    • 摘要: 目的建立BALB/c-Nude裸小鼠为背景的人源化ACE2(hACE2)转基因裸小鼠动物模型。方法利用hACE2转基因小鼠与雄性BALB/c-Nude裸小鼠杂交获得F1代,将F1代hACE2小鼠与BALB/c-Nude裸小鼠回交获得F2代,再将F2代hACE2小鼠互交获得F3代hACE2转基因裸小鼠。对F3代中hACE2转基因裸小鼠的生长发育、生理指标及免疫指标与C57BL/6J野生型小鼠、hACE2小鼠和BALB/c-Nude裸小鼠对比分析。结果(1)hACE2转基因裸小鼠生长发育指标与C57BL/6J野生型小鼠、hACE2小鼠和BALB/c-Nude裸小鼠无明显差异。(2)hACE2转基因裸小鼠生理指标中,建立的hACE2转基因裸小鼠与BALB/c-Nude裸小鼠相似,病理解剖观察发现,小鼠体内均无胸腺。脏器系数结果与BALB/c-Nude裸小鼠比较,脾系数和肝系数出现显著性差异(P<0.05)。血常规检测指标与C57BL/6J野生型小鼠、hACE2小鼠比较,中性粒细胞百分比(NEU)、淋巴细胞百分比(LYM)和单核细胞百分比(MONO)均出现显著性差异(P<0.01)。(3)hACE2转基因裸小鼠免疫指标中,与C57BL/6J野生型小鼠和hACE2小鼠比较,CD45+CD3+、CD45+CD3+CD4+、CD45+CD11B+和CD45+NK+均出现显著差异(P<0.01),与BALB/c-Nude裸小鼠无明显差异。结论成功建立了BALB/c-Nude为背景的hACE2转基因免疫缺陷裸小鼠动物模型。

    • 摘要: 背景与目的:目前对于中晚期结直肠癌患者尚无统一有效的治疗方法,探索影响结直肠肿瘤发生、发展的因子,有利于提高老年患者的治疗效果.通过裸小鼠移植瘤实验探究缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)对人结直肠癌细胞增殖及成瘤性的影响.方法:采用RNA干扰技术下调HIF-1α的表达,通过蛋白质印迹法(Western blot)和反转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)验证细胞转染率;采用AnnexinⅤ-异硫氰酸荧光素(fluorescein isothiocyanate,FITC)/碘化丙啶(propidium iodide,PI)流式细胞术检测空白对照组(PBS组)、实验组(si-HIF-1α组)和阴性对照组(si-NC组)细胞的凋亡情况;将空白对照组、实验组和阴性对照组的细胞分别注入裸小鼠体内,进行肿瘤细胞移植瘤实验,然后测量各组移植瘤的体积和质量,评估抑制HIF-1α的表达水平对人结直肠癌细胞增殖的影响.结果:细胞转染后,si-HIF-1α组HIF-1α的表达降低.与空白对照组相比,实验组细胞的凋亡率明显上升(P0.05).结论:下调HIF-1α的表达水平,能够抑制人结直肠癌细胞的增殖及成瘤性.

    • 摘要: 目的 探讨双歧杆菌-单纯疱疹病毒胸苷激酶基因-丙氧鸟苷(BI-HSV-TK/GCV)系统对裸小鼠胃癌细胞增殖的抑制效应和分子机制.方法 建立裸小鼠胃癌实验模型,检测裸小鼠的体重、肿瘤体积.运用TUNEL法观察裸小鼠胃癌细胞的凋亡情况,运用Western blot实验、RT-PCR实验分别测定胃癌组织中Caspase 3、Caspase 8、Fas及FAP-1蛋白及其mRNA表达情况.结果 BI-HSV-TK/GCV系统能够改善裸小鼠的体重,有效抑制其胃癌肿瘤的体积,促进胃癌细胞的凋亡,诱导胃癌细胞凋亡相关基因Caspase 3、Caspase 8、Fas蛋白以及mRNA表达水平增加,FAP-1蛋白以及mRNA表达水平降低.结论 BI-HSV-TK/GCV系统能够有效抑制胃癌细胞在裸小鼠体内成瘤的体积,并促进胃癌细胞的凋亡.

