    • 摘要: SPECT/CT融合技术、图像重建算法、光子补偿技术等的不断完善和应用使得单光子发射计算机断层扫描(single photon-emission computed tomography,SPECT)已经能够开展类似于正电子发射型计算机断层扫描(positron-e-mission tomography,PET)的定量分析.SPECT/CT定量分析中使用最多的是锝-99m(technetinum-99m,99m Tc)标记的放射性药物,该文综述了99m Tc定量SPECT/CT在骨关节、甲状腺等疾病中的应用及局限,并对定量SPECT/CT的应用前景进行了展望.

    • 摘要: 为了发展99 Tcm标记的葡萄糖代谢显像剂,合成MAMA偶联的二苯并环辛炔配体和四种2-位含有叠氮基的小分子葡萄糖衍生物,依据体外点击速率实验筛选出两种可能的葡萄糖衍生物(N3-DG-1和N3-DG-2).首先完成N3-DG-1和N3-DG-2在荷S180肿瘤小鼠体内的预着靶,再注射99 Tcm标记的环辛炔探针进行寻靶,发生体内点击反应.动物分布实验结果表明,N3-DG-2的瘤血比和瘤肉比优于N3-DG-1,说明99 Tcm标记的环辛炔探针和N3-DG-2组合更适用于葡萄糖代谢显像.

    • 摘要: 目的:使用放射性核素99mTc标记程序性死亡受体-配体1(programmed cell death-ligand 1,PD-L1)纳米抗体,制备靶向非小细胞肺癌(non-small cell lung cancer,NSCLC)的显像剂(99mTc-PD-L1纳米抗体),评价其在SPECT/CT上显示肿瘤PD-L1表达的可行性.方法:采用三羰基化合物试剂盒制备标记前体三羰基锝化合物盐溶液,标记PD-L1纳米抗体,并测定标志物的标记率、放射化学纯度及体外稳定性;分别构建PD-L1高表达(HCC827)和PD-L1阴性(A549)的 NSCLC裸鼠移植瘤模型,经尾静脉注射99mTc-PD-L1纳米抗体30、90 min后,观察和分析SPECT/CT显像.结果:99mTc-PD-L1纳米抗体的标记率在95%以上,且在磷酸盐缓冲液(phosphate buffered saline,PBS)和血清中均表现良好的稳定性.小动物SPECT/CT显像表明99mTc-PD-L1纳米抗体在PD-L1高表达组中具有良好的肿瘤摄取,而在PD-L1阴性组无明显肿瘤摄取,表明99mTc-PD-L1纳米抗体能够显示肿瘤PD-L1的表达情况,具有良好的体内特异性;此外,99mTc-PD-L1纳米抗体主要分布在肾脏,通过泌尿系统排泄,肝脏、脾脏、心脏、肺及软组织等显影微弱,甲状腺区及胃肠未见核素浓聚,证实显像剂具有良好的生物分布.结论:该研究利用三羰基化合物试剂盒成功制备了99mTc-PD-L1纳米抗体,标志物具有良好的标记率和体外稳定性,能够在PD-L1高表达的NSCLC中特异性浓聚,有潜力成为一种新型的显示肿瘤PD-L1表达的显像剂.

    • 摘要: Objective:99mTc labeled BmK CT was prepared and its potential as a tumor-specific agent for SPECT imaging of glioma was performed via a xenografted nude mouse model.Methods:BmK CT was linked with DTPA to form BmK CT-DTPA,followed by radiolabeling with 99mTc.After purification from PD-10 column,the stability of 99mTc-BmK CT in vitro was studied.SPECT imaging at different time points was performed both in normal rabbits and tumor bearing nude mice after intravenous injection of 99m-Tc-BmK CT.At 4 h post-injection,one tumor bearing mouse was sacrificed to remove the tumor and major organs.Their relative radioactivity ratios were recorded by analyzing the regions of interest.Results:The radiochemical yield of 99mTc-BmK CT was above 80% and its mean radiochemical purity was more than 99%.The radiochemical purity of purified 99mTc-BmK CT was greater than 90% within 24 h both in PBS and FBS.SPECT images of 99mTc-BmK CT in normal rabbits showed that the major radioactivity was accumulated in the liver,kidney and spleen,while other tissues including thyroid had very low level.99mTc-BmK CT could be slowly cleared from the body through urinary system.The SPECT imaging in tumor bearing nude mice suggested that mildly increased SPECT signal intensity was found in the tumor site at 0.5 h post-injection and much higher was displayed at 1 h and 2 h,followed by the highest at 4 h.After that,the tumor SPECT signal descended steadily and could still be detected at 8 h post-injection.Conclusion:99mTc-BmK CT was successfully prepared with a high radiochemical purity and stability and could be used as a tumor-specific ligand for SPECT imaging of xenografted glioma model in vivo.%目的:通过放射性核素99mTc标记BmK CT多肽制备靶向胶质瘤的显像剂,探讨99mTc-BmK CT用于胶质瘤显像的可行性.方法:采用BmK CT多肽游离的氨基与DTPA酸酐反应得到BmK CT-DTPA,经99mTc标记后通过柱层析分离纯化制备99mTc-BmKCT.测定标记物在PBS溶液和血清中不同时间点放射性化学纯度,评价BmK CT-99mTc体外稳定性.新西兰白兔耳缘静脉注射99mTc-BmK CT进行SPECT显像,观察不同时间点体内的放射性分布.皮下胶质瘤裸鼠经尾静脉注射99mTc-BmK CT,观察不同时间点肿瘤的摄取情况;注射后4h处死裸鼠,分离肿瘤和主要器官进行离体SPECT显像,并用勾画感兴趣区法分析相对放射性计数.结果:99Tc标记BmK CT多肽标记率大于80%,经柱层析分离纯化后放射性化学纯度大于99%.标记物在PBS和血清稳定性良好,6h内放射性化学纯度均大于95%,12h内放射性化学纯度大于90%.正常白兔SPECT显像表明99mTc-BmK CT主要浓聚在肝脏、脾脏和肾脏,软组织持续显影微弱,甲状腺区及胃肠未见核素浓聚;显像剂主要通过泌尿系统排泄,24h肾脏与肝脏显影接近.胶质瘤裸鼠SPECT显像表明,注射后4h肿瘤显像清楚,ROI分析结果显示肿瘤/肌肉比4.26±0.25,标记物在肿瘤内代谢缓慢,8h肿瘤部位仍有较高摄取.结论:本研究成功制备了99mTc标记BmK CT多肽,标记物主要被肝、脾和肾摄取,经泌尿系统排泄;99mTc-BmK CT能够在皮下胶质瘤中浓聚,注射后4h肿瘤显影清晰,瘤内代谢缓慢,有潜力成为一种新型胶质瘤分子探针.

