    • 摘要: 目的 研究青翘的质量等级标准。方法 采集全国连翘主产区的138批青翘药材,按2020年版《中国药典》相关方法检测青翘药材中杂质、水分、醇溶性浸出物、挥发油、连翘苷、连翘酯苷A等质量指标的含量,并筛选合格样品。采用层次分析与主成分分析(AHP-PCA)混合加权法确定上述指标(杂质限量除外)的综合权重,并计算青翘合格样品的综合得分,再根据K-均值聚类分析结果提出青翘质量等级划分建议。结果与结论 青翘中杂质、水分、醇溶性浸出物、挥发油、连翘苷、连翘酯苷A的含量范围分别0~7.80%、1.60%~8.18%、13.13%~61.60%、0.21%~3.47%、0.02%~2.15%、0.79%~14.04%,平均含量分别为1.24%、4.97%、34.88%、2.01%、0.42%、6.86%,138批青翘药材共有47批合格。建议青翘质量等级标准可划分为3级:一级青翘的挥发油含量≥2.40%,连翘苷含量≥0.59%,连翘酯苷A含量≥8.34%,醇溶性浸出物含量≥38.66%,水分含量≤4.99%;二级青翘上述指标含量分别≥2.26%、≥0.41%、≥7.47%、≥32.58%、≤5.33%;三级青翘上述指标含量分别≥2.15%、≥0.32%、≥4.60%、≥31.52%、≤7.23%。

    • 摘要: 目的:建立青翘和老翘药材的指纹图谱并比较,同时进行聚类分析和主成分分析.方法:采用高效液相色谱法.色谱柱为Hypersil C18,流动相为乙腈-0.1%甲酸水溶液(梯度洗脱),检测波长为235 nm,柱温为25°C,流速为1.0 mL/min,进样量为10μL.以连翘苷为参照,绘制8批青翘(Q1~Q8)和6批老翘(L1~L6)药材的HPLC指纹图谱;采用《中药色谱指纹图谱相似度评价系统(2012版)》进行相似度评价,确定共有峰;采用SPSS 23.0软件进行聚类分析和主成分分析.结果:青翘和老翘药材共有19个共有峰,指认了其中6个,分别为连翘酯苷A、芦丁、松脂素-β-D-葡萄糖苷、连翘苷、槲皮素、连翘脂素;青翘与老翘药材的相似度为0.351~0.767.聚类分析结果显示,青翘和老翘药材样品可聚为4类,其中L1~L6聚为一类、Q1聚为一类、Q2~Q6聚为一类、Q7~Q8聚为一类.主成分分析结果显示,前3个主成分的累积方差贡献率为83.14%,L1~L6分布较近,Q2~Q6分布较近,Q7~Q8分布较近,Q1分布较为独立,与聚类分析结果一致.结论:青翘和老翘的指纹图谱共有峰经相似度评价、聚类分析及主成分分析均有明显差异,所建HPLC指纹图谱可用于综合评价和比较青翘和老翘药材的质量.

    • 摘要: To study the active ingredients of green fructus forsythiae in heat-clearing and detoxifying.The 60%ethanol extract of green fructus forsythiae was separated into 6 chemical composition groups F1~F6 by using C18 as the separation material.According to the ability of eliminate DPPH·, antibacterial effect, and ameliorate the degree for inflammatory of mouse macrophage RAW264.7 induced by LPS, the antioxidant, antibacterial, and anti-inflammatory activities of different chemical composition groups were evaluated.The chemical composition of F2 has strong activities in antioxidant, antibacterial and anti-inflammatory.Further chemical composition analysis showed that the active ingredients F2 of green fructus forsythia in heat-clearing and detoxifying was mainly composed of phenylethanoid glycosides, and the main active ingredients were forsythiaside A and forsythiaside I.The active ingredients of green fructus forsythia in heat-clearing and detoxifying are mainly phenylethanoid glycosides.%探讨青翘清热解毒活性组分.以C18为分离材料将青翘60%乙醇提取物分离为6个化学成分群F1~F6;以清除DPPH·的能力、抑菌效果和改善LPS诱导的小鼠巨噬细胞RAW264.7产生炎症反应的程度为评价指标, 评价不同化学成分群的抗氧化、抑菌和抗炎活性.F2化学成分群在抗氧化、抑菌和抗炎活性等方面均具有较强的活性;进一步化学成分分析表明青翘清热解毒活性组分F2主要是由苯乙醇苷类成分组成, 主要成分为连翘酯苷A和连翘酯苷I.青翘清热解毒活性活性成分主要为苯乙醇苷类成分.

