To explore the preparation method of the absolute of peony flower by subcritical extraction at low temperature and dewax with ethanol, the preliminary experiments were conducted to find out the influence of extraction solvent on yield of concrete and to determine the parameters for dewax process. L9 (34) Orthogonal test was used to optimize the extraction conditions, and the components in the obtained absolute were analyzed by GC - MS. The results showed that the butane was a suitable extraction solvent in preliminary experiment. The best technological conditions were as follows: extraction time 30 min for 4 times, extraction temperature 40 °C, and extraction pressure 0.35 MPa. The reasonable process conditions for dewax were as follows: concrete(g) / anhydrous ethanol(mL) = 1 / 10, dissolving at 40 °C with medium stirring speed for 45 min, then storing in the refrigerator (-10 °C) for 12 h, filtering, and recovering ethanol under vacuum distillation, finally the oil yield was 0.06%. Fifty - five kinds of components were identified in the absolute of peony flower by GC - MS analysis. The low temperature and pressure process effectively maintained high natural quality of product, and the industrial production would be realized as these relative low requirements for equipments and investments.%为探索亚临界萃取牡丹花浸膏结合乙醇除去浸膏中蜡质制备牡丹花净油的工艺,采用预试验考察萃取溶剂对浸膏收率的影响,并确定乙醇除蜡质的工艺参数;以L9(34)正交试验优选萃取浸膏的条件;并用GC-MS分析牡丹花净油的组分.预试验发现丁烷为较合适的萃取溶剂,萃取浸膏的最佳工艺条件为:单次萃取时间30 min,萃取次数4次,萃取温度40°C,萃取压力0.35 MPa.预试验得到乙醇除蜡质的合理工艺参数为:浸膏(g):无水乙醇(mL)=1:10,中速搅拌,40°C溶解45 min,在-10°C下静置12 h,过滤,减压回收乙醇得牡丹花净油,收率为0.06%.利用GC-MS方法分析牡丹花净油,从中检出55种成分.本研究确定的工艺条件,全过程温度低、压力低,有效保持了产品的天然品质,且对设备材质要求不高,投资小,易实现工业化生产.
