    • 摘要: 凝集素是一类能够识别特异性糖并与之非共价结合的蛋白或糖蛋白,因其具有特定的识别受体,故在免疫系统和发育过程中发挥了重要的作用.使用DEAE-Sepharose FF弱阴离子交换层析、SP-Sepharose强阳离子交换柱层析、Sephadex G 75分子筛凝胶层析对姬松茸凝集素进行分离纯化,研究理化因素对其凝血活性的影响及其抗菌活性.结果表明:纯化的样品经SDS-PAGE电泳分析得到单一条带,姬松茸凝集素是含有2个相同亚基的同源二聚体,其分子量都为1.20×105.纯化后的姬松茸凝集素相对粗提液的回收率为28.99%,纯化倍数为19.06倍.姬松茸凝集素对热有较好的耐受性,但温度为90°C时失去凝血活性;在强酸或强碱下丧失活性,最适pH为7~9;凝集活性不受金属离子的影响;葡萄糖是其专一性结合糖.姬松茸凝集素对褐腐菌的生长起到了明显的抑制作用,而对大肠杆菌、枯草芽孢杆菌、绿色木霉、西瓜枯萎病菌和黑曲霉的生长没有明显的影响.

    • 摘要: 目的 探讨中药海藻羊栖菜(Sargassum fusiforme)中的硫酸喹诺糖基二酰基甘油酯(SQDG)对凝血作用的影响,初步探讨SQDG促凝血作用途径.方法 采用断尾法评价SQDG对小鼠出血时间的影响,利用玻片法评价SQDG对小鼠凝血时间的影响;血凝仪法检测SQDG对活化部分凝血活酶时间APTT、凝血酶原时间PT、凝血酶时间TT的影响.结果 SQDG组的出血时间、凝血时间比空白对照组分别缩短了28.96%、52.92%,SQDG可以显著缩短TT 40.42%,但对APTT和PT没有影响.结论 SQDG具有促凝血作用,可能是在凝血过程的纤维蛋白形成阶段发挥作用的.

    • 摘要: 建立面包树果实凝集素(frutalin,FTL)的提取纯化方案,并对FTL纯品进行部分理化性质分析.本试验以新鲜的面包树果实籽为材料,经Tris-HC1缓冲液浸提、过滤、硫酸铵沉淀和阳离子交换层析后超滤浓缩得FTL纯品,最后用纯品开展了糖抑制,热稳定性,pH稳定性和金属离子耐受性等试验.提取优化得到的最佳条件为:料液比1∶2(g∶mL),浸提时间2h,浸提温度50°C,硫酸铵饱和度70%,沉淀时间4h.经阳离子交换层析后FTL回收率为22%.研究结果表明,FTL是一种热稳定性凝集素,在温度不超过60°C时,其凝血活性不受影响,并且不依赖于金属离子来发挥其凝血活性.本研究建立了一个简单高效的FTL分离纯化方案,并对FTL纯品做了部分理化性质研究,为FrL可能的生物医学应用提供研究基础.

    • 摘要: 目的 分析1例凝血因子Ⅹ (coagulable factor Ⅹ,FⅩ)缺陷症家系的表型及基因型,并探讨F10基因突变与表型的关系.方法 用一期凝固法测定先证者及其家系成员的凝血酶原时间(prothrombin time,PT)、活化部分凝血酶时间(activated partial thromboplastin time,APTT)、纤维蛋白(fibrinogen,FIB)及血浆FⅡ活性(FⅡactivity,FⅡ∶C)、血浆FⅦ活性(FⅦactivity,FⅦ∶C)、血浆FⅨ活性(FⅨactivity,FⅨ∶C)、血浆FⅩ活性(FⅩ activity,FⅩ∶C)等指标进行表型诊断;用ELISA法检测FⅩ抗原含量(FⅩ antigen,FⅩ∶Ag);用PCR对先证者及其家系成员F10基因8个外显子及其侧翼序列、5'和3'非翻译区进行扩增,产物纯化后直接进行正向和反向测序;同时对100名健康对照DNA相应突变区域测序以排除基因多态性;应用软件分析突变位点序列和蛋白质构象改变特点及同源比对分析.结果 先证者PT、APTT、FⅩ∶C、FⅩ∶Ag均有异常,分别为16.1s、49.0 s、27%、56%,其他凝血表型指标均正常;先证者母亲PT、APTT、FⅩ∶C、FⅩ∶Ag分别为14.8s、37.4s、44%、34%;先证者外祖母PT、APTT、FⅩ∶C、FⅩ∶Ag分别为15.8 s、42.2 s、31%、45%;父亲的凝血指标均正常.先证者、母亲和外祖母均为F10第8外显子g.28076G>A杂合突变,导致p.Gly363Ser,父亲无此突变.p.Gly363Ser突变导致FⅩ蛋白二级结构和三维空间结构的改变,导致活性降低.结论 该先证者的家系中存在F10遗传性杂合突变g.28076G>A(p.Gly363Ser),且该突变与FⅩ水平降低有关,为国内尚未见报道的突变.%Objective To explore the correlation between F10 gene mutation and its phenotype in a Chinese pedigree affected with F Ⅹ deficiency.Methods Prothrombin time (PT),activated partial thromboplastin time(APTT),fibrinogen,F Ⅱ activity(F [Ⅱ ∶ C),F Ⅶ activity (F Ⅶ ∶ C),F Ⅸ activity (FⅨ ∶ C),FⅩ activity(FⅩ ∶ C) were determined with a one-stage clotting assay.The FⅩ antigen(FⅩ ∶ Ag) was detected with an enzyme linked immunosorbent assay(ELISA).The 8 exons,introns and 5'and 3'untranslated regions(UTR) of the F10 gene of the proband and her family members were subjected to PCR amplification and Sanger sequencing.Suspected mutation was confirmed by reverse sequencing.Polymorphisms were excluded by direct sequencing of 100 healthy individuals.Results The PT and APTT of the proband have prolonged to 16.1 s and 49.0 s,respectively.Her FⅩ ∶ C and FⅩ ∶ Ag were reduced by 27% and 56%,and her mother's PT,APTT,FⅩ ∶C and FⅩ ∶Ag were 14.8 s,37.4 s,44%,34%,respectively.Her grandmother's PT,APTT,FⅩ ∶ C and FⅩ ∶ Ag were 15.8 s,42.2 s,31%,45%,respectively.The results of her father and other family members were all within the normal range.Genetic analysis has revealed a heterozygous G>A mutation in the proband at position 28076 in exon 8 of the F10 gene,which resulted in a p.Gly363Ser substitution.The same mutation was also found in her mother and grandmother.No mutation of the F10 gene was found in her father.Gly363Ser may result in changes in the secondary structure of the F Ⅹ protein and reduction of its activity.Conclusion The g.28076G> A (p.Gly363Ser) mutation of the F10 gene probably underlies the F Ⅹ deficiency in this pedigree.The mutation was discovered for the first time in Chinese patients.

