    • 摘要: 目的 评价慢性疲劳综合征(CFS)大鼠模型的行为学特征。方法 选取力竭游泳与慢性束缚两种方法进行长期复合刺激建立CFS大鼠模型,造模持续21 d。观察大鼠的一般状态、饮水量与摄食量、体质量变化等,进行定时检测旷场悬尾指标变化,并在实验过程中对这两种行为学检测方法进行改进。造模结束后,采用酶联免疫吸附试验(ELISA)法检测免疫球蛋白A(IgA)、免疫球蛋白G(IgG)、补体C3以及血乳酸(BLA)水平,并取大鼠双侧股四头肌组织,进行常规苏木精-伊红(HE)染色观察形态学。结果 与空白组比较,模型组大鼠体型明显减小,毛发稀疏暗淡,部分大鼠出现烦躁好斗的现象,饮水量与摄食量均存在下降趋势。旷场实验结果显示,造模21 d,模型组水平跨格数与站立次数较空白组均明显减少(P0.05)。悬尾实验结果显示,与空白组相比,模型组大鼠造模21 d挣扎次数明显减少(P<0.05),不动时间明显变长(P<0.05)。与空白组比较,模型组大鼠血清中IgA、IgG及补体C3含量明显降低(P<0.05),血乳酸含量明显升高(P<0.05)。与空白组比较,模型组大鼠股四头肌部分纤维结构破坏,胞核固缩、凝聚。结论 力竭游泳与慢性束缚方法可以成功构建CFS大鼠模型,体质量、旷场实验和悬尾实验、血清学指标以及组织形态学指标可作为检测模型成功的标准,同时对旷场悬尾实验方法进一步完善,得到可被重复验证与实践的较成熟方案。

    • 摘要: 目的:研究刺五加-云芝不同比例混合后醇提物对小鼠的抗抑郁作用,并筛选出具有最强抗抑郁活性的比例,并探讨其抗抑郁作用机理.方法:通过75%乙醇超声辅助提取有效成分,小鼠给药剂量为4.55 mg/kg,连续给药14 d;测定刺五加-云芝(1:1、1:2、1:3、1:4、1:5、5:1、4:1、3:1、2:1)不同比例醇提取物对小鼠行为绝望模型的影响.通过腹膜内注射利血平建立小鼠抑郁模型;ELISA法检测小鼠海马中单胺类神经递质5-HT、NE以及小鼠血清中促炎和抗炎细胞因子IL-1β、IL-10的含量和水平变化.结果:刺五加-云芝(1:2、1:1)组和单一实验组均能不同程度降低小鼠在行为绝望模型中的失望时间(P<0.05),抑制由利血平引起的体温下降及眼睑下垂(P<0.05),1:2组能显著提高小鼠脑中5-HT、NE及小鼠血清中IL-10的含量(P<0.01,P<0.05),下调血清中IL-1β的含量(P<0.01).刺五加-云芝1:2配比组作用效果最好.结论:刺五加-云芝配伍组合对小鼠抑郁模型有一定的作用,作用机制可能与增加抑郁模型小鼠脑中5-HT、NE及小鼠血清中IL-10的含量,降低小鼠血清中IL-1β的含量有关.

    • 摘要: 目的:探究中药制剂右归胶囊对小鼠抑郁模型的疗效,研究其抗抑郁作用机理.方法:100只健康雌性昆明小鼠,随机分为空白组、模型组、西药组(10mg/kg)、右归胶囊组(3.6g/kg).空白组给予生理盐水,模型组、西药组、右归胶囊组给予氢化可的松,四组小鼠连续造模35d,每周称量1次质量,于第15天开始给药,连续给药21d.于第14d、第35d通过悬尾实验、强迫游泳实验对各组小鼠进行行为学指标观测.结果:造模14d后,除空白组外,各组小鼠均出现明显的体质量下降;悬尾实验、强迫游泳实验各组小鼠不动时间增加(P<0.01或P<0.05),给药21d后,与模型组相比,各组小鼠抑郁行为均出现明显改善(P<0.01或P<0.05).结论:综合上述结果,右归胶囊可以改善由肾阳虚导致的小鼠抑郁行为.

