    • 摘要: 目的:分析髓外造血的CT及MRI表现,提高其影像诊断准确率,降低漏误诊率。方法:回顾性分析经临床随访证实的14例髓外造血患者的影像资料,总结该病的特征性影像学表现。结果:14例中11例表现为脊柱两侧多发软组织肿块,其中4例伴肝大、脾大,脾大为著;3例伴胸膜丘状或结节状软组织肿块;2例伴胸骨旁软组织肿块。2例表现为脾大伴脾脏异常低密度灶。1例仅表现为胸骨旁软组织肿块。2例行CT增强扫描呈均匀轻度强化,脂肪成分不强化。2例行MRI平扫及增强扫描,T1WI呈等信号,T2WI呈低信号,DWI呈等信号,增强扫描呈轻度强化。结论:髓外造血的CT及MRI表现具有一定特征,结合临床资料能够提高其诊断正确率。

    • 摘要: Recent genomic studies have revealed the existence of many somatic gene mutations associated with hematologic malignancies.These somatic mutations drive the expansion from a single cell to a detectable hematopoietic clone.Clonal hematopoiesis (CH) is age-associated and related to increased risk of hematologic malignancy.Thrombopoietin (TPO) stimulates megakaryocytogenesis,platelet activation and hematopoietic stem cell(HSC) maintenance after combining with c-MPL on cell surface.Increasing researches have revealed the relationship between the level of TPO and CH.This article reviews the reasearch progress in the relationship between level of TPO and CH,in order to provide evidences for clinical prevention,diagnosis and treatment of hematologic malignancies.%目前,基因组学的发展揭示了许多与血液系统恶性肿瘤密切相关的体细胞基因突变,这些基因突变驱动单个细胞增殖为可检测到的造血克隆.克隆性造血(CH)与血液系统肿瘤发病风险增加有关.随着年龄增长,健康个体CH频率增加.血小板生成素(TPO)与其细胞表面c-MPI结合后,刺激巨核细胞生成、血小板活化和造血干细胞(HSC)维持.越来越多的研究发现,TPO水平与CH密切相关.笔者通过总结TPO水平与CH关联性的研究进展,旨在为血液系统肿瘤的预防和诊治提供思路.

    • 摘要: 多种化学和物理因素均能损害人类的血液系统,引起血液系统恶性疾病.造血功能重建是各种治疗血液系统恶性疾病方案的首要目的.造血干细胞移植(HSCT)是恢复血液系统恶性疾病患者造血功能的有效途径.探讨造血干细胞(HSC)的生物学特征是目前组织工程学研究的热点,其中包括间充质干细胞(MSC)和内皮祖细胞(EPC).HSCT联合输注MSC和EPC对HSCT后患者恢复造血功能有积极影响.笔者拟就MSC和EPC的生物学特征,及其对HSCT后患者造血功能重建的影响的研究进展进行综述.%A variety of chemical and physical factors can damage the human blood system,causing malignant diseases of blood system.Reconstruction of hematopoietic function is the essential purpose of various treatments for malignant diseases of blood system.Hematopoietic stem cell transplantation (HSCT) is an efficient way to reconstitute hematopoietic function.To explore the biological characteristics of hematopoietic stem cell (HSC) is a hotspot of tissue engineering research,including mesenchymal stem cell (MSC) and endothelial progenitor cell (EPC).HSCT combined infusion of EPC and MSC,has a positive effect on patients to reconstitute hematopoietic function.In this article,the biological characteristics and effects of MSC and EPC on hematopoietic function reconstitution after HSCT were reviewed respectively.

    • 摘要: 环状RNA(circRNA)为一类广泛存在于生物界的内源性非编码RNA,其主要结构特征为包含封闭环状结构,不具有5'-、3’-末端与多聚腺苷酸尾结构.封闭环状结构使circRNA不易被RNA外切酶降解,以及避免腺苷酸脱支酶的脱帽效应,因此circRNA的结构与功能均稳定.目前研究发现,circRNA主要参与转录及转录后基因表达的调控,尤其可以作为微小RNA(miRNA)海绵,充当竞争性内源RNA(ceRNA)调控靶基因的表达.circRNA直接或者间接参与骨髓造血发育及血液系统恶性肿瘤的发生、发展,因此其有望成为血液系统恶性肿瘤诊断的分子标志物与治疗靶点.为探讨circRNA与血液系统恶性肿瘤的恶性转化、疗效与预后之间关系,笔者拟就circRNA的分类、结构特征、生物学功能及其在骨髓造血发育及血液系统恶性肿瘤中作用的研究进展进行综述.%Circular RNA (circRNA) is a group of widely existed endogenous non-coding RNA.Its main structural feature is closed covalently cyclic loops and lack of the 5′-end,the 3′-end,and poly (A) tail,which resistant to degradation by RNA exonucleases and avoid debranching by adenylate debranching enzymes.Therefore,circRNA is stable in structure and function.Recent researches have revealed that circRNA mainly participates in transcriptional and posttranscriptional regulation,especially acts as microRNA (miRNA) sponge to regulate target gene expression as competitive endogenous RNA (ceRNA).circRNA is directly or indirectly involved in development of hematopoiesis in marrow and hematological malignancies,and is expected as a new diagnostic biomarker or molecular therapeutic target for hematological malignancies.In order to investigate relationships between circRNA and malignant transformation,therapeutic efficacy and prognosis of hematologic malignancies,this review summarizes the classification,characteristics,biological functions of circRNA and its role in hematopoiesis and hematologic malignancies.

