    • 摘要: 非 小 细 胞 肺 癌(non-small cell lung cancer, NSCLC)是世界上最常见、多发的恶性肿瘤之一,约 占所有肺癌的 85%[1],其临床检测包括 8号染色体着 丝粒探针(centromeric enumeration probes for chro⁃ mosomes 8,CEP8)荧光原位杂交法与 CD45 免疫荧 光 抗 体 技 术 两 种[2]。 虽 然 两 种 检 测 方 法 应 用 在 NSCLC 检测的准确性均较高,但多以单独检测,两 者联合却鲜有报道。故本次研究将通过 CEP8联合 CD45检测 NSCLC患者血液循环肿瘤细胞(circulat⁃ ing tumor cells,CTC)检测的价值,为该病的临床诊 治提供理论与实践依据。现报道如下。

    • 摘要: 复制骨骼肌钝挫伤模型,探讨骨骼肌损伤后给予干细胞移植治疗后效果。实验兔随机分为3组:正常组(3只)、对照组(15只)、BM-MSC移植组(15只)。除正常组外均用钝性暴力法制造骨骼肌钝挫伤模型,损伤后对照组注入无细胞低糖DMEM培养液,BM-MSC组注射自体BM-MSC,并于伤后1、3、7、14、21d处死动物,进行相关检测。兔骨髓间充质干细胞CD90阳性,CD45阴性;损伤区肿胀,72h后肿胀消至实验前;模型组于损伤后1d均可见血细胞渗出,第3天炎症细胞开始浸润至7d达峰值,伤后第21天肌纤维形态基本恢复正常。

    • 摘要: 目的通过检测冠状动脉粥样硬化病灶内CD45蛋白表达水平,分析其与病灶结构变化的关系,探讨CD45在人冠状动脉粥样硬化病灶发生发展中的作用。方法收集贵州医科大学法医司法鉴定中心司法鉴定中尸体检验及组织学检查确认有冠状动脉粥样硬化病例的冠状动脉组织,根据冠状动脉病变情况将其分为对照组、单纯动脉粥样硬化组(单纯As组)和动脉粥样硬化并继发病变组(As并继发病变组),常规石蜡切片HE染色观察各组冠状动脉的组织结构,免疫组织化学染色法、蛋白免疫印迹法及实时荧光定量PCR检测CD45蛋白的表达分布及水平,分析CD45表达水平与粥样硬化病灶结构变化的关系。结果与对照组比较,有动脉粥样硬化及并发继发病变的冠状动脉斑块内纤维帽厚度变薄,病灶厚度、坏死灶厚度及血管腔面积增加(P均<0.05)。与对照组比较,有动脉粥样硬化及并发继发病变的血管组织CD45蛋白表达显著升高(P<0.05),且As并继发病变组显著高于单纯As组(P<0.05)。与对照组比较,有动脉粥样硬化及发生继发病变的冠状动脉病灶内CD45的表达显著升高(P<0.05),且As并继发病变组显著高于单纯As组(P<0.05),CD45阳性蛋白表达主要分布于斑块肩部和底部的白细胞(棕黄色着色)。与对照组比较,有动脉粥样硬化及并发继发病变的血管组织CD45 mRNA表达显著升高(P<0.05),且As并继发病变组显著高于单纯As组(P<0.05)。病灶内CD45表达水平与病灶结构变化具有相关性。结论人冠状动脉粥样斑块内CD45分子作为炎症反应的标志物,其表达水平反映病灶内的炎症反应程度,而病灶内炎症反应程度能影响病灶结构,从而导致斑块稳定性改变。