    • 摘要: 目的:观察细辛水煎液及挥发油抗抑郁作用.方法:将初筛过的小鼠随机分为模型组、阳性组、细辛挥发油组,细辛水煎液高、中、低剂量组(2、1、0.5 g/mL)6组,每组小鼠10只,灌胃给药连续7 d,1次/d,第7天灌胃结束后1 h,进行行为学实验:小鼠悬尾实验(TST)、强迫游泳实验(FST)和开场实验(OFT),观察细辛水煎液及挥发油对小鼠抑郁行为的干预作用.结束后处死小鼠,用Elisa法测定小鼠脑内五羟色胺(5-HT)、谷氨酸(Glu),γ-氨基丁酸(GABA)的含量.结果:1)细辛水煎液高剂量及细辛挥发油可使小鼠在悬尾实验TST中不动时间显著性缩短(高剂量P<0.01,挥发油P<0.01);细辛水煎液中剂量及细辛挥发油可使小鼠在强迫游泳实验FST的不动时间均显著性缩短(P<0.05).2)细辛挥发油剂量组和水煎液高、中、低剂量组结果与模型组比较均能显著增加小鼠脑内GABA的含量(P<0.01);细辛水煎液高、中剂量组与模型组比较能显著增高小鼠脑内Glu的含量(P<0.05);细辛水煎液中、低剂量组与模型组相能显著增加小鼠脑内5-HT的含量(中剂量P<0.01,低剂量P<0.05).结论:首次证实细辛水煎液及挥发油具有抗抑郁作用.细辛挥发油和水煎液在小鼠悬尾模型和强迫游泳模型上均表现出抗抑郁作用,细辛水煎剂和挥发油与盐酸氟西汀作用机制有相似之处.作用机制可能与提高脑内类神经递质5-HT、Glu、GABA含量有关,尤其是与提高单胺类神经递质5-HT浓度关系更加密切.%Objective:To observe the antidepressant effect of Radix et Rhizoma Asari decoction and volatile oil.Methods:The screened mice were randomly divided into 6 groups with 10 rats in each group,respectively model group,positive control group,Ra-dix et Rhizoma Asari volatile oil group,Radix et Rhizoma Asari decoction of high,medium and low dose group(2,1,0.5 g/mL). Each group were intragastric administration for 7 d,1 time a day.1 h after the last administration,the tail suspension test(TST), forced swimming test(FST)and open field test(OFT)were performed to observe the intervention effect of decotion and volatile oil of Radix et Rhizoma Asari on depression behavior of mice.Then the mice were killed,and Enzyme linked immunosorbent assay (ELISA)was used to determine the contents of GABA,glutamate(Glu)and 5-HT in the brain of mice.Results:The high dose and the volatile oil of Radix et Rhizoma Asari could significantly reduce the immobility time of mice in TST(P<0.01,P<0.01). Radix et Rhizoma Asari decoction medium dose and volatile oil could significantly shorted the immobility time of mice in FST(P<0.05).Compared with model group,the volatile oil and decoction of high,medium and low dose group could significantly increase the content of GABA in the brain(P<0.01),and Radix et Rhizoma Asari decoction high and medium dose group could signifi-cantly increase Glu content in the brain(P<0.05).Radix et Rhizoma Asari decoction medium and low dose group could signifi-cantly increase the content of 5-HT in the mouse brain(medium dose P<0.01,low dose P<0.05).Conclusion:It is the first time to confirm that Radix et Rhizoma Asari Decoction and volatile oil have antidepressant effect.Radix et Rhizoma Asari volatile oil and decoction both shows antidepressant effect on the model of tail suspension and forced swimming in mice,whose mechanism was consistent with that of fluoxetine hydrochloride.The mechanism of action may be related to increasing the content of GABA,Glu and 5-HT,especially is closely related with 5-HT concentration.

    • 摘要: 目的:比较赤芍、白芍及其有效成分芍药苷、芍药内酯苷对小鼠的抗抑郁作用及其与NO/cGMP信号转导通路的相关性.方法:选取小鼠悬尾实验模型,给予赤芍水提物、白芍水提物,记录小鼠悬尾不动时间.采用相同实验方法给予小鼠芍药苷和芍药内酯苷,并检测小鼠大脑皮质及海马组织中NO/cGMP通路相关指标.结果:与模型组比较,2g/kg赤芍和2 g/kg、1 g/kg白芍组小鼠悬尾不动时间减少,差异有统计学意义(P<0.05).与2g/kg赤芍同剂量组比较,白芍的小鼠悬尾不动时间减少,差异有统计学意义(P<0.05).与模型组比较,20 mg/kg芍药苷组、20 mg/kg芍药内酯苷组的小鼠悬尾不动时间及NO、cGMP含量明显减少,差异有统计学意义(P<0.05);且与10 mg/kg芍药苷同剂量组比较,芍药内酯苷组的小鼠悬尾不动时间及NO含量明显减少,差异有统计学意义(P<0.05).与模型组比较,20 mg/kg芍药苷组及20mg/kg芍药内酯苷组的小鼠nNOS、GluRl mRNA表达明显减少,差异有统计学意义(P<0.05).结论:白芍具有抗抑郁作用,芍药苷和芍药内酯苷是白芍抗抑郁作用的物质基础;与白芍比较,赤芍的抗抑郁作用较弱,与其主要含有抗抑郁作用较弱的芍药苷,而不合抗抑郁作用较强的芍药内酯苷有关.