    • 摘要: The chemical constituents of Cortex Moutan charcoal were studied in depth,and their coagulation activity was screened.The chemical constituents were isolated by polyamide,silica gel and Sephadex G-Sepharose chromatography purification,and their structures were identified by NMR spectroscopy and other analyses.A total of 10 compounds were obtained and identified as follows:3-(2-furan)-l-(2-hydroxy-4-methoxy-phenyl)-2-propylene ketone (1),2-(2,5-hydroxy-4-methyl phenyl) propionate ethyl ester (2),methyl tetradec-5-enoate (3),1,4-diethylcyclohexane (4),catechol (5),methyl-4-hydroxybenzoate (6),3-hydroxy-2-methyl-4-pyrone (7),3,8-dihydroxy-2-metyl-chromone (8),3β-hydroxy-olean-12-en (9),1-monolinolein (10).Among them,compounds 1 and 2 were new natural products,and 3-10 were isolated from the Cortex Moutan charcoal for the first time.The potential coagulation activities of compounds were evaluated,and compounds 2,9 and 10 had strong hemostatic effects.While compounds 1 and 8 could significantly activate blood circulation.%对中药牡丹皮炭的化学成分进行深入研究,并对其凝血活性进行筛选.通过聚酰胺、硅胶、葡聚糖凝胶等柱色谱方法进行分离纯化,利用核磁共振波谱等方法鉴定结构.共分离鉴定了10个化合物,分别为3-(2-呋喃)-1-(2-羟基-4-甲氧基苯基)-2-丙烯酮(1)、2-(2,5-羟基-4-甲基苯基)丙酸乙酯(2)、5-十四烯酸甲酯(3)、1,4-二乙基-环己烷(4)、儿茶酚(5)、对羟基苯甲酸甲酯(6)、3-羟基-2-甲基-4-吡喃酮(7)、3,8-二羟基-2-甲基色酮(8)、3β-羟基-齐墩果烷-12-烯(9)、1-亚油酸单甘油酯(10).其中1,2为新天然产物,3-10为首次从牡丹皮炭中分离得到.在体外评估化合物潜在的凝血活性,化合物2,9和10具有强烈的止血作用,而化合物1和8可以活化血液循环.

    • 摘要: 目的:本论文旨在采用不同的提取和微生物转化方法,从宽体金线蛭中得到一种既不影响内皮细胞凝血相关因子mR NA的表达,又能够抑制动脉粥样硬化发展的水蛭提取物。方法:本实验以宽体金线蛭的干燥全体为原材料,采用不同酶解法制备得到4种水蛭提取物,分别命名为水蛭水提物(LP)、胃蛋白酶酶解水蛭提取物(PHL)、胃蛋白酶和胰蛋白酶酶解水蛭提取物(PTHL)和粗酶酶解水蛭提取物(CEHL)。