    • 摘要: Objective To investigate the role of microglial activation in choroidal neovascular membranes(CNM) of age-related macular degeneration (AMD) patients.Methods Paraffin-embedded sections of CNM excised from 11 patients with wet AMD were collected.The microglial of sections were analyzed by immunohistochemistry with antibodies against HLA-DR antigen,CD 45 or CD 68.CNM were also dual labeled with CD 68 and vascular endothelium growth factor (VEGF).Results In inflammatory active CNM,abundant microglial cells were appeared characteristically amoeboid,with hypertrophic cell body and shorter process.In inflammatory inactive CNM,most of positive labeled microglial cells showed ramified,with small cell body and numerous nodular and slender processes.Double labeling study demonstrated that VEGF was mainly expressed in microglial cells.Conclusion Microglial are involved at different stages of the disease.Microglial activation is a prominent feature of the disease process.%目的 探讨小神经胶质细胞在湿性年龄相关性黄斑变性(AMD)患者脉络膜新生血管膜(choroidal neovascular membranes,CNM)形成中的作用.方法 选取11例湿性AMD,利用其手术切除的CNM,石蜡包埋后切片,分别用抗HLA-DR抗原、CD 45、CD 68的抗体对小神经胶质细胞进行免疫染色.同时用抗CD 68抗体和抗血管内皮生长因子(VEGF)抗体对CNM进行双重免疫染色.结果 在炎症活跃的CNM中,可见大量阿米巴样小神经胶质细胞,胞体肥大,突起短小,并可见新鲜出血.在炎症非活跃的CNM中,可见分叉较多的小神经胶质细胞,胞体小、突起呈细长的结节状且数量多.双染的结果显示VEGF主要表达在小神经胶质细胞.结论 小神经胶质细胞存在AMD病程的各个阶段,小神经胶质细胞活化是该病的重要病理特征.

    • 摘要: 目的 探讨dl-扁桃酸热敏凝胶(dl-mandelic acid thermosensitive in situ gel,MA Gel)对兔阴道黏膜的刺激性及炎症反应情况.方法 将40只雌兔随机分为基质对照组、MA Gel低剂量组(10g/L)、MA Gel中剂量组(20 g/L)、MA Gel高剂量组(40 g/L)和市售壬苯醇醚凝胶(nonoxynol 9,N-9 Gel)组,每组8只,阴道给药2.5 mL,qd,连续10d.在给药前、给药第4日以及末次给药后24 h与72 h,收集基质对照组、MA Gel高剂量组和N-9 Gel组阴道灌流(cervicovaginal lavage,CVL)液,流式细胞仪计数CVL中炎性细胞和中性粒细胞数目,并对CD45+细胞进行分析.取阴道组织做组织病理切片,免疫组织化学检测阴道黏膜内CD45的表达.结果 MA Gel高剂量组CVL液中炎性细胞、中性粒细胞以及CD45+ 细胞数目显著低于N-9 Gel组(P=0.036).MA Gel各剂量组与基质对照组相比,阴道黏膜CD45+表达无显著上升,主要表达在黏膜上皮层.结论 dl-扁桃酸热敏凝胶仅引起家兔阴道黏膜较轻微的炎症反应,未诱导局部炎性细胞的明显活化,安全性较好,具有研发成为阴道外用避孕药的前景.%Objective To study the vaginal mucosal irritation and inflammatory response ofdl-mandelic acid thermosensitive in situ gel (MA Gel) in rabbits.Methods Forty female rabbits were randomly divided into 5 groups:matrix control group,MA Gel low-dose group (10 g/L),MA Gel middle-dose group (20 g/L),MA Gel high-dose group (40 g/L),and nonoxynol 9 (N-9) Gel group.Gels were administered to rabbits through vagina at 2.5 mL once a day for 10 consecutive days.Cervicovaginal lavage (CVL) fluid of matrix control group,MA Gel high-dose group and N-9 Gel group was collected at 4 different time-points (before administation,4th day of administration,24 h and 72 h after last administration).The number of inflammatory cells,neutrophilic granulocytes,and CD45+ cells in CVL was detected by flow cytometry.The vaginal tissue was taken for histopathological slices and immunohistochemistry was used to detect CD45+ cells in vaginal mucosa.Results The number of inflammatory cells,neutrophilic granulocytes and CD45+ cells of MA Gel high dose group was significantly lower than that of N-9 Gel group (P=0.036).Compared with the matrix control group,the number of CD45+ cells in all MA Gel groups showed no significant increase,and the CD45+ cells mainly located in the vaginal epithelium.Conclusion MA Gel caused far lower vaginal mucosal irritation in rabbits than N-9 Gel,and it was not likely to induce significant local activation of inflammatory cells.Owing to its promising pharmaceutical effects and safety profile,MA Gel has the potential to be developed as a novel spermicide.

