摘要:
Sortilin was discovered in 1997 by Petersen and his colleagues.The single transmembrane receptor was isolated as a 95 kDa membrane glycoprotein by receptor-associated protein affinity chromatography and therefore initially named gp95.The cDNA library screening revealed sequence homology between the luminal segment of gp95/sortilin and the yeast vacuolar sorting protein 10 protein(Vps10p)domain(Petersen et al.,1997).The Vps10p domain was first identified in the Saccharomyces cerevisiae protein Vps10p that directs lysosomal enzymes from the Golgi to the vacuole(Marcusson et al.,1994).

摘要:
The choroid plexus(ChP)is a highly vascularized and secretory tissue in each of the brain ventricles that represents the key structure between the blood and the cerebrospinal fluid(CSF).Besides its essential role in CSF production and brain waste clearance pathways,the ChP also contributes to the regulation of central nervous system(CNS)immunosurveillance(Ghersi-Egea et al.,2018).Indeed,the ChP forming the blood-CSF barrier(BCSFB)regulates the entry of immune cells and solute molecules into the brain and vice versa.When antigen-specific,autoreactive immune activation occurs in the periphery,inflammatory cells migrate through the brain barriers towards the CNS(Strominger et al.,2018),initiating neuroinflammatory diseases such as multiple sclerosis(MS).

Synaptosome:Synapses are the most critical portion of neuron connections,necessary for cellular organization of the brain.Synapse integrity is uttermost important for healthy brain functioning.Any perturbation in the synapse structure and/or function initiates neurological disorders.Synapses are the prime targets that are smashed in almost all neurodegenerative diseases.The vital components of synapse are essential for neurotransmission,synaptic plasticity and overall synapse function.Synapse dysfunctions are well studied in Alzheimer’s disease(AD)and other neurodegenerative diseases(Gowda et al.,2021).The best way to study the synapse dysfunctions in neurological diseases is the biochemical analysis of“synaptosome”.

In neuroscience research,neuronal models are crucial tools for elucidating the molecular and cellular processes involved in the disorders of the nervous system.Facilitating easily and reproducibly executable studies,in vitro models,such as the SH-SY5Y cell line culture,help us explore the pathophysiological mechanisms of neurodegenerative diseases;they are also essential for the efficient screening of drugs for treating the diseases of the nervous system(Peng et al.,2021;Strother et al.,2021).

Ketamine,a multimodal dissociative anesthetic,produces powerful analgesia at subanesthetic doses in traumatically injured patients.As ketamine does not induce respiratory depression or hemodynamic instability,the Committee on Tactical Combat Casualty Care for the US military recommends the use of subanesthetic doses of ketamine for acute pain management(Butler et al.,2014).Additionally,ketamine may have immunomodulatory effects after injury at subanesthetic doses,mediating the balance of pro-and anti-inflammatory processes(Loix et al.,2011;De Kock et al.,2013).

Ischemic retinopathy is a retinal disease caused by ischemic attacks.Ischemia is a common pathologic mechanism in various retinal disorders and diseases such as age-related macular degeneration,diabetic retinopathy,glaucoma,or vascular occlusion(Osborne et al.,2004).Although various murine models have been developed to understand a series of metabolic mechanisms induced by retinal ischemia and further used to test promising therapeutics,effective treatment in ischemic retinopathy has not been clearly suggested.This is associated with the notion that the contributing pathologic metabolic pathways might be enormously complex.

The P2X4 receptor belongs to the P2X receptor family of trimeric ligand-gated ion channels and was the first member of this family for which a crystal structure was obtained(Kawate et al.,2009).This structure confirmed the trimeric stoichiometry of P2X receptors and subsequent studies from the same group revealed the orthosteric binding site of the natural ligand adenosine 5′-triphosphate(ATP)in a cleft between each adjacent subunit(Hattori and Gouaux,2012).Now synonymous with structural descriptions of P2X receptors,these original studies described the structure of each P2X4 receptor subunit as that resembling a dolphin.

摘要:
Hevin/Sparcl1(hereafter referred to as Hevin)is an extracellular matrix protein encoded by the SPARCL1 gene.Recently,it has been revealed that Hevin has various functions,such as synapse formation,neuronal migration,inflammation,and angiogenesis(Gongidi et al.,2004;Naschberger et al.,2016;Singh et al.,2016;Liu et al.,2021).In addition,genome-wide association studies uncovered de novo and familial mutations of the SPARCL1 gene associated with a risk for autism spectrum disorder(ASD)(De Rubeis et al.,2014).However,the relationship between ASD-associated Hevin mutant and cellular phenotype has not been clarified.Recently,we have reported that ASD-associated mutation in Hevin reduces secretion efficiency and induces endoplasmic reticulum(ER)stress caused by structural instability(Taketomi et al.,2022).In this perspective,we discuss the relationship between the molecular functions of Hevin and ASD risk(Figure 1A).Also,we introduce our recent findings that link ASD-associated Hevin mutant to the cellular phenotype of ASD.

通过干湿循环加速试验模拟了Al表面沉积沙漠浮土情况下的大气腐蚀过程,利用失重分析、扫描电镜(SEM)和大气腐蚀监测仪(ACM)研究了浮土中可溶性盐对Al的大气腐蚀的影响.结果表明:浮土中的可溶性盐会使发生大气腐蚀的相对湿度范围变宽,随着可溶性盐含量的提高A1的腐蚀不断恶化.在可溶性盐成分中,MgCl2对Al的腐蚀影响最大,Nacl次之,MgSO4和Na2SO4的影响较小.
