摘要:
猪流行性腹泻病毒(PEDV)是猪肠道冠状病毒,可引起猪急性肠胃炎、水样腹泻、呕吐症状,是造成仔猪腹泻的主要病毒,对全球养猪业造成重大损失.先天免疫作为机体防御病毒感染的第一道防线具有重要作用,它与多种蛋白和机制有关,包括Interferons(IFNs)、炎性细胞因子、凋亡、自噬等.其中干扰素作为宿主免疫防御病毒感染最主要的细胞因子,其大量表达帮助宿主拮抗病毒的感染,最终将病毒从宿主体内清除.然而PEDV感染可抑制干扰素的表达,PEDV通过抑制TBK1、降解STAT1、阻碍NF-κB、IRF3及IRF1核移位、干扰RIG-I/STING泛素化等信号通路等方式,干扰IFN表达或其功能发挥,从而协助病毒免疫逃逸.本文主要阐述PEDV免疫逃逸中IFN应答机制研究,旨在为PEDV的疫苗和新型抗病毒药物研究提供思路.

摘要:
Under non-random uncertainty, a new idea of finding a possibly optimal solution for linear programming problem is examined in this paper. It is an application of the intuitionistic fuzzy set concept within scope of the existing fuzzy optimization. Here, we solve a linear programming problem (LPP) in an intuitionistic fuzzy environment and compare the result with the solution obtained from other existing techniques. In the process, the result of associated fuzzy LPP is also considered for a better understanding.

但都高于漏出液组(P<0.05);胸水IFN-γ在结核性胸水组中高于漏出液组和恶性组(P<0.05).联合检测血清、胸水CYFRA21-1诊断结核性胸膜炎的敏感度为93.5%,特异度为74.1%;单独检测胸水IFN-γ诊断结核性胸膜炎的敏感度为87.1%,特异度为79.3%;联合检测血清、胸水CRFYA21-1以及胸水IFN-γ 诊断结核性胸膜炎的敏感度为90.3%,特异度为89.6%.结论联合检测血清、胸水CYFRA21-1以及胸水IFN-γ对结核性胸膜炎的早期诊断有较高的临床参考价值.%Objective To explore the significance of combined detection of cytokeratin 19 soluble fragment (CYFRA211)and pleural gamma interferon (IFN - gamma)in pleural effusion for early diagnosis of tuberculotic pleurisy. Methods A total of 89 patients with different causes of pleural effusion were selected for this study,and serum and pleural effusion levels of CYFRA21 - 1 and hydrothorax interferon - gamma were analyzed,at the same time the best threshold figures of sensitivity and specificity had been detected by using ROC curve analysis. Results The serum concentration of CYFRA21 - 1 in malignant group was higher than that of tuberculous and transudative effusion groups (P < 0. 05). There was no significant difference between the concentration of CYFRA21 - 1 in malignant pleural effusion and tuberculous pleural effusion groups (P = 0. 13),but it was higher than that of transudate group (P < 0. 05). Level of pleural IFN - gamma in tuberculotic pleural effusion group was higher than that of transudate group and malignant group (P < 0. 05). The sensitivity of combined detection of serum and pleural fluid levels of CYFRA21 - 1 for diagnosis of tuberculous pleurisy was 93. 5%,and its specificity was 74. 1% . The sensitivity of separate detection of pleural IFN - gamma for diagnosis of tuberculous pleurisy was 87. 1%,and its specificity was 79. 3% . The combined detection of serum and pleural ef-fusion levels of CRFYA21 - 1 and pleural IFN - gamma CRFYA21 - 1 for diagnosis of tuberculous pleurisy was 90. 3%,and its specificity was 89. 6% . Conclusion The sensitivity of combined detection of CYFRA21 - 1 in serum and pleural effusion and IFN - gamma in pleural effusion is good for the early diagnosis of tuberculous pleurisy with higher value for clinical reference.

