The hippocampus is involved in important brain functions such as learning and memory,spatial navigation,fear processing,and social behavior(Dudek et al,2016).The most prominent areas of the hippocampus are typically denoted as the dentate gyrus and the three areas of the cornu ammonis(CA1,CA2,and CA3).Discovered by Lorente de Nó(1934),the CA2 region of the hippocampus is a relatively small area interposed between CA3 and CA1 that forms the nexus linking the input of the entorhinal cortex to the output of CA1(Chevaleyre and Siegelbaum,2010).

Selective vulnerability of excitatory neurons in Alzheimer’s disease(AD):AD is the most common form of dementia;however,the pathogenesis of AD is largely unknown.One of the characteristic features of AD is the formation of intracellular neurofibrillary tangles(NFTs).NFTs are abnormal accumulates of misfolded tau protein,which may eventually cause neuronal death and neurodegeneration(Jack et al.,2018).In the early stages of AD progression,not all neurons are equally vulnerable to tau aggregates.Previous studies have shown that large pyramidal neurons in the entorhinal cortex(EC)are specifically vulnerable to pathological tau accumulation(Fu et al.,2017).This selective vulnerability of excitatory neurons to tau pathology is one of the fundamental questions needed to be answered in AD research.

The adult cortex has long been regarded as non-neurogenic.Whether injury can induce neurogenesis in the adult cortex is still controversial.Here,we report that focal ischemia stimulates a transient wave of local neurogenesis.Using 5′-bromo-2′-deoxyuridine labeling,we demonstrated a rapid generation of doublecortin-positive neuroblasts that died quickly in mouse cerebral cortex following ischemia.Nestin-Cre^(ER)-based cell ablation and fate mapping showed a small contribution of neuroblasts by subventricular zone neural stem cells.Using a mini-photothrombotic ischemia mouse model and retrovirus expressing green fluorescent protein labeling,we observed maturation of locally generated new neurons.Furthermore,fate tracing analyses using PDGFRα-,GFAP-,and Sox2-Cre^(ER) mice showed a transient wave of neuroblast generation in mild ischemic cortex and identified that Sox2-positive astrocytes were the major neurogenic cells in adult cortex.In addition,a similar upregulation of Sox2 and appearance of neuroblasts were observed in the focal ischemic cortex of Macaca mulatta.Our findings demonstrated a transient neurogenic response of Sox2-positive astrocytes in ischemic cortex,which suggests the possibility of inducing neuronal regeneration by amplifying this intrinsic response in the future.

随着社会快速发展,系统性、行业性火灾风险持续增加,对人们生命财产安全造成严重威胁。为址,全国各地先后新建、改建消防综合训练基地,以提升复杂灾情条件下消防救援队伍灭火救援综合能力。然而传统实战训练演练虽能有效提高消防救援队伍整体救援能力,但其依然存在弊端。为有效解决这些弊端,基于集成Cortex嵌入式技术,构建出一套消防装备模拟训练系统,测试结果显示,基于Cortex的消防装备模拟训练系统稳定性较强,训练成本低、效率高。

The prelimbic cortex(PL),a key brain.region of the medial prefrontal cortex(mPFC),plays an important role in cognitive ability,emotional regulation and pain modulation.However,there was stil a lack of systematic connective mapping in the PL.Therefore,to analyze the afferent projections to the mouse PL,the fluorescence gold(FG)and rabies virus(helper viruses(AAV2/9-DIO-EGFP-TVA and AAV2/9-DIO-G)and RV-EnvA-OG-dsRed)were injected respectively into the PL of C57 mice,PV-Cre and CaMKII-Cre mice.The FG labeled projections neurons were observed in some cortex regions,including mPFC,anterior cingulate cortex(ACC),insular cortex(IC),claustrum(CLA).They were also found in some nuclei of the thalamus,such as paraventricular nucleus of thalamus(PVT),mediodorsal nucleus of thalamus(MD),paratenial nucleus of thalamus(PT),centrolateral nucleus of thalamus(CL),.ventromedial nucleus of thalamus(VM),rhomboid nucleus of thalamus(Rh),and reuniens nucleus of thalamus(Re).

The basal ganglia is a critical subcortical nuclei in several parallel and closed circuits,which mainly modulate thalamus-cortex activity.Dysfunction in any one of these loops connecting cortex can lead to movement and/or mood disorders.To understand cellular complexity,differential gene expression and transcriptional regulatory network of human basal ganglia under normal condition,we performed single cell RNA sequencing of 2424 cells sampled from the basal ganglia of human postmortem brains including caudate,putamen,substantia nigra and globus pallidus.

Ventral tegmental area(VTA),which is well-documented as a modulatory center of reward and addiction,has been demonstrated to participate in pain processing.In addition,insula cortex(IC)has also been proved to be related with maintenance of neuropathic pain.In the present study,in order to clarify whether there exist dopaminergic projections from the VTA to the IC,retrograde and anterograde tracing methods combined with immunofluorescent histochemical staining were applied.The results revealed that almost all VTA-IC-projecting neurons originated from the VTA were dopaminergic neurons,with smaller amount of GABAergic neurons sending projection fiberss to the IC.
