    • 摘要: 目的 评估化学发光免疫系统在临床甲状腺素检测中的应用效果.方法 选取我院2018年6月至2019年11月区间内的100例甲亢患者、100例甲低患者和100例甲状腺功能正常者作为试验对象,将化学发光免疫法(CLIA)与酶联免疫吸附法(ELISA)、放射免疫测定法(RAI)检测结果比较.结果 使用化学发光免疫系统实测血清促甲状腺素(TSH)、游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)时,所得线性范围、准确度、总精密度均较好,标准变异系数(CV)1.58%~6.44%、灵敏度较高且方法总回收率在99.7%~104.6%;且实际样本检测结果优于ELISA法、RAI法.结论 利用化学发光免疫法对甲状腺功能异常患者进行血清中甲状腺素及其抗体检测,可显著提高检测结果准确性并缩短检测周期,具有较高的临床医学推广价值.

    • 摘要: 血清雌二醇(estradiol,E2)水平的检测是重要的临床指标和实验评价手段.由于生物种属的差异、测定目的和测定要求的不同,所选择的分析方法也不尽相同.本文对目前血清E24种常用的分析方法:色谱质谱联用法、放射免疫测定法、酶免疫分析法、化学发光免疫分析法进行综述和评价,以期为从事临床检测、临床研究、药学研究和分析方法开发的工作人员提供参考和依据.

    • 摘要: 近年来,为有效保护野生熊类种群,提高其繁殖性能,圈养黑熊的人工受精、妊娠诊断等技术也成为研究热点。目前,用于动物妊娠诊断的方法主要有:雄性动物试情法、阴道检查法、超声波诊断法、血小板计数法、放射免疫测定法(RIA)、酶免疫测定法、生殖激素诊断法及早孕因子诊断法等,其中放射免疫测定法是一种将同位素的放射测量与免疫反应原理相结合的微量分析方法。

    • 摘要: 奶牛人工授精(或配种)后,有效地进行早期妊娠诊断是促进奶牛繁殖的关键.也是加强怀孕母牛饲养管理,保证顺利分娩或对未孕母牛及时配种,缩短产仔间距。降低饲养管理成本的关键所在。生产实践中广泛应用的是直肠检查方法,尽管此法简单易行,不需特殊设备,但检出时间长,需有丰富经验的技术人员才能准确判断,影响奶牛及时配种,因此奶牛早期妊娠诊断方法的研究引起了普遍的重视。据有关资料介绍,目前现有的奶牛早孕妊娠诊断方法有十几种。但有些方法由于受各种因素的限制(如仪器、药品等)还不能有效地在生产实践中推广应用.有些方法则还处于研究阶段.有待进一步验证其可靠性和准确率。即使同一种方法,报道的结果也有一定差别。本实验从基层奶牛场的实际情况和实验室的条件考虑,选取血小板计数法;硫酸铜检测法;血清酸滴定法;放射免疫测定法等四种操作相对简单的诊断方法进行比较.筛选出快速、准确和生产现场易于操作的早期妊娠方法,为缩短奶牛产犊间距。提高奶牛繁殖率服务。

    • 摘要: Objective To explore the value of Serum fa - MG in diagnosing malignant bone tumor. Methods Serum β2-MG of 38 patients with malignant bone tumor,12 patients with benign bone tumors and 16 normal healthy adults was examined by radioimmunoassay,and comparative analysis was conducted. Results Three groups of test samples of serum β2- MG levels were (4.17 ± 0.85) mg/L,(1.82 ± 0.37) mg/L and (1.73 ± 0.40) mg/L,which showed that patients with malignant bone tumor of β2 - MG levels were significantly higher than that of benign bone tumors and normal adults,and the difference was statistically significant (P<0. 01). It was also found that the level of β2 - MG and malignant bone tumors,clinical staging,and response to treatment had a significant correlation. Conclusion Determination of serum β2 - MG can be an aid of diagnosis of malignant bone tumors and assessment of treatment,and one of the serological markers of the condition.%目的 探讨血清β2-MG检测在恶性骨肿瘤中的临床应用价值.方法 采用放射免疫测定对38例恶性肿瘤、12例良性骨肿瘤及16例正常健康成人的血清β2-微球蛋白(β2 - MG)水平进行了检测,并进行对比分析.结果 三组检测样本血清β2-MG水平分别为(4.17±0.85) mg/L、(1 82±0.37) mg/L和(1.73±0.40) mg/L,表明恶性骨肿瘤患者β2- MG水平明显高于良性骨肿瘤与正常人,有显著性差异(P<0.01);同时还发现β2 - MG水平与恶性骨肿瘤的临床分期、治疗反应有明显相关性.结论 血清β2 - MG测定可作为恶性骨肿瘤的辅助诊断及评估治疗、病情的血清学指标之一.

