摘要:
Objective :To reveal the possible mechanism of Fuzheng Jiedu Huayu granule on myocardial pro-tection of multiple organ injury induced by Klebsiella pneumoniae ,through cytokines and myocardial histopathology . Methods :128 KM mice were randomly divided into control group ,model group ,western medicine group and tradi-tional Chinese medicine group with 32 animals in each group .The multi-organ injury model was prepared by direct injection of Klebsiella pneumoniae by tracheal intubation method .8 mice were sacrificed on the 1st ,3rd ,5th ,and 7th days after modeling respectively ;the heart was taken out and divided into two parts .One part of the heart tissue was homogenized and the levels of TNF-α ,IL-1β ,IL-6 ,IFN-γ ,IL-10 were detected in different infection phases ;the oth-er part of the heart tissue was HE staining ,and the pathological changes of different infection phases were observed under light microscope.Results :Compared with the control group ,the cytokines level of TNF-α ,IL-1β ,IL-6 ,IFN-γ ,IL-10 in the myocardial tissue of the model group were significantly increased on the 1st and 3rd day after infec-tion ,and the pathological damage of myocardial tissue was obvious .Compared with the model group ,on the 1st day after infection ,the level of pro-inflammatory cytokines such as TNF-α ,IL-1β ,IL-6 and IFN-γ in myocardial tissue were significantly decreased ,and the level of anti-inflammatory cytokine IL-10 was obviously increased in the tradi-tional Chinese medicine group .Conclusion :Pro-inflammatory/anti-inflammatory cytokines are involved in the dam-age of myocardial tissue in pneumonia mice .Fuzheng Jiedu Huayu granule can reduce the expression levels of pro- inflammatory cytokines and improve the expression level of anti-inflammatory cytokine in myocardial tissue ,thereby reducing the degree of pathological damage of myocardial tissue and playing a protective role on myocardial tissue .%目的:从心肌组织细胞因子及病理角度揭示扶正解毒化瘀颗粒对克雷伯杆菌肺炎所致小鼠多器官功能损伤心肌保护的可能作用机制.方法:将128只KM小鼠随机分为对照组、模型组、西药组和中药组,每组32只;采用气管插管法直接注入肺炎克雷伯杆菌制备多器官功能损伤模型;分别于造模后第1 、3 、5 、7天每组各取8只小鼠;取出心脏,一部分心脏组织匀浆,待测不同感染时相细胞因子TNF-α 、IL-1β 、IL-6 、IFN-γ 、IL-10水平;另一部分心脏组织 HE染色,光学显微镜下观察不同感染时相病理变化.结果:与对照组比较,模型组小鼠在感染后第1天、第3天心肌组织中TNF-α 、IL-1β 、IL-6 、IFN-γ 、IL-10等细胞因子水平显著增高,心肌组织病理损伤明显;与模型组比较,中药组小鼠在感染后第1天心肌组织中T N F-α 、IL-1β 、IL-6 、IFN-γ等促炎细胞因子水平显著降低,抗炎细胞因子IL-10水平显著增高.结论:促炎/抗炎细胞因子参与了肺炎所致小鼠心肌组织的损伤,扶正解毒化瘀颗粒能够降低心肌组织中促炎细胞因子表达,提高抗炎细胞因子表达,从而减轻心肌组织的病理损伤程度,对心肌组织起到一定保护作用.