    • 摘要: 目的 探讨慢性乙型肝炎患者外周血中T细胞受体重排删除环(TRECs)含量以及CD4+C D45RA+、CD4+CD45RO+、CD8+CD45RA+、CD8+CD45RO+T淋巴细胞亚群表达的临床意义.方法 流式细胞技术与实时荧光定量PCR技术分别检测32例轻度慢乙肝患者、42例中度慢乙肝患者、38例重度慢乙肝患者以及30例健康对照人群外周血中TRECs的含量以及CD4+CD45RA+、CD4+CD45RO+、CD8+CD45RA+、CD8+CD45RO+T淋巴细胞亚群表达.结果 慢性乙型肝炎患者中轻、中、重组以及健康对照组间相比CD3+/CD4+/CD8+、CD4+CD45RA+、CD4+CD45RO+变化均无明显差异(P>0.05);CD8+CD45RA+均有不同程度的下降,尤其在重度组中下降最为明显,具有统计学差异(P<0.05),CD8+CD45RO+存在明显的升高,同样在重度组中升高最为显著具有统计学差异(P<0.05);TRECs含量慢乙肝组较正常对照组明显降低(P<0.05);在慢性乙型肝炎患者中轻、中、重慢乙肝组间TRECs含量也存在统计学差异(P<0.05).结论 TRECS是胸腺近期输出功能指标,能够反映慢性乙型肝炎近期免疫功能,外周血TRECS与CD45+T淋巴细胞亚群联合检测较常规CD4+和CD8+T淋巴细胞亚群更能全面了解慢性乙型肝炎的进展及预后.

    • 摘要: 目的 通过分析CD45阳性(CD45+)多发性骨髓瘤(MM)浆细胞的免疫表型特征及其与CD45阴性(CD45-)MM浆细胞免疫表型的区别,探讨CD45表达与MM临床特征的相关性.方法 回顾性分析192例初诊MM患者浆细胞的免疫表型,收集2个组的相关临床信息及实验室数据进行比对研究.结果 根据浆细胞CD45的表达情况将192例MM患者分为CD45+MM组(37例,浆细胞的CD45平均表达荧光强度>102)和CD45-MM组(155例).CD45+MM组浆细胞CD138、CD38、CD19、CD56、CD20、CD71、CD117、CD33的阳性率分别为83.8%、97.3%、32.4%、64.9%、21.6%、51.4%、35.1%、24.3%;胞浆κ轻链和λ轻链的阳性率均为45.9%,有8.2%不表达轻链.2个组浆细胞CD19、CD71和CD138的阳性率差异有统计学意义(P0.05).结论 CD45+MM患者浆细胞CD19和CD71阳性率明显升高,而CD138阳性率明显降低,推测其分化阶段可能处于浆细胞的偏早期.%Objective To investigate the plasmocyte immuno-phenotype characteristics of CD45-positive (CD45+) multiple myeloma(MM) and its difference with the plasmocyte immuno-phenotype characteristics of CD45-negative(CD45-)MM,and to analyze the correlation of CD45 expression with MM clinical characteristic. Methods The data of 192 MM patients were reviewed for immuno-phenotype analysis.Results According to CD45 expression,192 MM patients were classified into CD45+ MM(CD45 average fluorescence intensity on plasma cells >102) group(37 cases) and CD45-MM group(155 cases). In CD45+ MM group,the positive rates of CD138,CD38,CD19,CD56,CD20,CD71,CD117 and CD33 were 83.8%,97.3%,32.4%,64.9%, 21.6%,51.4%,35.1% and 24.3%. The positive rates ofκ andλ light chains were 45.9%. There was 8.2% with no light chain expression. The positive rates of CD19,CD71 and CD138 had statistical significance between the 2 groups (P0.05).Conclusions For CD45+ MM patients,their CD19 and CD71 positive rates increase,and CD138 positive rate decreases. CD45+ MM cells may be in the early differentiation stage of plasma cells.