    • 摘要: 分别采用点击化学方法对DNA三角双锥结构进行氟-18标记研究,采用二氯亚锡还原法对DNA三角双锥结构进行锝-99m标记研究.结果表明,点击化学方法不适用于DNA三角双锥结构的氟-18标记;而采用二氯亚锡还原法则制得了以DNA三角双锥结构为载体的分子影像探针99m Tc-DBNs.%DNA bipyramid nanostructures were radiolabeled with fluorine-18 through click chemistry. Reactant concentration, catalyst dosage, reaction time, temperatureand reaction frequency in the CuAAC reaction were studied. DNA bipyramid nanostructures were also radiolabeled with technetium-99m through stannous chloride lableling method. The results showed that it was not a good idea to radiolabel DNA bipyra-mid nanostructures with fluorine-18 through click chemistry. At the same time 99m Tc-DBNs were successfully prepared . Radioactive molecular imaging probes would be obtained based on the nanoplatforms of DNA bipyra-mid nanostructures.

    • 摘要: 分析比较了亚急性甲状腺炎(亚甲炎)、原发性甲状腺功能减退及正常体检者的甲状腺及唾液腺核素显像结果.亚甲炎患者唾液腺的靶/非靶比值明显降低.亚甲炎患者的甲状腺和唾液腺功能会同时受损,这可能是其出现咽部症状的原因之一.%The function of salivary glands in patients with subacute thyroiditis was evaluated.The data of patients with subacute thyroiditis,primary hypothyroidism,and normal controls undergoing radionuclide scanning were reviewed and compared retrospectively.The ratio of target to non-target values in the salivary glands were decreased in patients with subacute thyroiditis.Thyroid and salivary glands in patients with subacute thyroiditis might be damaged simultaneously and both developed pharyngeal symptoms partly due to the damaged salivary glands.

    • 摘要: 目的 探讨甲状腺核素显像在诊断亚急性甲状腺炎中的应用.方法 实验组80例,经病理学确诊为亚急性肉芽肿性甲状腺炎和亚急性淋巴细胞性甲状腺炎;对照组40例,无甲状腺疾病.两组均用血清放射免疫检查和核素(99mTc)甲状腺静态显像的方法对比分析.结果 核素显像实验组T/N=(2.18±0.56),对照组T/N=(6.34±0.88),差异有显著性(P﹤0.05).血清放射免疫检查两组无显著性差异(P﹥0.05).结论 亚急性甲状腺炎病理过程为变化过程,不同阶段的血清放射免疫检查结果不同,但是亚急性甲状腺炎发病过程中,核素显像有其特异性,即甲状腺显影不清,边界模糊,放射性分布不均匀稀疏.核素显像对亚甲炎的诊断有不可替代的作用.

    • 摘要: 通过对比几种标记方法,选择预亚锡化法对一种抗植物蛋白抗体进行99mTc标记,标记条件相对简单,初始标记率约为65%,经SephadexG25凝胶柱纯化后该标记抗体的放射化学纯度达到80%以上.进一步研究了标记配合物在正常小鼠以及荷瘤(H22)小鼠体内的生物分布.生物性能研究表明:99mTc标记的抗体在小鼠体内主要经肾代谢,其在肿瘤内有一定的初始摄取,且清除较慢;靶与非靶比值随时间的增加而提高.但遗憾的是该标记配合物仍存在血液本底较高的缺点,希望通过进一步对该抗体片断的标记研究为肿瘤的早期诊断提供一种新的显像剂.
    • 摘要: 高血压病人或做运动试验时血压反应过高的人,常有运动心电图(ECG)“假阳性”,但运动后心肌灌注情况(MPI)有什么变化报道甚少(AmHeartJ,2008,155:130—137)。该文研究运动试验时血压升高过多是否会合并异常MPI增多。方法:病人7205名因胸痛或呼吸困难做运动试验,检查血压反应及锝^99m-SPECTMPI研究(1999—2005年)。