    • 摘要: Objective To compare the efficacy difference of Green Forsythiae Fructus and Ripe Forsythiae Fructusby using Lianqiao powder and Yinqiao powder in the classic formula; To provide experimental evidence for the guidance of "one for dual-use" of the Forsythiae Fructus. Methods The guinea pig sore model was made with Staphylococcus aureus. 40 guinea pigs were randomly divided into blank group, model group, Lianqiao powder Green Forsythiae Fructus group and Lianqiao powder Ripe Forsythiae Fructus group. The blank group and the model group were fed with normal saline, while Lianqiao powder Green Forsythiae Fructus group and Lianqiao powder Ripe Forsythiae Fructus group were treated with 1.2 g raw medicine/kg liquid Lianqiao powder Green Forsythiae Fructus and Lianqiao powder Ripe Forsythiae Fructus for 7 d. The symptom score, blood and pathological changes of guinea pig soreness were observed. The model of acute lung injury was induced by 10% LPS aerosol inhalation. 40 rats were randomly divided into blank group, model group, Yinqiao powder Green Forsythiae Fructus group and Yinqiao powder Ripe Forsythiae Fructus group. The blank group and the model group were fed with normal saline, while Yinqiao powder Green Forsythiae Fructus group and Yinqiao powder Ripe Forsythiae Fructus group were treated with 4 g raw medicine/kg liquid Yinqiao powder Green Forsythiae Fructus and Yinqiao powder Ripe Forsythiae Fructus for 10 d. The levels of IL-6, TNF-α and IL-1β in the lung lavage fluid were detected. Results The effect of Lianqiao powder Green Forsythiae Fructus on the wound healing of guinea pig sore wound was faster than that of Lianqiao powder Ripe Forsythiae Fructus, but there was no significant difference between Lianqiao powder Green Forsythiae Fructus group and Lianqiao powder Ripe Forsythiae Fructus group in inhibiting the secretion and pathological changes of guinea pig sore wound. The levels of IL-6, TNF-α and IL-1β in Yinqiao powder Green Forsythiae Fructus group was lower than that in Yinqiao powder Green Forsythiae Fructus group, without statistical significance. Conclusion It is verified that there is efficacy differences between Green Forsythiae Fructus and Ripe Forsythiae Fructus in the different Chinese herbal compound.%目的 采用经典方剂连翘散和银翘散验证青翘和老翘的功效差异,为指导连翘"一体两用"提供实验依据.方法 采用金黄色葡萄球菌制作豚鼠疮疡模型,将40只豚鼠随机分为空白组、模型组、连翘散青翘组和连翘散老翘组,空白组和模型组灌服生理盐水,连翘散青翘组和连翘散老翘组灌服1.2 g原药材/kg药液,连续7 d,观察各组模型豚鼠疮疡症状积分、血常规及病理变化.采用10%脂多糖雾化吸入法诱导大鼠急性肺损伤模型,将40只SD大鼠随机分为空白组、模型组、银翘散青翘组和银翘散老翘组,空白组和模型组灌服生理盐水,银翘散青翘组和银翘散老翘组灌服4 g原药材/kg药液,连续10 d,观察各组大鼠肺洗液白细胞介素(IL)-6、肿瘤坏死因子(TNF)-α、IL-1β 水平.结果 连翘散青翘组抑制豚鼠疮疡伤口红肿作用较老翘组起效快,但在抑制豚鼠疮疡伤口分泌物和病理改善方面与连翘散老翘组差异无统计学意义.银翘散青翘组大鼠较老翘组IL-6、TNF-α、IL-1β 含量降低,但差异无统计学意义.结论 不同复方中青翘与老翘功效有一定差异.
