    • 摘要: AIM To explore the protective effects of Huoxue Kangnao Granules (Astragali Radix,Morindae officinalis Radix,Puerariae lobatae Radix,etc.) on cerebral ischemia-reperfusion injury in rats.METHODS The rat model for middle cerebral artery occlusion (MCAO) was established,SD rats were randomly divided into sham operation group,model group,positive control group,Huoxue Kangnao Granules low-,middle-and highdose groups.Nerve function score was tested.Volume of cerebral infarction was calculated by TTC staining.SOD,MDA,6-keto-PGF1α,TXB2,IL-1 β and TNF-α were detected by ELISA.The expression levels of MMP-9 and ICAM-1 were detected by RT-PCR.To observe the influence of Huoxue Kangnao Granules on platelet aggregative function,clotting time was analyzed by automatic blood coagulation analyzer,plasma T-PA and PAI activities were determined by ELISA,and hemorheology indices were detected by automatic biochemical analyzer.RESULTS Huoxue Kangnao Granules could significantly decrease cerebral infarction volume,improve nerve function,reduce MDA content,increase SOD activity;increase 6-keto-PGF1 α level,reduce TXB2 content;decrease TNF-α and IL-1β expressions;inhibit MMP-9 and ICAM-1 expressions;extend KPTT and PT;elevate t-PA activity,inhibit PAI activity;decrease whole blood viscosity,enhance erythrocyte deformation ability.Each dosage group of Huoxue Kangnao Granules had a positive correlation between dose and response.CONCLUSION Huoxue Kangnao Granules can protect cerebral ischemia-reperfusion injury in rats,whose mechanisms may be related to anti-free radical injury,anti-thrombus,anti-cerebral vasospasm,improving cerebrovascular permeability,anti-inflammatory,anti-coagulation and improving abnormal changes of hemorheology.%目的 探究活血康脑颗粒(黄芪、巴戟天、葛根等)对大鼠脑缺血再灌注损伤的保护作用.方法 建立大鼠大脑中动脉栓塞(MCAO)模型,SD大鼠随机分为假手术组,模型组,阳性对照组,活血康脑颗粒低、中、高剂量组.测试神经功能评分,TTC染色计算脑梗死体积,酶联免疫吸附(ELISA)法检测超氧化物歧化酶(SOD)、丙二醛(MDA)、6-酮-前列腺素(6-keto-PGF1α)、血栓素B2(TXB2)、白细胞介素-1β (IL-1β)、肿瘤坏死因子-α(TNF-α),逆转录-聚合酶链反应(RT-PCR)法检测基质金属蛋白酶9(MMP-9)、细胞间黏附分子-1(ICAM-1)表达水平.为了观察活血康脑颗粒对血小板聚集功能的影响,全自动血凝分析仪分析凝血时间,ELISA法测定血浆组织型纤溶酶原激活物(t-PA)和抑制物(PAI)活性,全自动生化分析仪检测血液流变学指标.结果 活血康脑颗粒能显著减少脑梗死体积,改善神经功能,降低MDA含有量,增加SOD活性;升高6-keto-PGF1α水平,降低TXB2含有量;减少IL-1β和TNF-α表达;抑制MMP-9、ICAM-1表达;延长白陶土部分凝血活酶时间(KPTT)、血浆凝血酶原时间(PT);升高t-PA活性,抑制PAI活性;降低全血黏度,提高红细胞变形能力.活血康脑颗粒每个剂量组的量效呈正相关.结论 活血康脑颗粒可保护大鼠脑缺血再灌注损伤,其机制可能与抗自由基损伤、抗血栓、抗脑血管痉挛、改善脑血管通透性、抗炎、抗凝血、改善血液流变学异常变化有关.

    • 摘要: 目的 研究维吾尔药牛舌草提取物的抗自由基作用及体内抗氧化功能.方法 采用体外分析法观察牛舌草提取物对DPPH·及超氧自由基的清除作用,并进一步针对牛舌草大孔树脂洗脱部位GZB-3及GZB-AB80的提取物,以小鼠血清SOD、GSH-Px的活性和MDA的含量变化,探讨牛舌草提取物在小鼠体内的抗氧化能力.结果 牛舌草各提取物具有体外抗自由基功能,其中牛舌草GZB-3及GZB-AB80提取物有较好的清除DPPH·及超氧自由基的作用;牛舌草提取物可显著增强小鼠血清SOD、GSH-Px活性,降低MDA含量,其中GZB-AB80提取物有较好的体内抗氧化能力.结论 体内外抗氧化实验表明,牛舌草提取物具有明显的抗自由基作用及抗氧化功能.%Objective To study the antioxidation and anti-free radical effects of the extract from traditional Uyghur medicine A nchusa italica .Methods The scavenging ability to DPPH · free radicals and the rate of inhibiting superoxide anion radical were used to test the in vitro anti-free radical function of the different extracts .The changes of the SOD and GSH-Px activities ,and the MDA content in mice serum were determined to study the antioxidant function in vivo of Anchusa italica different extracts .Results An-chusa italica extracts can significantly strengthen the SOD ,GSH-Px activity and reduce the MDA content in mice .A nchusa italica extracts showed a good scavenging effect on free radicals .Conclusion A nchusa italica extracts had obvious anti-oxidation and scav-enging free radical effects .

    • 摘要: 对异甘草素的合成工艺进行优化和体外清除自由基活性进行研究.通过间苯二酚、冰醋酸、对羟基苯甲酸合成异甘草素,考察物料比、温度、反应时间对产率的影响,通过紫外分光度法对其清除自由基活性进行研究.酚与酸物料比1:2.5,240 W微波辐射3 min,2,4-二羟基苯乙酮产率最高为65.75%;当醛酮物质的量比为3:1,温度控制在120~130°C,回流6 h,异甘草素产率最高为28.10%;反应时间控制20 min,异甘草素加入量为1.00 mL清除DPPH自由基能力最强,为66.4%.异甘草素具有一定的抗自由基活性.%The synthesis process of isoliquiritigenin was optimized and its activity of scavenging free radicals in vitro was studied. Firstly, isoliquiritigenin was prepared from resorcinol, acetic acid and p-hydroxybenzoic acid. The influence of molar ratio of reactants, reaction temperature and reaction time on the yield of isoliquiritigenin product was investigated. The results showed that, when the molar ratio of resorcinol and acetic acid was 1:2.5, microwave radiation was 240 W for 3 min, the product yield of 2, 4-dihydroxyacetophenone reached the maximum value, 65.75%; When the molar ratio of aldehyde and ketone was 3:1, the temperature was about 120~ 130 oC with reflux time of 6 h, the yield of isoliquiritigenin product was as high as 28.10%. Then, the activity of scavenging DPPH free radicals was the highest, about 66.4 when 1.00 mL isoliquiritigenin was added to the solution with the reaction time of about 20 min. The results indicated that isoliquiritigenin has good anti-free radicals ability.
