    • 摘要: 目的:建立同时测定人血浆中奥卡西平(OXC)及其活性代谢物10,11-二氢-10-羟基卡马西平浓度的实验方法.方法:以阿普唑仑为内标,血浆样本采用甲基叔丁基醚萃取,以RP-HPLC测定,使用KromasilTMC18(150mm×4.6mm,5μm)色谱柱,流动相为CH3CN-H2O(45:55,V/V),流速1.0mL/min,检测波长220nm,柱温40°C,灵敏度为0.01AUFS.结果:OXC、MHD血浆浓度分别在0.70~11.2 μg/mL、2.40~38.40μg/mL范围内线性关系良好,最低检测限分别为0.35 μg/mL、1.20 μg/mL;两者低、中、高浓度方法平均回收率分别为83.23%~103.17%、87.99%~116.07%;日内RSD(n=3)、日间RSD(n=5)均低于10%.结论:该方法灵敏、准确、简便、快速,可用于OXC与MHD的临床治疗药物监测(TDM)与药动学研究.

    • 摘要: 目的:建立快速、灵敏的高效液相色谱-串联质谱(HPLC-MS/MS)方法同时测定人血浆中替诺福韦艾拉酚胺(TAF)及其活性代谢产物替诺福韦(TFV)的浓度,并研究该药物在人体内的药动学特征.方法:采用Capcell pake ADME色谱柱(75 mm×2.1 mm,5 μm),以0.1%甲酸水溶液-甲醇为流动相,梯度洗脱,流速0.4 mL·min-1,进样量5 μL;采用多反应监测(MRM),选择质荷比(m/z)477.3/346.1(TAF)、288.2/176.0(TFV)、484.3/346.1(内标1:TAF-d7)及294.2/182.0(内标2:TFV-d6)进行检测.结果:TAF与TFV质量浓度分别在0.1~400 ng·mL-1及0.4~20 ng·mL-1范围内线性关系良好,定量下限分别为0.100 0ng·mL-1及0.400 0 ng· mL-1,日内、日间精密度均小于9.0%,准确度分别为94.1%~99.2%和95.5%~112.8%.健康中国人服用富马酸替诺福韦艾拉酚胺片25mg后,TAF主要药动学参数:Cmax=(207.97±61.08) ng· mL-1,T1/2=(0.54±0.15)h,AUC0-∞=(126.07±42.67)h·ng·mL-1;TFV主要药动学参数:Cmax=(8.76±1.46)ng·mL-1,T1/2=(29.47±4.16)h,AUC0-∞=(193.19±45.75)h·ng·mL-1.结论:该方法适用于TAF及其代谢产物TFV的血药浓度检测,并提供药动学参数.

    • 摘要: 杀线虫海洋枝顶孢霉BH0531是以松材线虫为靶标从渤海水域分离获得的一株海洋真菌.为进一步探明其活性代谢物的性质和类型,实验采用非靶向代谢组学分析,对发酵液及菌体代谢物进行定性检测鉴定测定结果显示,发酵液与菌体共有的代谢物160种,只存在于发酵液中的代谢物87种.其中与乙酰胆碱酯酶活性相关物质有3种;促进植株生长作用相关物质有6种;抗氧化相关物质有6种;抑菌相关物质有3种.从代谢物角度为BH0531菌株的进一步开发利用提供实验依据.

    • 摘要: 研究北芪内生细菌及其次生代谢产物,旨在深入开发新资源。采用平板分离法从北芪中分离内生细菌,利用滤纸片法测定北芪内生细菌的抑菌活性。以芒柄花素及北芪生药为对照,采用HPLC法分析北芪内生细菌活性代谢物。同时对筛选出的目的菌株结合形态特征观察、生理生化试验及16S rDNA序列分析进行菌种鉴定。所得北芪内生细菌中,有31株内生细菌发酵液对11株试验菌表现出不同的抑菌活性,占总分离菌株的86.11%。HPLC分析表明,2株内生细菌发酵产物与北芪生药样品在相同保留时间内有相应的色谱峰出现,其中菌株HX8经鉴定为甲基营养型芽孢杆菌。北芪内生细菌具有较好的抑菌活性,其中内生细菌HX8发酵产生北芪活性成分或类似化合物,这为利用内生菌发酵生产北芪活性成分提供了新思路和途径。

    • 摘要: 乳酸菌作为机体代谢重要的益生菌,不仅可以提高机体对食物的转化吸收,作为机体内重金属离子的吸附剂,而且其活性代谢物可以抑制肠道内有害菌的生长,调节肠道微生物平衡,故乳酸菌在医疗保健方面的应用日益突出.本文主要阐述了乳酸菌益生方面的研究进展,对其作用机制进行了实例举证,并针对目前乳酸菌益生行业所面临的各种问题与不足,提出了对其未来研究方向的一些建议.

