    • 摘要: the author research the Acute toxicity tests of PVP -I ,cupric sulfate and cypermethrin on the ParalichthysLethostigma juveniles(70 d) In this study .The results showed that the safe concen‐tration of the PVP -I to ParalichthysLethostigma juveniles was 4 .062 mg/L ,the semi -lethal con‐centrations of 24 h was 55 .296 mg/L and 34 .560 mg/L was the semi-lethal concentrations of 48 h . The safe concentration ,of cupric sulfate was 0 .667 mg/L .the semi-lethal concentrations of 24 h was 2.812 mg/L and 2 .600 mg/L was the semi -lethal concentrations of 48 h .The safe concentration of effective cypermethrin was 0 .079 mg/L .Semi -lethal concentrations of 24 h was 7 .112 μg/L and 2.371 μg/L was its semi-lethal concentrations of 48 h .%研究了聚维酮碘、硫酸铜、高效氯氰菊酯三种药物在一定条件下对漠斑牙鲆幼鱼(70日龄)的毒性效应。结果表明:聚维酮碘的安全浓度为4.062 g/m L ;24 h半致死浓度为55.296 m g/L ,48 h半致死浓度为34.560 m g/L。硫酸铜的安全浓度为0.667 m g/L ,24 h半致死浓度为2.812 m g/L ,48 h半致死浓度为2.600 m g/L。高效氯氰菊酯的安全浓度为0.079μg/L ,24 h半致死浓度为7.112μg/L ,48 h半致死浓度为2.371μg/L。

    • 摘要: 应用解剖、常规石蜡切片和显微摄影等技术,观察了漠斑牙鲆消化道的组织结构,对饥饿前后漠斑牙鲆的食道、胃和肠道纹状缘高度、柱状细胞高、壁厚、肌肉层厚以及食道、肠道杯状细胞大小进行了测量。结果表明,漠斑牙鲆消化道由内向外分为粘膜层、粘膜下层、肌肉层和浆膜层。食道粘膜上皮为复层上皮,胃本体和肠道粘膜上皮为单层柱状上皮,直肠粘膜上皮为假复层纤毛柱状上皮。除胃本体外,从食道到直肠粘膜上皮中均有杯状细胞分布,但其数量呈减少趋势。粘膜下层主要由疏松结缔组织构成。食道肌肉层为内纵外环的横纹肌,胃本体、肠道及直肠肌肉层为内环外纵的平滑肌。漠斑牙鲆饥饿10 d后各段消化道的褶皱高度降低,纹状缘边界不光滑,甚至局部断裂,肌层和粘膜下层连接疏松。消化道显微结构的测量结果显示,饥饿后粘膜上皮中的杯状细胞显著变小,胃、肠、直肠的柱状上皮细胞高度显著下降,肠前段和中段肌肉层厚和壁厚度呈显著下降。%The techniques of anatomy, paraffin section and photomicrography were used in this paper. Histological structure of the digestive tract of Paralichthys lethostigma and histological changes including thickness of striated border, height of columnar cell, thickness of wall and muscle layer in digestive tract as well as the size of mucous cell in esophagus and intestine following starvation for 10 days were assessed by light microscopy. The results show that the digestive tract of Paralichthys lethostigma consisted of mucosa, submucosa, muscles and serosa layer. The mucosal epithelium of the esophagus was stratified squamous epithelium, while epithelium in stomach and intestine’s was simple columnar epithelium, and it was pseudostratified ciliated columnar epithelium in rectum. Goblet cells existed between the epithelial cells in digestive tract except stomach. Submucosa layers were loose connective tissue. The muscle of esophagus was striated muscle arranged in the inner longitudinal and outer circular. However, in other segments of digestive tract, the muscle was composed of smooth muscle cells and the cells arrangement patterns were opposite to that of esophagus musclar layer. Starvation for 10 days resulted in the decrease of height of mucosa rugae, disruption and exfoliation of the striated border, and the connection between the size of goblet cells significantly reduced. The height of columnar cells in stomach, intestine and rectum, as well as the thickness of muscle layer and the wall of foregut and midgut significantly decreased. This study provided scientific data for understanding Paralichthys lethostigma to the starvation tolerance and artificial culture of the fish.

