摘要:
目的:探讨E选择素、P选择素在溃疡性结肠炎 (UC) 大鼠肠黏膜中的表达水平及意义.方法:选取2018年3-6月健康的雄性 (Sprague Dawkey, SD) 大鼠65只, 分为空白对照组 (n=12) 和实验组 (n=53) .空白对照组不处理, 于实验组中随机选取12只SD大鼠进行预实验, 筛选最合适剂量的TNBS (2, 4, 6-三硝基苯磺酸) 造模剂构建溃疡性结肠炎的模型.剩余大鼠进行正式造模, 按不同的解剖时间将其分为A、B、C组, 各12只.通过每天观察体重及一般情况 (精神状态, 粪便形状、颜色等) 的改变, 依据疾病活动指数 (disease activity index, DAI) 评分, 记录实验大鼠状态, 初步判断大鼠溃疡性结肠炎的严重程度.分别于第3、7、14天处死大鼠, 使用大体形态损伤指数 (colon macroscopic damage index, CMDI) 对结肠评分, 判断溃疡程度, 采用ELISA法检测肠黏膜中E选择素、P选择素的含量指标, 同时评估结肠病理改变, 综合得出E选择素与P选择素的含量与溃疡程度的关系.结果:实验组大鼠的DAI、CMDI评分均明显高于空白对照组, 且实验组结肠组织的病理情况镜下溃疡改变显著.空白对照组、A、B、C组间的E选择素、P选择素含量比较, 差异均有统计学意义 (P<0.05) .溃疡性结肠炎由逐渐严重至恢复的过程中, 各实验组肠黏膜组织中的E选择素、P选择素水平均先逐渐升高随后下降, 但C组含量仍均明显高于A组.结论:在溃疡性结肠炎发展过程中, 随着结肠溃疡程度加重, E选择素和P选择素水平升高.随着结肠溃疡程度减轻, E选择素和P选择素水平降低.E选择素与P选择素可能起到了促进UC病情发展的作用, 可作为UC诊断的动态参考指标, 以及为临床研究UC发病机制提供新的方向.%Objective:To investigate the expression levels and significance of E-selectin and P-selectin in intestinal mucosa of rats with ulcerative colitis (UC).Method:A total of 65 healthy male (Sprague Dawkey, SD) rats were selected from March to June 2018, they were divided into blank control group (n=12) and experimental group (n=53).The blank control group was not treated, 12 SD rats were randomly selected from the experimental group for the pre-experiment, and the most appropriate dose of TNBS (2, 4, 6-trinitrobenzene sulfonic acid) was screened to build the model of ulcerative colitis.The remaining rats were formally moulded and divided into group A, group B and group C according to different anatomical time, with 12 rats in each group.Through daily observation of body weight and changes in general conditions (mental state, shape and color of feces, etc.), according to disease activity index (DAI) score, the status of experimental rats were recorded and the severity of ulcerative colitis in rats was preliminarily determined.Rats were sacrificed on 3, 7 and 14 d respectively, and colon macroscopic damage index (CMDI) was used to assess the degree of ulceration.ELISA method was used to detect the content indexes of E-selectin and P-selectin in intestinal mucosa, and meanwhile the pathological changes of Colon were evaluated.The correlation between the content of E-selectin and P-selectin and the degree of ulcer was obtained.Result:The DAI and CMDI scores of rats in the experimental group were significantly higher than those in the blank control group, and the pathological changes of colonic ulcer in the experimental group were significantly changed under the microscope.The contents of E-selectin and P-selectin in blank control group, A, B and C groups were compared, the differences were statistically significant (P<0.05).In the course of recovery from ulcerative colitis, the levels of E-selectin and P-selectin in intestinal mucosa of experimental group increased gradually at first and then decreased, but the contents of E-selectin and P-selectin in group C were still significantly higher than those in group A.Conclusion:During the development of ulcerative colitis, the levels of E-selectin and P-selectin increased with the aggravation of the degree of colon ulcer.The levels of E selectin and P-selectin decreased with the decrease of degree of colon ulcer.E-selectin and P-selectin may play a role in promoting the development of UC, which can be used as a dynamic reference indicator for the diagnosis of UC and provide a new direction for the clinical study on the pathogenesis of UC.
