The experiment is to study the security of two kinds of herbal prescriptions by testing the herbal prescription acute toxicity in mice.In this study, two herbal prescriptions were prepared and fed to the mice, with high, medium and low dose as experimental groups and one blank as control group, once a day and lasting for 7d.During the 7d, the mice behavior was observed.After 7d, blood and tissue organs were collected to determine blood indicators and observe pathological changes.The results showed that: (1) Comparing No.I prescription with the control group, the blood indicators of mice such as WBC, GRAN%, RBC decreased significantly (P<0.05).But the liver organ coefficient of high dose group decreased extremely (P<0.01), medium dose group was decreased significantly (P<0.05).The liver pathological changes of each group all presented a strong to weak trend.(2) Comparing No.Ⅱ prescription with the control group, the blood indicators of mice such as WBC, GRAN%, RBC were decreased significantly.The liver organ coefficients of high and medium dose group were decreased significantly (P<0.01), only the high dose group had liver pathological changes.Therefore, it is concluded that No.Ⅰ prescription had toxic effects on the liver, which was dependent on the dose.The high dose of No.Ⅱprescription had toxic effects on the liver, while medium and low-dose had no toxic effect.%为研究两种缓解马应激中草药方剂的安全性,进行了中草药对小鼠的急性毒性试验.配制Ⅰ、Ⅱ号两种中草药方剂,各设高、中、低3个剂量试验组和1个空白对照组,每天灌服1次,连续灌服7 d,每天观察小鼠行为等情况, 7 d后,采集血液和组织脏器,测定血常规并观察组织病理变化.结果显示:1)Ⅰ号方剂与对照组相比,小鼠WBC、GRAN%、RBC等血液指标显著降低(P<0.05),高剂量组肝脏脏器系数极显著降低(P < 0.01),中剂量组肝脏脏器系数显著降低(P<0.05),各剂量组肝脏病理变化呈现由强到弱趋势;2)Ⅱ号方剂与对照组相比,小鼠WBC、GRAN%等血液指标显著降低(P<0.05),高、中剂量组肝脏的脏器系数均显著降低(P<0.05),仅高剂量组肝脏有病理变化.结论:Ⅰ号方剂对肝脏有毒副作用,作用大小呈现剂量依赖性;Ⅱ号方剂高剂量对肝脏有毒副作用,中、低剂量无毒副作用.