    • 摘要: 建立了一种定量测定比格犬血浆中氯吡格雷活性代谢物(active metabolize, AM)的液相色谱-串联质谱(LC-MS/MS)法.采血管中加入酯酶抑制剂敌敌畏,采样后立即离心处理,取上层血浆,加入2-溴-3''-甲氧基苯乙酮(MPB)进行衍生化,反应完成后直接加入含0.1%甲酸的冰乙腈沉淀蛋白,取上清液,测定氯吡格雷活性代谢物的衍生化产物(MP-AM).以乙腈-0.1%甲酸为流动相,梯度洗脱,API4000电喷雾离子源,多反应监测(MRM)模式扫描.在优化的实验条件下,获得了峰宽较窄且对称的色谱峰;MP-AM的线性范围为1~1 000 μg/L,相关系数大于0.995;生物基质效应较小,提取回收率在92.8%~95.1%之间,相对标准偏差小于8.27%.该方法的准确度和精密度均符合国家食品药品监督管理总局(China Food and Drug Administration, CFDA)的相关要求,适用于比格犬血浆中氯吡格雷的药代动力学研究.%A method of liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) method was developed for determining clopidogrel active metabolite (AM) in beagle dog plasma.LC separation was performed using water containing 0.1% formic acid and acetonitrile as mobile phases with the gradient elution at a flow rate of 1 mL/min.Quantitative analysis was achieved after the chromatographic separation on an Ascentis C18 column (5 cm×4.6 mm×5 μm).The electrospray ionization (ESI) source in positive mode and multiple reaction monitoring (MRM) were used for the detection of clopidogrel AM derivative (MP-AM).The sample pretreatment conditions such as the extraction method, the derivative reagents were optimized.The alkylating reagent 2-bromo-3''-methoxyacetophenone (MPB) was used to stabilize clopidogrel AM in blood.Blood samples were centrifuged immediately after collection to take plasma and MPB was then added.Protein precipitation with iced acetonitrile (containing 0.1% formic acid) as precipitation solvent was used for sample extraction.The post-extracted samples were analyzed by LC-MS/MS.The peaks are narrow and symmetrical.The linear range of MP-AM is 1-1 000 μg/L, and the linear correlation coefficient is more than 0.995.The method has no matrix interference.The recoveries of MP-AM range from 92.8% to 95.1% at three spiked levels of 2.4, 24 and 800 μg/L with the relative standard deviations (RSD) less than 8.27% (n=6).The intra-assay/inter-assay accuracy and precision are all acceptable.The method is simple, accurate and robust, which is suitable for research the pharmacokinetic of AM in beagle dog plasma.

    • 摘要: 目的:研究北芪内生真菌及其活性代谢产物,旨在深入开发新资源.方法:采用平板分离法从北芪中分离内生真菌,利用滤纸片法对内生真菌活性代谢物进行抑菌活性测定,以芒柄花素及北芪生药为对照,采用HPLC法对北芪内生真菌活性代谢物进行检测分析,同时对筛选出的目的菌株结合形态特征观察及rDNA ITS序列分析进行菌种鉴定.结果:在所得北芪内生真菌中,有25株内生真菌发酵液对11株试验菌表现出不同的抑菌活性,占总分离菌株的83.33%;HPLC分析表明,1株内生真菌发酵产物与北芪生药样品在相同保留时间内有相应的色谱峰出现,其中菌株HZ13经鉴定为链格孢属菌(Alternaria alternata).结论:北芪内生真菌的发酵液具有较好抑菌活性,其中内生真菌HZ13发酵产生北芪活性成分或类似化合物,这为利用内生菌发酵生产北芪活性成分提供了新思路和途径.%Objective:To investigate the endophytic fungi from Astragalus membranaceus (Fisch.) Bge.and their secondary metabolites in order to find novel resources.Methods:Endophytic fungi were isolated from Astragalus membranaceus (Fisch.) Bge.by using the method of plate cultivation,and the antimicrobial activities of their secondary metabolites were determined by filter paper method.Meanwhile,with formononetin and Astragalus membranaceus (Fisch.) Bge.real samples as reference substances,the secondary metabolites of these endophytic fungi were analyzed by HPLC,and the target endophytic fungi were identified by rDNA ITS sequence homology analysis combined with their physiological and biochemical characteristics.Results:The fermentation broths of 25 strains of endophytic fungi (83.33% of the total isolated strains) showed different antimicrobial activities,and the HPLC analysis indicated that there were the same corresponding chromatographic peaks appeared in the fermentation broths of HZ13 as its host plants at the same retention times,then the HZ13 strain was identified as Alternaria alternata.Conclusion:Diverse antimicrobial natural products exist in the secondary metabolites of endophytic fungi from Astragalus membranaceus (Fisch.) Bge.,and HZ13 can produce similar or new active metabolites as its host.These findings provided a new idea and approach for using endophytic HZ13 to ferment and produce the components of Astragalus membranaceus (Fisch.) Bge.

    • 摘要: 经过近十年的发展,合成生物学研究的对象从单细胞的生物元件和装置的研究,逐渐过渡到多细胞的复杂体系.植物合成生物学被称为合成生物学研究的“下一篇章”.从复杂而多样的植物代谢切入进行植物合成生物学研究,有助于人类在更复杂的层面上理解生命运行的本质规律,及更深入地认知复杂人造生命的设计和构建的科学及工程原理;也有望在药用植物活性代谢物的合成生物学设计和创新生产方面实现突破.本文综述了该领域的国内外进展,并提出新本草计划的研究设想,通过基于合成生物学的药用植物活性代谢物研究,使数千年传统的本草学研究焕发新生.%In the last decade,synthetic biology research has been gradually transited from monocellular parts or devices toward more complex multicellular systems.The emerging plant synthetic biology is regarded as the "next chapter" of synthetic biology.The complex and diverse plant metabolism as the entry point,plant synthetic biology research not only helps us understand how real life is working,but also facilitates us to learn how to design and construct more complex artificial life.Bioactive compounds innovation and large-scale production are expected to be breakthrough with the redesigned plant metabolism as well.In this review,we discuss the research progress in plant synthetic biology and propose the new materia medica project to lift the level of traditional Chinese herbal medicine research.
