo-EM)structures of two types ofα-synuclein filaments extracted from the brains of individuals with MSA(6).Each filament type is made of two different protofilaments.Here we report that the cryo-EM structures ofα-synuclein filaments from the brains of individuals with PD,PDD and DLB are made of a single protofilament(Lewy fold)that is markedly different from the protofilaments of MSA.These findings establish the existence of distinct molecular conformers of assembledα-synuclein in neurodegenerative disease.

Through the last decades,our knowledge about ocular adnexal lymphoma(OAL)including orbital lymphoma has increased markedly due to the focus on lymphoma subclassification and the focus on improved staging of OAL.Our retrospective study of 797 patients with orbital lymphoma(1),discussed in the Editorial by Stagner and Zukerberg(2),provides additional insight into this relatively rare disease.Though orbital lymphoma is uncommon,lymphoma is the most common malignant neoplasm to arise within the orbit;hence it is of great importance to investigate the clinical and histopathological features of lymphomas of this specific site.As pointed out by Stagner and Zukerberg(2),a cohort with the size of nearly 800 orbital lymphoma patients has never been analyzed.The material is unique with seven contributing eye cancer centers from different parts of the world.

Objective To evaluate the effects of BQ-123 on cardiac function and ventricular remodelling after coronary microembolization (CME) in rats. Methods We created a rat model of CME by injecting a suspension of autogenic microthrombotic particles into left ventricle. Three days after the procedure, the 30 surviving rats were randomly divided into 3 groups, each consisted of 10 rats: sham-operation group(SO), CME model group(CM) and BQ-123 intervention group(BQ). Rats in the BQ group received BQ-123 (400μg/kg per day, intraperitoneally) for 4 weeks. Plasma and myocardial endothelin-1 (ET-1) were measured by radioimmunoassay. And serial echocardiography was performed to monitor alterations of left ventricular end-systolic and end-diastolic diameter (LVESD, LVEED), and left ventricular short-axis fraction shortening(LVFS) and ejection fraction (LVEF), and physiologicography to document the changes of left ventricular systolic pressure (LVSP) and end-diastolic pressure pressure(LVEDP), and left ventricular maximum positive and negative dp/dt (±LVdp/dtmax). Results Compared with sham-operated group, both LVEDD and LVESD were increased (P0.01), whereas LVFS and LVEF were significantly decreased (P0.01) in CME group; LVEDP was markedly increased, while LVSP and±LVdp/ dtmax markedly reduced in CME group (P0.01); plasma and tissue ET-1 levels increased in CME group (P0.01). BQ-123 intervention significantly decreased both the plasma and tissue ET-1 levels (P0.01), and markedly increased LVFS and LVEF, with significant improvement of LVSP and±LVdp/ dtmax (P0.01). Conclusions Treatment with BQ-123 prevents ventricular remodeling after CME due to suppression of the endothelin system.

Objective To investigate the expression of HSP90α in human hepatocellular carcinoma (HCC) and explore its relation to biological behavior and prognosis of the tumor. Methods Immunohistochemical S - P method was used to detect the expression of HSP90α in liver tissue from 10 normal individuals and liver and juxtacancerous tissues from 40 patients with HCC. Results The expression rate of HSP90α was 10% ,52.5% and 72.5% in normal liver tissue,juxtacanerous tissue and HCC tissue,respectively. A significantly higher distribution of HSP90α expression in HCC tissue compared with juxtacancerous tissue and normal liver tissue was found (P 0. 05). HSP90α was markedly correlated to tumor differentiation and size (P 0. 05) but not to tumor number (P 0. 05). Meanwhile,it was correlated with prognosis of HOC. The mean tumor-free survival time in HSP90α-negative patients was 38. 6 months while that in HSP90α-positive ones 25. 5 months. There was significant difference between these 2 groups of patients (

To investigate the relationship between left ventricularmass index(LVMI),left ventricular end-diastolic volume(EDV)andmyocardial ischemia induced with treadmill exercise test in patientswith normal coronary angiogram.Method:Sixty-one male patientswith normal coronary angiogram were divided into two groupsaccording to treadmill test:positive(23 cases)and negative group(38 cases).Left ventricular end-diastolic diameter(EDD),interventricular septum thickness(IVST),posterior wall thickness(PWT),EDV,LVMI,etc.were measured or calculated withechocardiography.Results:LVMI in positive treadmill test group(114.2±35.8 g/m^2)was markedly larger than that in negative group(90.7±16.1,p=0.0009).IVST and PWT in positive group weresignificantly thicker than those in negative group(p=0.00002,0.00003).EDD and EDV were not different between the groups.Conclusions:The left ventricular mass increased markedly inpatients with normal coronary angiogram but positive treadmillexercise test.Left ventricular hypertrophy or hypertrophy-relatedchanges,e.g.reduction of coronary reserve,may be the cause ofischemia at exercise.The end-diastolic size of left ventricle was notrelated to ischemia induced by exercise in patients with normalcoronary angiogram.
