该研究旨在分析评估甘肃省河西走廊地区羊肉中重金属含量,为消费者提供食品安全保障。在河西走廊地区5个地级市采集羊肉及羊肝样品100份,采用电感耦合等离子体质谱法(ICP-MS)和原子荧光法对样品中重金属铅(Pb)、镉(Cd)、汞(Hg)、砷(As)、铬(Cr)5种元素的含量进行检测。结果表明:羊肉样品中铅、铬、砷、镉元素平均含量分别为0.206±0.160 mg/kg、0.141±0.169 mg/kg、0.055±0.087 mg/kg和0.160±0.189 mg/kg,汞元素未检出。重金属平均值铅>铬>镉>砷>汞,依据GB 2762-2017判定均无超标;羊肝样品中重金属含量高于羊肉样品;嘉峪关、酒泉、金昌3个地区的重金属检测值略高,差异不显著(P>0.05)。建议有关部门加大冶金工厂的三废排放监督力度,合理选择肉羊养殖场场所,定期对饲料和水进行质量安全监控,定期对羊肉产品进行质量安全检测。

摘要:
本文以冷冻羊肝为原料,加入自制复合发酵菌种,通过响应面法优化得到最佳发酵工艺。用单因素试验研究鸡油添加量、混合糖粉比例、发酵底物湿度对发酵综合评分的影响,发酵综合评分中的pH以5.0为最佳标准,筛选出参数并采用Box-Benhnken响应面法优化三个因素获得了最佳发酵工艺。结果表明,最佳发酵工艺参数为每百克羊肝泥发酵底物湿度43%,鸡油添加量24 g,7 g混合糖粉比例为乳糖:蔗糖(W∶W)3.2∶3.8。在此条件下,37°C发酵7 d,发酵综合得分为97.01分,和预测值96.71分基本一致,说明复合微生物发酵剂发酵羊肝工艺可行。

为优化羊肝蛋白酶解工艺条件及探讨其体外抗氧化活性,以水解度为评价指标,采用碱性蛋白酶酶解羊肝,在单因素试验基础上,通过响应面法优化羊肝蛋白的酶解工艺条件.结果表明,羊肝蛋白最佳酶解条件为酶解温度51°C、pH 8.5、酶解时间4.1h、加酶量0.40%.在此条件下,进行3次验证试验,测得羊肝酶解液实际水解度为(40.31±0.24)%.体外抗氧化试验结果表明,羊肝酶解产物具有一定的抗氧化活性,其清除羟自由基(·OH)、超氧阴离子自由基(O2-·)和DPPH自由基的IC50值分别为17.01 mg/mL、13.21 mg/mL和10.42 mg/mL,并具有一定的还原能力.%In order to optimize the sheep liver enzymolysis technology and explore itsin vitro antioxidant activity,using hydrolysis degree as evalua tion index,sheep liver was hydrolysed by alkaline protease.On the basis of single factor experiments,the enzymatic hydrolysis conditions of sheep liver proteins were optimized by response surface methodology.The results showed that the optimum enzymatic hydrolysis conditions were as follow:hydrolysis temperature 51 °C,pH 8.5,hydrolysis time 4.1 h and enzyme addition 0.40%.Under the conditions,through three validation tests,the actu al hydrolysis degree of sheep liver hydrolysates was (40.31 ±0.24)%.The results ofin vitro antioxidant experiments showed that sheep liver hydrolysates had a certain antioxidant activities.The 1C50 of its scavenging hydroxyl radical,superoxide anion and DPPH free radical were 17.01 mg/ml,13.21 mg/ml and 10.42 mg/ml,respectively.Sheep liver hydrolysates had a certain reduction capacity.

Using the sheep liver as raw material,the hydroxyl free radical scavenging rate and peptide yield of enzymatic hydrolysis solution was obtained from sheep liver,the activity of five protease were determined.The enzyme screening results showed that hydroxyl free radical scavenging rate (84.50%) and peptide yield (22.49%) of enzymatic hydrolysis solution by flavor protease were the optimal.On the basis of single factor experiments,with hydroxyl free radical scavenging rate as evaluation index,the optimum enzymolysis conditions were optimized by response surface method.The results showed that the optimum hydrolytic conditions of sheep liver antioxidant peptides preparation were material-liquid ratio 1:3 (g:ml),enzyme addition 2 400 U/g,enzymolysis temperature 50 °C,pH 7.50 and enzymolysis time 2.8 h.Under the conditions,the actual hydroxyl free radical scavenging rate was 93.70%,which was close to the model theoretical value.The total amino acid content in the sheep liver enzymolysis products was 57.23 g/100 g,in which the contents of hydrophobic amino acid and essential amino acid were 46.47% and 41.63%,respectively.It indicated that sheep liver enzymolysis products by flavor protease had high antioxidant activity and nutritional value.%以羊肝为原料,分别测定5种蛋白酶酶切羊肝所得的酶解液的羟基自由基清除率和肽得率.结果表明,风味蛋白酶羟自由基清除率(84.50%)和肽得率(22.49%)最好.在单因素试验基础上,以羟基自由基清除率为评价指标,采用响应面法优化最佳酶解条件.结果表明,羊肝抗氧化肽制备的最佳酶解条件为料液比1∶3 (g∶mL)、加酶量2400 U/g、酶解温度50°C、pH7.50、酶解时间2.8h.在此优化条件下,测得实际羟自由基清除率为93.70%,与模型理论值相接近.羊肝酶解产物中总氨基酸含量为57.23 g/100 g,其中疏水性氨基酸和必需氨基酸占总氨基酸含量分别为46.47%和41.63%,表明以风味蛋白酶酶解羊肝产物具有较高的抗氧化活性和营养价值.

采用原子吸收法和原子荧光法对阜新市市售不同批次的科尔沁、朝阳及阜新出产的羊肉与羊肝的Zn、Cu、Pb、Cd、Cr、Hg、As的含量进行检测.结果表明,三个地区均表现为羊肝中的重金属含量显著高于羊腿肉和羊背肉,而羊腿肉和羊背肉之间无显著差异.羊肉与羊肝中重金属含量表现为:科尔沁>朝阳>阜新,其中科尔沁显著高于其它两地区,仅科尔沁地区羊肝中As和Pb含量超标.%The heavy metal(Zn,Cu,Pb,Cd,Cr,Hg and As) content were determined by atomic absorption and atomic fluorescence spectrometry in the mutton and sheep liver of different batches(Horqin,Chaoyang and Fuxin).Results showed that Fuxin,Chaoyang and Horqin all showed that the heavy metal content in liver was higher than that in leg meat and back meat.However there was no significant difference between the sheep's leg meat and sheep's back meat.The contents of heavy metals in mutton and sheep liver in three areas indicated that Horqin>Chaoyang>Fuxin.The heavy metal content of Horqin was higher than that of the other two regions.Only the contents of As and Pb exceeded food national standard in Horqin region.