    • 摘要: 目的:研究重组甘精胰岛素注射液连续给药不同时间在犬体内的药动学.方法:采用125I胰岛素放射免疫分析药盒测定犬血中胰岛素水平.取犬随机分为高、中、低剂量(0.26、0.13、0.065 mg·kg-1)组,每组6只,皮下注射相应重组甘精胰岛素,每日1次,连续给药12周,分别测定给药6、12周末各组犬血中胰岛素水平,并用3p97软件计算药动学参数.结果:胰岛素检测浓度线性范围为40~1 280mU·L-1(r=0.991 7),最低定量限为40 mU· L-1,平均相对回收率为(96.34±4.91)%,RSD均<12.0%.犬血中重组甘精胰岛素的药动学参数分别为给药6周末的t1/2α:(0.89±0.40)~(1.43±1.01)h,cmmax:(2.30±1.16)~(11.69±4.26) mU·mL-1,AUC0~12h:(12.53±1.44)~(65.80±39.12) mU·h·mL-1;给药12周末的t1/2α:(0.68±0.48)~(1.18±0.83)h,Cmmax:(1.92±0.79)~(8.62±1.51) mU·mL-1,AUC1~12h:(19.36±19.04)~(60.25±17.62) mU·h·mL-1,2时间点药动学参数比较无明显差异.结论:重组甘精胰岛素短时(给药6周)与长时(给药12周)给药的药动学参数未见明显差异.%OBJECTIVE: To study the pharmacokinetics of recombined insulin glargine in beagle dogs after different time of consecutive administration. METHODS: 125Ⅰ radio immunoassay kit was used to analyze the level of insulin in blood of beagle dogs. Beagle dogs were randomly divided into high-dose, medium-dose and low-dose groups (0.26, 0.13, 0.065 mg·kg-1) with 6 dogs in each group once a day for consecutive 12 weeks. The levels of insulin in blood sample were determined after 6 or 12 weeks of treatment and the pharmacokinetic parameters were calculated by 3p97 software. RESULTS: The linear range of glargine was 40~ 1 280 rnU·L-1 (r=0.991 7), the lowest limit of quantification was 40 mU·L-1. Average recovery rate was (96.34 + 4.91)% (RSD<12.0%). The pharmacokinetic parameters of recombined insulin glargine in beagle dogs after 6 weeks of treatment were as follows: t1/2α:(0.89±0.40)~(1.43±1.01) h; Cmax;(2.3O± 1.16)~(11.69+ 4.26) mU·mL-1; AUC0-12: (12.53 ± 1.44)~(65.80± 39.12) mU·h·mL-1. The pharmacokinetic parameters of recombined insulin glargine after 12 weeks of treatment were as follows: t1/2α: (0.68 ±0.48)~( 1.18±0.83) h,Cmax:(1.92 ±0.79)~(8.62± 1.51) mU·mL-1,AUC0-12b:( 19.36± 19.04)~(60.25± 17.62) mU·h· mL-1. There was no significant difference in pharmacokinetic parameters after 6 weeks and 12 weeks of treatment. CONCLUSION: The pharmacokinetic parameters of recombined insulin glargine have no significant difference after short-term (6 weeks) and long-term (12 weeks) treatment.

    • 摘要: 目的:观察毕格犬接受旋转刺激后血液应激相关激素的变化及前庭功能锻炼的影响,探讨应激相关激素与运动病的关系.方法:选取运动病敏感的8只毕格犬,进行旋转刺激,诱发运动病,测定呕吐潜伏期.用放射免疫分析法测定血浆精氨酸加压素(AVP)、肾上腺素与去甲肾上腺素水平,观察旋转刺激前后的变化.接着前庭功能锻炼3个月后,观察呕吐潜伏期的变化,并测定旋转刺激前后相应的激素水平.结果:(1)毕格犬经前庭功能锻炼后,旋转刺激引起的呕吐潜伏期显著延长;潜伏期为8.38±2.03min,功能锻炼后延长至27.75±2.25min(P<0.01).(2)前庭功能锻炼前旋转刺激前血浆AVP水平为9.34±1.24pg/mL,旋转刺激后16.14±1.20pg/mL;前庭功能锻炼后旋转刺激前血浆AVP水平为14.12±1.51pg/mL,旋转刺激后13.61±1.28pg/mL.前庭功能锻炼前,旋转刺激引起血浆AVP水平提高(P<0.05),前庭功能锻炼后,血浆AVP的基础水平升高(P<0.05),但对旋转刺激的反应明显减弱.(3)前庭功能锻炼前旋转刺激前血浆肾上腺素水平为2.31±0.08ng/mL,旋转刺激后2.42±0.11ng/mL,去甲肾上腺素水平为99.15±4.75pg/mL,旋转刺激后108.07±4.53pg/mL;前庭功能锻炼后旋转刺激前血浆肾上腺素水平为2.32±0.07ng/mL,旋转刺激后2.51±0.14ng/mL,去甲肾上腺素水平为97.45±4.58pg/mL,旋转刺激后105.35±4.14pg/mL.旋转刺激及前庭功能锻炼对血浆肾上腺素与去甲肾上腺素水平的改变,差异无统计学意义(P>0.05).结论:(1)毕格犬进行前庭功能锻炼可引起机体适应,抑制运动病;(2)前庭功能锻炼能提高血浆AVP基础水平,减弱对运动病诱发刺激的反应.