    • 摘要: 裸小鼠是肿瘤学实验研究中应用最为广泛的一种动物模型。在动物实验中,理想的麻醉是实验顺利进行的前提,麻醉方法和剂量的选择将对整个实验过程有非常重要的影响。目前,戊巴比妥钠已广泛应用于实验动物的麻醉。但其相关研究仅限于普通小鼠,对裸小鼠重复麻醉的具体用量及麻醉效果鲜有报道。本文旨在通过戊巴比妥钠对裸小鼠重复麻醉实验的研究,为相关研究提供麻醉时的参考依据。

    • 摘要: 目的 复方中药金龙胶囊(JLC)提取物及其制备的粗多糖、粗蛋白的抗脑肿瘤效果及安全性评价.方法 运用外科原位接种(surgical orthotopic implantation)技术,将肿瘤块种植到裸小鼠颅内,建立小鼠原位模型.用FluorVivo Model 100荧光成像仪对所有小鼠的脑肿瘤进行非侵袭性的实时成像观察.通过对空白对照组(无肿瘤且溶剂为溶媒)、阴性对照组(溶媒)、阳性对照组(替莫唑胺32 mg/kg)、JLC提取物低(750 mg/kg)、中(1200 mg/kg)、高剂量组(1500 mg/kg)及粗多糖(100 mg/kg)、粗蛋白组(100 mg/kg)的肿瘤抑制率进行比较,评价经灌胃给予金龙胶囊提取物对小鼠原位模型肿瘤生长的影响.结果 给药25 d后,JLC低、中剂量组肿瘤抑制率均为34.63%,与阴性对照组相比差异无显著性;粗多糖组和粗蛋白组肿瘤抑制率分别为22.26%、29.01%,肿瘤重量分别为(0.178±0.048)g、(0.144±0.047)g,与阴性对照组相比差异无显著性;阳性对照组和高剂量组,抑制率分别为99.57%、61.90%,肿瘤重量分别为(0.001±0.001)g、(0.088±0.045)g,差异有显著性(P0.05).

    • 摘要: 目的 研究番茄红素(Lycopene,LP)对荷肝癌裸小鼠移植瘤生长的影响及其机制.方法 通过背部皮下注射接种肝癌HepG2细胞的方法建立荷肝癌裸小鼠模型,并分别腹腔注射5、10、20 mg/(kg·d)的LP和2 mg/(kg·d)的顺铂进行干预治疗,疗程为14 d.治疗后,观察各组裸小鼠的一般生存状态及肿瘤生长状况,称取瘤体质量并计算抑瘤率;通过苏木精-尹红(HE)染色观察肿瘤组织形态结构改变,末端标记法(TUNEL)检测肿瘤组织细胞凋亡状况,免疫组织化学法(IHC)观察肿瘤组织凋亡相关蛋白(caspase-3、Bax、bcl-2)表达.结果 经LP治疗14 d,荷肝癌裸小鼠饮食状况和精神状态明显改善,瘤体减小,活动度和硬度降低,肿瘤组织呈现细胞皱缩、片状坏死等病理性改变,凋亡细胞数量明显增多;瘤体质量显著减轻、抑瘤率显著升高;肿瘤组织中caspase-3和Bax表达显著上调,而bcl-2显著下调,Bax/bcl-2比值显著升高;LP上述作用均具有一定的剂量依赖性.结论 番茄红素对荷肝癌裸小鼠移植瘤生长具有一定的抑制作用,其机制可能与番茄红素调节凋亡相关基因蛋白表达而促进肿瘤组织细胞凋亡有关.%Objective To investigate the effects of Lycopene (LP) on xenografts growth in liver cancer of nude mice.Methods The nude mice models bearing liver cancer were made by injecting inoculation human liver cell line HepG2 and were treated with 5,10 and 20 mg/(kg·d) of LP and 2 mg/(kg·d) of cisplatin for 14 d.The rats' general states and tumor growth conditions were observed,and the histopathological change of tumor tissue was observed by HE staining.The tumor cells apoptosis was detected by TUNEL.The expression of caspase-3,Bax and bcl-2 protein was detected by immunohistochemistry (IHC).Results The food and mental condition was improved after nude mice bearing liver cancer was treated with LP for 14 d,the tumor was reduced,and the activity and hardness was decreased.There were necrosis presentation,tumor cell shrinkage and other pathological morphological changes that appeared,and the number of apoptotic cells in tumor tissue was increased.The tumor weights of LP groups were decreased and the inhibition rates were increased.The expression of caspase-3 and Bax was significantly increased,while the expression of bcl-2 protein was decreased,and the ratio of Bax/bcl-2 was significantly increased;all the above effects were dose-dependent.Conclusion LP has inhibitive effects on xenografts growth in liver cancer of nude mice,which is perhaps related to its effects of regulating the expression of apoptosis-related genes and protein to promote tumor cell apoptosis.