    • 摘要: Objective To investigate the effect of fenbendazole on CD45R (+) B cells, IgG immune response and movement function recovery of rats with spinal cord injury (SCI). Methods Seventy⁃five adult female SD rats were randomly divided into sham operation group, model control group and fenbendazole group, 25 for each group. Routine rodent feed (containing 18% protein feed) was given in sham operation group and model control group for 4 weeks preoperatively, and fenbendazole and conventional rodents feed was given in fenbendazole group for 4 weeks preoperatively. The SCI was produced using a modified Allen method. Basso⁃Beattie⁃Bresnahan (BBB) scale was used to estimate the neurological recovery on days 1, 7, 14, 21 and 28 postoperation after SCI. The expression levels of IgG in the spinal cord tissue were evaluated using the immuno⁃histochemistry. Immunofluorescent assay was performed to identify the signal level of CD45R positive cells in the injured spinal cord. Results BBB scores in model control group and fenbendazole group were significantly lower than in control group (P<0.05) after SCI. With the extension of time, BBB scores in model control group and fenbendazole group were recovered gradually, but still lower than in control group. On the 1st day after SCI, BBB movement scores in model control group and fenbendazole group were 3.10 ± 0.29 and 3.23 ± 0.48, respectively, and no statistically significant difference between two groups. On the day 7, 14, 21, 28 after SCI, BBB scores in fenbendazole group were significantly higher than those in model control group (P<0.05). Immunohistochemistry revealed that IgG levels increased significantly after SCI. From day 7, the immune response of IgG levels in the fenbendazole group was weaker than that of model control group with the difference being statistically significant (P<0.05). Immunofluorescent assay confirmed that CD45R positive signal level increased significantly after SCI. From day 7, CD45R positive signal levels in the fenbendazole group were low⁃er than in the model control group at each time point with the difference being statistically significant ( P<0.05). Conclusion Fenbendazole pretreatment reduced the injury⁃induced CD45R⁃positive B cell signal inten⁃sity and IgG immunoreactivity at the lesion epicenter after contusive SCI, and promoted neural functional recov⁃ery after SCI.%目的:观察芬苯达唑对脊髓损伤大鼠的CD45R阳性B细胞、IgG免疫反应以及运动功能恢复的影响。方法将75只成年雌性SD大鼠随机分为假手术组、模型对照组和芬苯达唑组,每组25例。假手术组、模型对照组术前4周给予常规啮齿动物饲料喂食(含18%蛋白质的饲料),芬苯达唑组术前4周给予加入芬苯达唑的常规啮齿动物饲料喂食。用Allen法构建脊髓损伤模型,分别于术后第1、7、14、21、28天行Basso⁃Beatti⁃Bresnahan(BBB)运动功能评估,利用免疫组化检测损伤部位脊髓组织中IgG的表达水平,利用免疫荧光检测脊髓损伤部位CD45R阳性B细胞的信号水平。结果脊髓损伤后,模型对照组和芬苯达唑组的BBB评分均显著低于假手术组(均P<0.05),随着时间的延长,两组的BBB运动评分均逐渐有所恢复,但仍低于假手术组;脊髓损伤后第1天,模型对照组与芬苯达唑组的BBB运动评分分别为(3.10±0.29)分、(3.23±0.48)分,差异尚无统计学意义;而在脊髓损伤后的第7、14、21、28天,芬苯达唑组的BBB评分均高于模型对照组,差异均有统计学意义(均P<0.05)。免疫组化检查证实损伤部位脊髓标本的IgG水平显著升高,第7天开始,芬苯达唑组各个时间点的IgG免疫反应水平均低于模型对照组,差异均有统计学意义(均P<0.05)。免疫荧光检查证实脊髓损伤后损伤部位脊髓节段CD45R阳性B细胞信号水平显著升高,第7天开始,芬苯达唑组各个时间点脊髓损伤部位的CD45R阳性B细胞的信号水平均低于模型对照组,差异均有统计学意义(均P<0.05)。结论芬苯达唑预处理可降低脊髓组织中IgG的表达水平及脊髓损伤部位的CD45R阳性B细胞的信号水平,促进脊髓损伤后的神经功能恢复。
