    • 摘要: 为探讨虾夷扇贝横纹肌在冷藏期间微观结构和品质的变化,挑选活力良好的虾夷扇贝去壳,取横纹肌部分进行冷藏,分析冷藏期间其微观结构、ATP及其关联物含量、蛋白盐溶解性及肌原纤维蛋白ATPase活力的变化规律.结果显示:扇贝横纹肌在冷藏5 d后收缩明显、质地变硬、汁液流失严重,肌细胞中大量存在的线粒体发生了空泡样变,但横纹肌微观结构仍保持完整,经十二烷基硫酸钠-聚丙烯酰胺凝胶电泳检测后未观察到蛋白的降解.冷藏5 d后横纹肌中ATP含量仍为1.07μmol/g,肌原纤维蛋白Ca2+-ATPase活力从0.41μmol/(mg·min)升高到0.50μmol/(mg·min).横纹肌肌肉蛋白在1.0 mol/L NaCl溶液中的溶解度始终仅约为60%,在匀浆液中添加ATP或在均质前溶液中添加乙二醇双(2-氨基乙基醚)四乙酸螯合Ca2+,可将横纹肌在0.4 mol/L NaCl中的蛋白溶解度从25%提高至70%.以上结果说明横纹肌硬化过程中肌动蛋白和肌球蛋白的结合能够激活肌球蛋白的ATPase活性,并对横纹肌盐溶解度产生影响.

    • 摘要: 猪旋毛虫病是由旋毛虫成虫寄生于猪的小肠(终末宿主),幼虫寄生于横纹肌(中间宿主)而引起的人畜共患病。—、病原线形动物门线虫纲中毛首目、毛形科的旋毛形线虫。虫体很小,前细后粗,白色,雌虫长1.4-1.6毫米,雄虫长3-4毫米,肉眼勉强可见。幼虫(肌旋毛虫)长1-1.5毫米,在肌纤维膜内形成包囊,呈椭圆形,包囊很小,只有0.25-0.5毫米,囊内虫体呈螺旋状蜷缩。

    • 摘要: 目的:本文主要是对食用小龙虾所致横纹肌溶解症的护理进行经验总结.方法:选取本院2016年6月到2018年5月所收治的96例因食用小龙虾所致横纹肌溶解症患者,按照时间顺序将其划分为观察组和对照组,分别采用基础护理措施以及全面护理措施,通过统计学对比分析两者患者的护理满意度和并发生发生概率,进而对因使用小龙虾所致横纹肌溶解症患者的护理经验进行总结.结果:通过对比分析发现,对照组患者的患者满意度和并发症发生率都要优于观察组,因此,在治疗因食用小龙虾所致横纹肌溶解症患者方面,应当加强全面护理干预的推广.

    • 摘要: In this study, the histomorphology, protein profile and myofibrillar ( Mf) gelation at low temperature were analyzed in muscle of Manila clam Ruditapes philippinarum for exploring the relationship between processing characteristics and properties of muscle protein. The results showed that adductor muscle fibers were highly homon-ymous in direction, as well as, striated and smooth muscle were linear and helical columnar, respectively. Analysis of protein distribution in two types of muscle by SDS-PAGE electrophoresis revealed that the sarcoplasmic protein was focused on 23000 presented in striated muscle, while the thick filament was dissolved. Compared with striated muscle, more level of myorod was only found in smooth muscle, in which there was higher content of paramyosin, and lower content of myosin and tropomyosin, an approximate content of actin. The solubility of Mf from whole ad-ductor was depended on ionic strength and ATP. The maximum value of Ca2+-ATPase activity was observed in the middle ionic strength ( KCl concentration 0 . 30 mol/L ) , and the activity was reduced gradually as the deviation from the middle ionic strength. Proteins involved in crosslinking process contained myosin, paramyosin and tropo-myosin, with the increase in ionic strength, the more significant effect of crosslinking. The findings provide a scien-tific reference for the further study on the protein properties of edible parts from economically important shellfish.%为探讨菲律宾蛤仔Ruditapes philippinarum的加工特性与肌肉蛋白质特性的关系,对菲律宾蛤仔闭壳肌的组织形态、 肌原纤维蛋白(Myofibrillar,Mf)的分布特性,以及与其低温凝胶化相关的特性等进行了分析.结果表明:闭壳肌肌纤维具有高度同向性,横纹肌呈直线柱状,平滑肌呈螺旋柱状;利用SDS-PAGE电泳对两种类型肌肉的蛋白分布分析显示,横纹肌肌浆蛋白组分中存在相对分子质量为23000的差异蛋白,且溶出更多的粗丝蛋白,平滑肌的Mf组分中存在标志性蛋白Myorod,且副肌球蛋白含量高,肌球和原肌球蛋白含量低,肌动蛋白含量接近;整个闭壳肌Mf的溶解性依赖于离子强度和有无三磷酸腺苷(Adenosine Triphosphate,ATP)的添加;Ca2+-ATPase活性在中等离子强度(KCl浓度为0.30 mol/L)时最高,偏离中等离子强度后活性逐渐降低;参与交联的蛋白包括肌球蛋白、 副肌球蛋白、 原肌球蛋白,离子强度越高交联作用越显著.本研究结果可为探究经济贝类软体可食部位蛋白特性提供参考.

    • 摘要: To explore the protein distribution and physicochemical properties differences between striated(ST)and smooth(SM) muscle of Patinopecten yessoensis, muscle tissue sections were stained with hematoxylin and eosin and observed under microscope.Protein composition was analyzed by SDS-PAGE.The physicochemical properties were compared by salt-soluble and Ca2+-ATPase.It was found that the striated muscle fibers were thicker and denser than smooth muscle .Myorod only existed in the smooth muscle and was not present in the striated muscle.The content of paramyosin in the smooth muscle was 31%, however, it was only 11% in striated muscle.There was a complete difference between the two kinds of salt solution curves.The smooth muscle solubility was characterized by S-shaped curve, which was different from the striated muscle and showed a linear rise in the range of 0.05–1.0 mol/L salt concentration.Such differences were probably caused by paramyosin content.In addition, the optimal temperature of Ca2+-ATPase in smooth muscle myofibrillar and striated muscle myofibrillar was 35 °C and 30 °C, and the activation energy was 23.4 kJ/moL and 11.3 kJ/moL, respectively.%为探讨虾夷扇贝闭壳肌中横纹肌(ST)和平滑肌(SM)蛋白分布特征及理化性质差异.对肌肉组织进行切片、苏木精—伊红染色后,在光学显微镜下观察肌肉组织形态,SDS-PAGE分析蛋白组成,通过盐溶性和Ca2+-ATPase对理化性质进行了比较研究.研究表明,横纹闭壳肌的肌纤维更粗,肌细胞间更致密;Myorod蛋白仅存在于平滑闭壳肌中,在横纹闭壳肌中不存在;副肌球蛋白在平滑闭壳肌中含量为31%,横纹肌中仅为11%.2种肌肉盐溶曲线也完全不同,在0.05~1.0 mol/L盐浓度范围内平滑肌的溶解性呈S型曲线上升,而横纹肌呈直线型上升,这种差异可能与副肌球蛋白含量有关.此外,平滑肌和横纹肌肌原纤维蛋白Ca2+-ATPase最适温度分别为35和30°C,活化能分别为23.4和11.3 kJ/moL.