    • 摘要: Objective To observe effects of proseal laryngeal mask airway on coagulative activity of patients who underwent total laparoscopic hysterectomy under general anesthesia.Methods 92 patients who underwent total laparoscopic hysterectomy treated in department of gynaecology of our hospital from January 2013 to October 2015 were selected as the research objects,they were divided by random number table into the control group (n =45) and the observation group (n =47).They all received total intravenous anesthesia,the observation group was given proper proseal laryngeal mask airway,the control group was given ID7.0 tracheal intubation.Heart rate,blood pressure and oxygen saturation before anesthesia (T0),induced intubation or laryngeal mask placed instant moment (T1) in the two groups were recorded,prothrombin time (PT),activated partial thromboplastin time (aPTT),fibrinogen (Fbg) and other coagulative indexes of the two groups preoperative 24 h and postoperative 24 h were tested,whether there was body movement,bucking or not when tube drawing of the two groups were recorded,all patients were followed up for 24 h after the operation to explicit whether there was pharyngalgia,trachyphonia,nausea and vomitting and other complications after the anesthesia.Results There was no significant difference in heart rate and mean arterial pressure at T0 of the two groups,oxygen saturation at T0 and T1 (P > 0.05);heart rate,mean arterial pressure at T1 of the observation group were significantly lower than the control group (P < 0.05).There was no significant difference in PT,aPTT,Fbg of the two groups preoperative 24 h (P > 0.05);PT,aPTT of the observation group postoperative 24 h were significantly longer than the control group,Fbg was significantly lower than the control group(P < 0.05).There was no significant difference in incidence of bloodstain on catheter side wall,trachyphonia,nausea and vomitting and other adverse reactions in the two groups (P > 0.05);incidence of body movement or bucking,pharyngalgia of the observation group were 10.6%,6.4% respectively,which were significantly lower than the control group (37.8 %,26.7 %) (P < 0.05).Conclusion Application of proseal laryngeal mask airway in patients who underwent total laparoscopic hysterectomy under general anesthesia is beneficial to maintaining stable intraoperative heart rate,blood pressure,and can reduce changes of eoagulative activity caused by trauma with less adverse reactions,controlling its indications fully and rational use is beneficial to patients who underwent total laparoscopic hysterectomy under general anesthesia.%目的 观察第三代喉罩对全麻下腹腔镜全子宫切除术患者凝血功能的影响.方法 收集我院2013年1月到2015年10月期间收治的行妇科腹腔镜下行全子宫切除手术患者92例作为研究对象,按随机数字表分为对照组(n=45)与观察组(n=47).均采取全凭静脉麻醉,观察组使用合适型号的第三代喉罩,对照组使用ID7.0气管导管.记录两组麻醉开始前(T0)、诱导插管或喉罩放置即时(T1)时刻的心率、血压与血氧饱和度,分别于术前24 h及术后24 h检测两组凝血酶原时间(PT)、活化部分凝血活酶时间(aPTT)、纤维蛋白原(Fbg)等凝血指标,记录两组拔管时有无体动、呛咳,术后24 h进行随访明确有无咽痛、声嘶、恶心呕吐等麻醉后并发症.结果 两组T0时刻的心率、平均动脉压及T0、T1时刻的血氧饱和度均无统计学意义(P>0.05);观察组T1时刻的心率、平均动脉压均低于对照组,差异有统计学意义(P<0.05).两组术前24 h的PT、aPTT、Fbg均无统计学意义(P>0.05);观察组术后24 h的PT、aPTT长于对照组,Fbg低于对照组,差异均有统计学意义(P<0.05).两组导管侧壁有血迹、声嘶、恶心呕吐等不良反应发生率的差异无统计学意义(P>0.05);观察组体动或呛咳、咽痛发生率分别为10.6%、6.4%,低于对照组的37.8%、26.7%,差异均统计学意义(P<0.05).结论 第三代喉罩在全麻下腹腔镜全子宫切除术患者中的应用利于维持术中心率、血压的稳定,同时减轻由于创伤所致的凝血活性变化,不良反应少.

    • 摘要: 针对菜豆凝集素二聚体复合物的结构特征,利用计算机辅助药物设计的方法设计抑制剂以破坏复合物结构;进一步采用标准的Fomc保护氨基酸NT氨基的固相法合成纯化了小肽抑制剂.体外兔血红细胞凝集实验结果表明,该小肽对菜豆凝集素的凝血能力有一定的抑制效果.该方法为植物凝集素凝血研究及菜豆相关的食品安全问题提供了一个新思路.%Common bean(Phaseolus vulgaris L.),rich in protein and low in fat,is one of the most important legume crops in the world.However,consumption of improperly prepared common bean can lead to food poisoning.Previous studies have largely attributed this toxic effect to the high content in lectin,a plant protein that binds specifically to carbohydrate or carbohydrate structures displayed on cell surface.Since most lectin forms a dimer complex for biological functions,a short peptide was designed to break the dimer interface.Detailed dynamical network and structural characteristic analysis were performed to select this peptide.In vitro hemagglutination assay showed that this peptide,upon binding to lectin,disrupts the dimer formation partially and weakens the hemagglutination effect.Taken together,a novel peptide inhibitor was designed whose potency and specificity can be further optimized for anti-hemagglutination and food safety applications.