    • 摘要: The role of the endocrine pathway in the regulation of southern flounder Paralichthys lethostigma reproduction has been unclear. Here we aimed to explore the underlying mechanisms by investigating the expression pattern of luteinizing hormoneβ subunit (LHβ). Using samples isolated from pituitary ofP. we obtained the full-length cDNA sequence of LHβ with the RACE method. The spatial and temporal expression patterns ofLHβmRNA in different tissues during an ovarian maturation cycle were analyzed using the qPCR assay. The levels of plasma LH and E2during an ovarian maturation cycle were also measured using the ELISA method. Our results revealed an asynchronous oocyte development and batch spawning in southern flounder. The cDNA sequence of LHβhad a length of 597 bp and it encoded 145 amino acids. LHβ expression was detected in all examined tissues except for muscle, which implied it could be involved in multiple physiological functions in southern flounder. The LHβ mRNA level in pituitary was high through the entire ovarian maturation cycle, and peaked at stage V and VI. In the ovary the LHβ mRNA level peaked at stage V. In the liver the LHβ mRNA level was the highest at stage II and relatively high at stage V. The levels of plasma LH and E2 fluctuated in a similar pattern during the ovarian maturation cycle. These results suggested that LHβmight regulate the ovarian maturation cycle via endocrine, paracrine and autocrine pathways. Our study provided insights into the functions of the endocrine pathway in the reproduction of southern flounder.%利用cDNA末端快速克隆(Rapid Amplification of cDNA Ends, RACE)方法获得了漠斑牙鲆(Paralichthys lethostigma)促黄体素(Luteinizing Hormone, LH)β亚基的cDNA全长序列,检测了LHβ亚基mRNA的组织表达水平,揭示了垂体、肝脏和卵巢中LHβ亚基mRNA在卵巢发育周期中的表达水平变化,利用酶联免疫技术(Enzyme-Linked Immuno Sorbent Assay, ELISA)测定了血浆LH和雌二醇(Estrodiol, E2)表达水平的变化。结果表明,漠斑牙鲆为卵巢非同步发育分批产卵性鱼类, LHβ亚基cDNA序列全长597bp,开放阅读框(Open reading frame, ORF)长438 bp,编码145个氨基酸,LHβ亚基mRNA具有广泛的组织分布特性。与肝脏和卵巢相比,垂体中LHβ亚基mRNA表达水平在卵巢发育各个阶段都有较高表达水平,在IV期表达水平迅速升高至较高水平并保持至VI期。卵巢中LHβ mRNA表达水平在V期达到峰值,而肝脏中LHβ mRNA表达水平在II期时达到峰值,在V期也有相对较高的表达水平。血浆LH和E2表达水平在卵巢发育周期中均呈现趋势一致的规律性变化。研究结果可为认识漠斑牙鲆生殖调控机制提供基础资料。

    • 摘要: 对人工养殖漠斑牙鲆肌肉进行一般营养组分、氨基酸和脂肪酸组成分析,并对其营养品质进行评价.结果表明:漠斑牙鲆肌肉鲜样中粗蛋白和粗脂肪含量分别为17.74%和3.02%.肌肉粗蛋白中检测出19种氨基酸,其中谷氨酸(120.4mg/g pro)、天冬氨酸(75.4mg/g pro)、甘氨酸(73.6mg/g pro)、亮氨酸(72.3mg/g pro)、赖氨酸(67.8mg/g pro)和精氨酸(63.4mg/g pro)含量较高.依据世界粮农组织/世界卫生组织/联合国大学(FAO/WHO/UNU)必需氨基酸基准模式,10种必需氨基酸评分均大于100,表明漠斑牙鲆肌肉蛋白是氨基酸平衡性较好的优质蛋白.脂肪酸检测结果显示漠斑牙鲆肌肉中饱和脂肪酸、单不饱和脂肪酸和多不饱和脂肪酸含量分别为32.23%、30.79%和27.01%.n-3系列多不饱和脂肪酸中二十碳五烯酸(EPA,C20∶5n-3)和二十二碳六烯酸(DHA,C22∶6n-3)含量较高,为7.38%和12.07%;n-6系列多不饱和脂肪酸中花生四稀酸(AA,C20∶4n-6)含量最高,为4.49%.n-6和n-3系列多不饱和脂肪酸比值为0.35,远低于我国和英国卫生部推荐的4.0最高安全限值.

    • 摘要: 为探讨工厂化养殖模式中,高浓度氨氮对养殖生物的生理影响,本试验以漠斑牙鲆为研究对象,研究了工厂化养殖模式不同氨氮浓度对其血液指标的影响.试验设置5个氨氮浓度梯度,分别对漠斑牙鲆血液生理指标进行了测定.结果显示,不同氨氮浓度对各项血液指标产生显著影响,随着氨氮浓度的上升,血浆总蛋白与白蛋白含量下降;谷丙转氨酶(GPT)、谷草转氨酶(GOT)与血清尿素氮含量升高;血清肌酐含量先升高后降低.结果表明,高浓度的氨氮刺激了漠斑牙鲆导致其生理调节机制出现紊乱,可能造成鱼类红细胞溶血,从而导致鱼类贫血,呼吸能力减弱并且影响鱼类肾的调节机能.