    • 摘要: Objective:To compare the differences of anatomical and histological characteristics of lymph nodes between BALB/c nude mice and BALB/c mice.Methods:Firstly,twenty BALB/c nude mice and twenty BALB/c mice were dissected by using a surgical microscope.Secondly,the differences of T cells and B cells at the lymph node were compared by the expressions of CD3 and CD20 immunohistochemistry dyes.Results:There were,on average,23 nodes per mouse contained within the large lymph node assembly in the BALB/c nude mouse.The anatomical features of the lymph node distribution in the nude mice were mainly found in the neck with relatively higher density.There were two lymph nodes both in the submandible lymph nodes group and in the superficial cervical lymph nodes group (the constituent ratios were 95% and 90%,respectively) in the BALB/c nude mice,but there were four lymph nodes (the constituent ratios were 95% and 90%,respectively) in the BALB/c mice.There were significant difference between the BALB/c nude mice and the BALB/c mice.Mostly there were two lymph nodes of deep cervical lymph nodes both in the BALB/c nude mice and the BALB/c mice (the constituent ratios were 95% and 100%,respectively).There were no significant difference between the BALB/c nude mice and the BALB/c mice.We confirmed that the number of CD3-positive T lymphocytes in lymph nodes of the nude mice decreased greatly as compared with the BALB/c mice.Expressions of CD3 in T cells were 95% and 100% in the BALB/c nude mice and in the BALB/c mice,respectively.There were significant differences between the BALB/c nude mice and the BALB/c mice.Expressions of CD2o in B cells were 95% and 100% in the BALB/c nude mice and in the BALB/c mice,respectively.There was no significant difference between the BALB/c nude mice and BALB/c mice.Conclusion:The anatomi cal pictures of lymph node distribution in the nude mouse will be benefit to those who are interested.The anatomical features of the lymph node local higher density in neck of the nude mouse and lack of CD3-positive T lymphocytes would be useful for obtaining a better understanding of localized lymph node metastasis of oral transplant tumors.%目的:研究裸小鼠淋巴结的解剖学和组织学特点.方法:使用手术显微镜解剖20只BALB/c裸小鼠和20只BALB/c小鼠,通过免疫组织化学方法比较了两者淋巴结中T淋巴细胞和B淋巴细胞表达的不同.结果:平均每只裸小鼠中包含23个淋巴结和一组大型淋巴结群.裸小鼠颈部淋巴结分布密度较高,其中颌下淋巴结及颈浅淋巴结在裸小鼠中大多数为2个(构成比分别为95%和90%),而在小鼠中大多数为4个(构成比分别为95%和90%),两者差异有统计学意义.无论小鼠还是裸小鼠的颈深淋巴结均为2个(构成比分别为100%和95%),两者差异无统计学意义.裸小鼠淋巴结中CD3阳性的T淋巴细胞减少.细胞膜和细胞质着色的CD3阳性表达的T细胞在裸小鼠的表达阳性率为15%,在小鼠的表达阳性率为100%,两者表达的差异有统计学意义.细胞膜阳性表达CD20的B细胞在裸小鼠中的阳性率为95%,在小鼠中的阳性率为100%,两者表达的差异无统计学意义.结论:裸小鼠淋巴结分布的解剖图对于初学者有一定的帮助,裸小鼠颈部淋巴结密度较高的解剖学特点和缺乏CD3阳性的T淋巴细胞将有助于更好地理解裸小鼠的淋巴结转移机制.

