摘要:
本研究根据鸭甲肝病毒1型(DHAV-1)和3型(DHAV-3)的5′UTR基因的保守区域,设计了1对特异性的引物,扩增目的片段大小约为199 bp.通过优化反应条件,建立了基于高分辨率熔解曲线(HRM)分析的鸭甲肝病毒1型和鸭甲肝病毒3型的鉴别检测方法.特异性试验结果表明,该方法能特异性地检测出DHAV-1和DHAV-3,检测其它常见鸭源病毒不能形成特异性的熔解峰;敏感性和重复性试验表明,对p-DHAV-1和p-DHAV-3阳性质粒的最低检测限分别为220 copies和28 copies,对不同批次的样品扩增效果良好,具有良好的批内和批间重复性.采用该方法检测临床样品,与传统的二重PCR方法比较,符合率为100%.该方法为DHAV-1和DHAV-3的临床疑似病例的鉴别检测、快速诊断以及分子流行病学调查等提供了技术支撑.

该研究通过高分辨率溶解曲线(HRM)分析法对36例乳腺癌组织TOX3基因的7号外显子、PPP2R1A基因的6号外显子、PTEN基因的5号外显子进行突变位点检测,利用DNA测序分析法进一步验证。结果显示TOX3基因的7号外显子与PPP2R1A基因的6号外显子中无突变,但在PTEN基因5号外显子第1408位核苷酸序列由A突变为T,导致其编码氨基酸由N突变为Y,所检测的36例乳腺癌组织中有11例检测到突变,HRM结果与DNA测序结果分析一致。

摘要:
目的 建立一种通过高分辨熔解曲线(H RM)-非标记探针基因分型分析方法,检测恶性疟原虫青蒿素耐药相关K13基因C580Y突变位点.方法 通过构建野生型和C580Y突变型质粒,设计相应引物、探针,应用HRM-非标记探针技术检测C580Y突变.结果 成功检测出标本中含有C580Y突变的基因型及野生基因型.检测野生型和突变型的探针峰的温度之差为5°C.结论 H RM-非标记探针技术是一种能够快速、准确、廉价、反应通量大的检测方法,适用于疫区恶性疟原虫青蒿素耐药相关K13基因突变的大批量检测.

Brassica rapa is one of the most important leafy vegetable crops with large cultivated area in China.To increase the availability of DNA markers in B.rapa,we developed insertion-deletion(InDel)markers utilizing high-resolution melting(HRM)curve analysis.We designed primers for 252 InDels(≥3 bp)evenly distributed in the genome and tested gene polymorphisms with eight accessions.In total,208 markers were specifically amplified,and 148 InDels with polymorphism were genotyped successfully using HRM.We further analyzed the correlation with InDel size,GC number,and predicted the difference in Tm values(Tm)using 208 markers with specific amplification.We found that the success rate of InDel markers was correlated with the GC number of InDel and the predicted-Tm,but not clearly correlated with the length of InDel.When the GC number within InDel was≥8,the successful rate exceeded 90.0%.When the predicted-Tm reached 0.5°C,the success rate was greater than 90.0%,and when it was≥0.6°C,the rate climbed to 100.0%,indicating their role as the optimal parameter for successful development of an applicable InDel marker.The polymorphic InDel markers can be easily genotyped using HRM.They are of great value in genetic analysis,construction of linkage map,and molecular marker-assisted selection in B.rapa.

摘要:
The development of next generation sequencing(NGS)and high throughput genotyping are important techniques for the QTL mapping and genetic analysis of different crops.High-resolution melting(HRM)is an emerging technology used for detecting single-nucleotide polymorphisms(SNPs)in various species.However,its use is still limited in maize.The HRM analysis was integrated with SNPs to identify three types of populations(NIL population,RIL population and natural population),and the useful tags were screened.The patterns of temperatureshifted melting curves were investigated from the HRM analysis,and compared these with the kit.Among all 48 pairs of primers,10 pairs of them were selected:six pairs of primers for the NIL population,three pairs of primers for the RIL population,and one pair of primer for the natural population.The marker for the natural population was developed with a matching rate of 80%for the plant height trait,based on the data of the phenotypic characteristics measured in the field.This study provides an effective method for maize genotyping in the classification of maize germplasm resources,which can be applied to other plants for high-throughput SNP genotyping or further mapping.

摘要:
目的 探讨EGFR基因突变对非小细胞肺癌(NSCLC)患者临床特征,预后的价值,揭示总生存期(OS)≥3年的生存特点.方法 回顾性分析2013年1月1日至2018年6月30日南京医科大学附属肿瘤医院接受个体化治疗的349例NSCLC患者临床资料.对应收集石蜡包埋组织标本,采用HRM法,检测组织中EGFR基因第18、19、20、21外显子的突变状态,分析EGFR基因突变与患者临床病理参数的相关性.Kaplan-Meier方法和χ2分析随访资料完整的249例患者EGFR基因突变对生存的影响,揭示OS≥3年生存影响因素.结果 在349例NSCLC肺癌患者中,EGFR基因突变率为50.4%,EGFR基因主要高突变人群是女性(62.2%)和腺癌患者(52.0%),0.05).截至2018年12月15日,249例随访完整的患者中,中位生存时间为23个月.EGFR基因的总突变和21突变,与野生型相比较,能显著提高无进展生存期(PFS)和OS,差异具有统计学意义(P0.05).OS≥3年和OS≤1年的患者在性别、TNM分期、淋巴结转移、EGFR总突变率、21的突变率及靶向治疗率方面差异有统计学意义(P<0.05).结论 EGFR基因的总突变和21突变的患者均能明显延长生存期.