    • 摘要: A self-designed drawer biofilter used in recirculating aquaculture system for Paralichthys lethostigma,the research is about the water treatment effect of drawer biofilter,weight gain and feed utilization rate.The results show that the P.lethostigma (225.4 ± 11.9) g during follow-up of 60 days culture in recirculating system gets a weight gain rate of 49.97% (337.5 ±10.3) g,the feed coefficient of 1.06,rearing density reaches 24.1 kg/ m3 and survival rate amounts to 100%.The removal efficiencies of drawer biofdter about NH4+ -N、NO2- -N and COD are (10.61 ±1.88)%,(14.90 ±3.06)% and (16.11 ±1.704)%,respectively.Therefore,using drawer biofilter to breed P.lethostigma in recirculating aquaculture system is feasible.%采用自行设计的抽屉式生物滤器应用于漠斑牙鲆(Paralichthys lethostigma)闭合循环水养殖系统,研究其对循环养殖水的处理效果及漠斑牙鲆的增重和饲料利用率的影响.结果表明:经过60 d的循环水养殖,漠斑牙鲆从初始时的(225.41±11.9)g增加到结束时的(337.5±10.3)g,增重率49.97%;试验饲料系数1.06,养殖密度24.1kg/m3,成活率100%;抽屉式生物滤器对于NH4+-N、NO2- -N和COD去除率分别为(10.61±1.88)%、(14.90±3.06)%和(16.11±1.70)%,可满足漠斑牙鲆养殖水体的水质要求.

    • 摘要: A repeat-enriched genomic library was constructed from southern flounder Paralich-thys lethostigma in order to evaluate the level of genetic diversity and population structure. In the first batch, 45 positive clones were successfully sequenced and 43 sequences were found to contain repeat motifs. According to Weber(1990) classification rules, the sequences were divided into three categories; 26 perfect repeat sequences without interruptions in the runs of CA or GA dinucleotides (60.5%), 14 imperfect repeat sequences with one or more interruptions in the run of repeats (32. 6%) , and 3 compound repeat sequences with adjacent tandem simple repeats of a different sequence (6. 9%). Fourteen pairs of microsatellite primers were selected and used to analyze the polymorphism and genetic structure of the wild and cultured population of southern flounder from America. The results were shown as follows: two loci were monomorphic, and 12 loci were polymorphic, inwhich 7 loci showed high polymorphic levels judged by PIC value (Pic>0. 5). The averages of observed and expected heterozygosities of the 12 polymorphic loci were 0. 36 and 0. 57, respectively. In addition, a total of 90 alleles (50 effective alleles) were detected at 12 loci in total samples. The Hardy-Weinberg equilibrium ( HWE) analysis showed that 9 loci deviated from genetic equilibrium to different extent in the two populations and heterozygote deficiency occurred in total samples (Fzs>0). The level of genetic diversity in the wild population was significantly higher than that of the cultured population, and the genetic distance between two populations was 0.111 5, which indicated that the genetic differentiation between two populations was quiet weak. Twelve polymorphic microsatellites were identified in this study, and these microsatellite loci would be usful to analyze genetic diversity, and protect germ plasm resource of southern flounder.%采用磁珠富集法筛选适合漠斑牙鲆遗传多样性分析的微卫星分子标记.筛选共获得43条序列,其中完美型26个,占60.5%;非完美型14个,占32.6%;复合型3个,占6.9%.选取其中14对特异性好且扩增效率高的微卫星引物,对采自美国北卡罗来纳州沿海的漠斑牙鲆野生群体和养殖群体进行遗传多样性及遗传结构比较分析.研究结果表明,12对引物的扩增产物具有多态性,其中7个位点为高度多态(PIC>0.5).两个群体中共检测到90个等位基因.12个多态性微卫星位点在两个群体中的平均观察杂合度(Ho)和期望杂合度(He)分别为0.36和0.57.9个位点在整个群体中呈现出不同程度的偏离遗传平衡(P<0.05),且偏离平衡的位点均表现为杂合子缺失(Fis>0).野生群体和养殖群体间的遗传距离为0.111 5,群体间的遗传分化微弱(Fst=0.043 8).

The purpose is to provide information for it's researching, breeding and promotion.%漠斑牙鲆(Paralichthys Lethostigma)是我国2002年从美国引进的一种新型海产鱼类,其肉质细嫩,营养丰富,具有很高的食用价值,是一种极具开发价值的鱼类。本文在国内现有的对漠斑牙鲆研究的基础上针对其特征进行了说明,从苗种的选择、优化,饵料,池塘养殖,痍病防治等方面对漠斑牙鲆的养殖技术进行了阐述,另外还结合其生物学特性和养殖的经济效益对漠斑牙鲆的推广前景进行分析,意在为漠斑牙鲆在我国研究、养殖及推广提供参考资料。