    • 摘要: 目的 研究大蒜素(allitridi)对荷子宫内膜癌裸小鼠移植瘤生长的影响及其机制.方法 取对数生长期子宫内膜癌Ishikawa细胞,皮下注射接种的方法制备荷子宫内膜癌裸小鼠模型,成瘤后取60只裸小鼠模型随机分为模型组、大蒜素高(40mg/kg)、中(20mg/kg)、低(10mg/kg)剂量组和顺铂2mg/kg组,每组12只;各治疗组均隔天腹腔注射给药1次,共给药治疗5次.治疗完成后剥取肿瘤组织并称量、计算抑瘤率;HE染色法观察肿瘤组织形态学变化;TUNEL染色法观察肿瘤组织细胞凋亡状况并计算凋亡指数(apoptosis index,AI),免疫组织化学法(immunohistochemistry,IHC)观察肿瘤组织bcl-2、Bax、caspase-3蛋白表达并进行半定量分析,计算Bax/bcl-2比值.结果 大蒜素各组和顺铂组裸小鼠移植瘤肿瘤组织细胞呈皱缩、片状坏死等病理性改变,凋亡细胞数量明显增多,以大蒜素高剂量组最为显著.与模型组比较,大蒜素高、中剂量组和顺铂组瘤体重量显著减轻(P <0.05,P<0.01),抑瘤率显著升高(P<0.01),肿瘤组织bcl-2蛋白表达显著下调(P <0.05,P<0.01),Bax和caspase-3蛋白表达显著上调(P <0.05,P<0.01),Bax/bcl-2表达比值显著升高(P<0.01).与顺铂组比较,大蒜素高剂量组Bax/bcl-2表达比值显著升高(P<0.05).结论 大蒜素对荷子宫内膜癌裸小鼠移植瘤生长具有抑制作用,其机制可能与大蒜素能够下调bcl-2表达、上调Bax和caspase-3表达、提高Bax/bcl-2表达比值进而促进肿瘤细胞凋亡有关.

    • 摘要: Objective To study the effect of gender on the tumorigenicity and growth trend after human hepato-cellular carcinoma HepG2 cells were inoculated into BALB/C nude mice. Methods Eighteen 4-6 weeks' old BALB/C nude mice were divided into two groups (male group and female group) according to the gender, with 9 in each group. All the nude mice scapular were inoculated with 0.2 mL HepG2 cells in logarithmic growth phase and at the concentra-tion of 1×107/mL. After 10 days, the tumor formation of the two groups was observed, and the tumor volume was mea-sured. Results The number of tumorigenicity model in female group was 4, and tumorigenicity rate was 44.44%. The number of tumorigenicity model in male group was 9, and tumorigenicity rate was 100.00%, with statistically significant difference (P=0.029). The transplanted tumor volume trend figure of the successful model showed that the tumor growth in male group was higher than that in the female group. Conclusion Tumorigenicity rate and growth rate of transplant-ed tumor have obvious gender difference after human hepatocellular carcinoma HepG2 cells were inoculated into BALB/C nude mice.%目的 研究性别对人肝癌细胞HepG2接种于BALB/C裸小鼠成瘤及移植瘤生长趋势的影响.方法 18只4~6周龄BALB/C裸小鼠雌雄各半,按照性别分成雌雄两组,每组9只,将全部裸小鼠肩胛部接种处于对数生长期且浓度为1.0×107/mL的HepG2细胞0.2 mL,10 d后观察两组的成瘤情况并测量肿瘤体积.结果 雌性组成瘤模型4只,成瘤率为44.44%;雄性组成瘤模型9只,成瘤率为100.00%,差异有统计学意义(P=0.029);造模成功的移植瘤体积走势图显示移植瘤生长速度雄性组大于雌性组.结论 人肝癌细胞HepG2接种于BALB/C裸小鼠成瘤率及移植瘤生长速度有明显的性别差异.