    • 摘要: Objective To explore the effects of Saussurea involucrata cultured cell extracts on hematopoietie function in the mice with radiation injuries.Methods By using digital random table method,100 Kunming male mice were averagely divided into 5 groups:blank control group,pure radiation group,Saussurea involucrata cultured cell extracts high,medium and low dosage group.The high,medium and low dosage group were respectively administrated Saussurea involucrata cultured cell extracts with the dosages of 1.0 g/kg,0.5 g/kg,0.1 g/kg and the others were given distilled water for 28 days.Except blank control group,the other groups were given 2.5 Gy 60Co γray irradiation after daily intragastric administration for 14 days.The number of peripheral blood leukocytes in the mice was respectively checked 14 d and 1 d before irradiation and 3 d,6 d,10 d and 14 d after irradiation.The spleen index,the endogenous splenic nodule count,the bone marrow nucleated cells count and the bone marrow DNA content were respectively checked 10 d and 14 d after irradiation.Results Mice weight was all raised during administration in 4 dosage groups (t =76.58-203.34,P<0.01).Compared with those of blank group,the number of peripheral blood leukocytes,the bone marrow DNA content and the spleen index of pure radiation group were significantly decreased (P<0.01),but the endogenous splenic nodules significantly increased (P<0.01).Compared with those of pure radiation group,the number of peripheral blood leukocytes,the bone marrow DNA content,the spleen index and the endogenous splenic nodule count of Saussurea involucrata cultured cell extracts groups were significantly increased (P<0.05).The spleen index and endogenous splenic nodules were significantly increased (P<0.05).The result was not in a dosedependent manner.Conclusions Saussurea involucrata cultured cell extracts had the effect of injury repair and regeneration on hematopoietic system of radiation injured mice.%目的 探讨雪莲培养细胞提取物对辐射所致小鼠造血功能损伤的影响. 方法 100只昆明种雄性小鼠,采用数字随机表法平均分为空白对照组、单纯辐射组、雪莲培养细胞提取物高剂量组(1.0 g/kg)、中剂量组(0.5 g/kg)、低剂量组(0.1 g/kg),每组20只.雪莲培养细胞提取物给予相应剂量的雪莲培养细胞提取物灌胃,其他各组给予蒸馏水灌胃,连续28 d.除空白对照组外,其余各组于灌胃14 d后给予2.5 Gy60Coγ射线全身辐射.辐射前第14天、1天及辐射后第3、6、10、14天检测外周血白细胞数,辐射后第6、14天检测脾脏指数、内源性脾结节数、骨髓有核细胞数和骨髓DNA含量. 结果 各组小鼠体重在饲喂过程中均明显增加(t=76.58~203.34,P<0.01);与空白对照组比较,单纯辐射组小鼠外周血白细胞数、骨髓DNA含量、脾脏指数均明显下降(P<0.01),脾结节数均明显增加(P<0.01);与单纯辐射组相比,雪莲培养细胞提取物各组小鼠外周血白细胞数、骨髓DNA含量和骨髓有核细胞数均明显升高(P<0.05),脾脏指数与内源性脾结节数增加(P<0.05),检测结果不呈剂量依赖性. 结论 雪莲培养细胞提取物对辐射致造血系统损伤小鼠有修复和促再生作用.

    • 摘要: Long non-coding RNAs (lncRNAs) , which are closely related with human critical illness, can regulate gene expression and involve the physiological and pathological processes on epigenetic, transcriptional and post-transcriptional levels through several mechanisms. LncRNAs also play important regulation roles in the maturation and differentiation process of hematopoietic cell. Besides, in-depth exploration of their mechanism will provide new ideas for the diagnosis and treatment of blood diseases.%长链非编码 RNA(lncRNA)可在遗传水平、转录水平、转录后水平等多层面上通过多种机制调控基因表达,参与生理、病理过程,与多种人类重大疾病关系密切。 lncRNA 在造血细胞成熟、分化过程中也起重要的调控作用,深入探索其作用机制可为血液疾病的诊治提供新思路。

    • 摘要: “滴答”一秒的时间,世界上就有3个人需要输血了。但是目前血液无法制造,也无法替代,只有来自献血者的奉献。因此我们经常会在学校、单位或人流密集区看到有采血车在号召大家无偿献血。但是,民间或者网上还是流传着“献血会造成血液功能的弱化甚至丧失,增加患高血压或者冠心病等疾病的风险”这样的说法。