    • 摘要: 目的:测定急性白血病(AL)患者血清LDH、β2-MG水平,探讨其对AL患者疗效及预后的价值.方法:对急性白血病患者154例分别采用速率法及放免法测定血清LDH、β2-MG水平.结果:(1)随着外周血白细胞总数的增加,LDH与化疗前外周血白细胞总数呈正相关(r=0.543,P<0.01);LDH与周围血、骨髓原始细胞数无相关性.(2)随着血清LDH、血β2-MG值升高,急性白血病患者的第1次缓解率逐渐下降、半年复发率逐渐上升;(3)急淋患者的LDH水平比急性非淋患者LDH水平高.结论:血清LDH、β2-MG可作为判断急性白血病疗效及预后可靠参考指标;LDH也是反映AL肿瘤负荷的一项可靠指标.

    • 摘要: 选用4月龄伊普吕肉兔12只,雌雄各半,分别于白天(13:00)、夜晚(0:00)、饥饿24h、热应激(38°C,15min)状态下经耳缘静脉采血,以放射免疫分析法检测生长激素(GH)水平.试验结果显示,该品种兔血浆GH水平白天((0.80±0.08)μg/L)高于夜晚((0.73±0.05)μg/L);应激((0.93±0.12)μg/L)、饥饿((0.85士0.21)μg/L)状况下高于正常状态((0.77士0.07)μg/L);在夜间和饥饿状态下,雌性兔较雄性兔血浆GH水平升高;热应激时雄兔血浆GH水平稍高于雌兔,其他情况下无差异.结果提示,家兔血浆GH水平受昼夜时相的影响,可能参与应激的调节,在有些条件下种属内存在着性别差异.%A total of 12 4-month-old Hyplus rabbits, half male and half female,were chosen for following experiments.The ear vein blood was culled in the daytime (13:00) or in the night (0:00), after 24 h hunger or heat stress under 38°C for 15 min respectively,for determining their blood growth hormone (GH) levels.The results showed that the rabbit variety’s plasma GH level was higher in the daytime ((0.80±0.08) μg/L) than that at night ((0.73±0.05) μg/L).Compared to the normal conditions ((0.77±0.07) μg/L), the GH levelsin stress condition ((0.93±0.12)μg/L) and hunger ((0.85±0.21) μg/L) were higher.The blood plasma GH levels of female rabbits were higher than those of male rabbits at night and in hunger, and the blood plasma GH levels of male rabbits were slightly higher than those of female rabbits under heat stress.No differences were observed in other situations.

    • 摘要: 使用放射免疫法(RIA),以甲状腺激素(T3、T4)和雌激素(E2)为检测指标,将不同生长发育阶段转基因克隆羊的激素水平与同龄非转基因羊作比较,从激素水平上分析转基因和克隆是否会对动物生长发育产生影响.结果表明:转基因克隆羊的激素浓度与同龄非转基因羊在未成熟时T3、E2无显著差异,成熟时T3、T4、E2无显著差异,仅未成熟时T4有显著差异(P≈0.02).可见,转基因克隆羊与非转基因羊生长发育相关激素水平无显著差异,从激素水平初步否认了转基因和克隆对动物生长发育的影响.%The thyroid hormones(T3 and T4)and estrogen(E2)were determined by radioimmunoas-say(RIA)among both transgenic cloned goats at different growth stages and the same-age non-trans-genic goats in order to study the effect of transgenosis and cloning on the animal growth and development. The results showed that there were not significant differences(P>0.05)in the T3 and Ej levels between the transgenic cloned goats and the same-age non-transgenic goats at immature stages,and also there were not significant differences in the T3, T4 and E2 levels between both groups at mature stage,though there was a significant difference in the T4 level between both groups at immature stages (P≈0.02). It could be concluded that there were not significant differences in the growth-related hormone levels between transgenic cloned goats and non-transgenic goats,preliminarily denying the influence of transgenosis and cloning on the animal growth and development in terms of hormone levels.