AIM To investigate the mechanism by which hepatitis C virus(HCV) core protein-induced mi R-93-5 p up-regulation regulates the interferon(IFN) signaling pathway.METHODS HCV-1 b core protein was exogenously expressed in Huh7 cells using pc DNA3.1(+) vector. The expression of mi R-93-5 p and interferon receptor 1(IFNAR1) was measured using quantitative reverse transcriptionpolymerase chain reaction and Western blot. The protein expression and phosphorylation level of STAT1 were evaluated by Western blot. The overexpression and silencing of mi R-93-5 p and IFNAR1 were performed using mi R-93-5 p agomir and antagomir, and pc DNA3.1-IFNAR1 and IFNAR1 si RNA, respectively. Luciferase assay was used to identify whether IFNAR1 is a target of mi R-93-5 p. Cellular experiments were also conducted.RESULTS Serum mi R-93-5 p level was increased in patients with HCV-1 b infection and decreased to normal level after HCV-1 b clearance, but persistently increased in those with pegylated interferon-α resistance, compared with healthy subjects. Serum mi R-93-5 p expression had an AUC value of 0.8359 in distinguishing patients with pegylated interferon-α resistance from those with pegylated interferon-α sensitivity. HCV-1 b core protein increased mi R-93-5 p expression and induced inactivation of the IFN signaling pathway in Huh7 cells. Furthermore, IFNAR1 was identified as a direct target of mi R-93-5 p, and IFNAR1 restore could rescue mi R-93-5 p-reduced STAT1 phosphorylation, suggesting that the mi R-93-5 p-IFNAR1 axis regulates the IFN signaling pathway.CONCLUSION HCV-1 b core protein-induced mi R-93-5 p up-regulation inhibits the IFN signaling pathway by directly targeting IFNAR1, and the mi R-93-5 p-IFNAR1 axis regulates STAT1 phosphorylation. This axis may be a potential therapeutic target for HCV-1 b infection.

摘要:
目的 探讨索菲布韦联合普通干扰素(Interferon IFN)和利巴韦林在1b型丙肝患者治疗中的临床效果,为1b型丙肝患者选取最优的治疗方案.方法 收集自2015年4月~10月在抚州市第一人民医院感染科进行1b型丙肝治疗的患者90名作为本次研究的观察对象,根据给药方式分为3组:对照A组30例:进行索菲布韦+利巴韦林治疗,治疗时间为24周;对照B组30例:普通IFN+利巴韦林治疗,治疗时间48周;治疗组30例:索菲布韦+普通IFN+利巴韦林治疗,治疗时间为24周.分别在治疗0周、4周、12周、治疗结束时、治疗结束后12周、24周行丙肝高灵敏度RNA测定.并比较3组患者在治疗中的疗效差异、观察不良反应.结果 接受索菲布韦治疗的治疗组与对照组A治疗效率分别为93.33%、90%明显高于对照组B的66.67%(P<0.05),在治疗过程中对照组A不良反应最少.结论 索菲布韦联合普通IFN和利巴韦林治疗1b型丙肝患者的临床效果优于传统治疗方案干扰素+利巴韦林,且不良反应少,在治疗丙型肝炎中具有一定的临床价值.

摘要:
Ojective] Researching the relationship of interferon, HOTAIR(HOX gene transcript antisense RNA) and malignant glioma to explore molecular mechanism of tumorigenesis and development. [Methods] Analyzing the differences of expression of HOTAIR in tumor tissue and normal tissue by GSE4290 datasets which is downloaded from gene expression database from NCBI website and analyzing the differences of expression of HOTAIR in glioma cells and normal glial cells by RT-PCR results. Testing the U87 glioma cell that treated by siHOTAIR transfection proliferation and cycle by MTT method and flow cytometry. Detecting the HOTAIR gene expression of U87 glioma cells stimulated by interferon alpha, beta or gamma via RT-PCR. [Results] The expression of HOTAIR in U87 cell is significantly higher than in normal tissue, inhibiting HOTAIR expression could reduce the proliferation rate of glioma cell and increase the cell cycle arrest. There was no significant differencein HOTAIR expression of U87 cells stimulated by interferon beta or gamma( P>0.05); but there was significant differenceinthe expression of HOTAIR in U87 cells stimulated by interferon alpha increased and this effect was further enhanced at the longer time and higher concentration(P0.05);IFN-α干预U87细胞后HOTAIR的表达量增加,干预时间越长、浓度越高表达量增加越显著,差异有统计学意义(P<0.05,P<0.01)。[结论]HOTAIR在U87细胞中异常表达可能对胶质瘤发生发展具有促进作用,IFN-α能够上调HOTAIR的表达,参与胶质瘤的发生发展。