    • 摘要: 动物行为学侧重以科学和客观的实验方法研究动物的各种行为,通过分析其行为目的、行为规律、行为方式等实现对动物情绪、学习、繁殖等现象的深入研究,是现代动物模型实验研究的重要技术之一.行为学实验中以鼠类动物行为研究的最为普遍,常用的经典行为学实验有旷场实验、悬尾实验、强迫游泳实验、迷宫实验、跑台实验等,其实验结果受诸多因素影响,了解行为学实验优势和局限性可为动物模型的建立和动物行为学分析提供参考.%Ethology,one of the important technologies in modern experimental research on animal models,lays particu-lar emphasis in researching all kinds of animal behaviors by scientific and objective experimental methods.It realizesin-depth study in animal emotions,learning and breeding,via analyzing their action aims,behavior regulation and behavioral pattern.In the field of behavioral experiment,the study of rodents is the most common.Regarding open field test,tail suspen-sion test,forced swimming test,maze test,treadmill test as the classic behavioral experiment,their results are influenced by lots of factors.Therefore,comprehending the advantages and limitations of these methods can provide references for animal model establishment and animal behavior analysis.

    • 摘要: Objective To measure and analyze the behavioral changes of Rncat congenital cataract mice. Methods Normal BALB/c mice and KM mice were used as control group,and inbred and random mated Rncat congenital cataract mice were used as experimental group. Behavioral tests, including the open field test, coat-hanger test, forced swimming test,and tail suspension test,were conducted on the mice. Results Compared with the inbred Rncat congenital cataract mice,the residence time in the open field test,the immobility time in the forced swimming test and tail suspension test of the BALB/c mice, randomly-mated Rncat congenital cataract mice and KM mice were significantly different. Conclusions There are certain differences in behavioral performance between the Rncat congenital cataract mice and the other mice. Our findings may provide a useful reference for future researchers.%目的 对先天性白内障Rncat小鼠行为学变化进行检测与分析.方法 以正常的 BALB/c小鼠和KM小鼠为对照组,以近亲交配和随机交配的先天性白内障 Rncat小鼠为实验组,分别进行小鼠的行为学检测(旷场实验、衣架实验、强迫游泳实验、悬尾实验).结果 与近亲交配的 Rncat先天性白内障小鼠相比,BALB/c小鼠、随机交配的先天性白内障Rncat小鼠、KM小鼠的旷场实验停留时间、强迫游泳和悬尾实验的不动时间差异有显著性.结论 先天性白内障Rncat小鼠与其他小鼠相比行为学检测结果有一定差异,可为实验人员提供参考.

    • 摘要: Objective Preliminary study on the anti-depressant effect of compound Chaijin Jieyu tablet (CJJY). Methods Two kinds of mice models of depression were established, the mice forced swimming test and hanging tail test replication stress behavioral despair models of depression, serotonin acid and reserpine induced drug-induced depression model; detection of mouse forced swimming test and the tail suspension test in the time of immobility, serotonin acid induced depression in mice the number of head twitches, reserpine induced depression of body temperature, eyelid closure, akinesi-a difference degree of mice. Results Compared with the control group, the immobility time of 4 times and 2 times or (1 times) dose CJJY groups by the forced swimming test and tail suspension test in mice was significantly reduced (P<0.05). Compared with the control group, the head-twitching number of 4 times and 2 times dose CJJY group, in 5-HT acid in-duced head twitches test, was significantly increased (P<0.05 or P<0.01). In the experiment of depression induced by reser-pine, compared with the control group, the number of mice in the model group, which the eyelids were unable to open 1/2 and akinesia, was significantly increased. After 1h and 4 h, the body temperature of the rest mice was decreased (P<0.01 or P<0.05). Compared with model group, the number of mice in the CJJY 4 times and 2 times dose groups, which the eyelids were unable to open 1/2 and akinesia. was significantly decreased. After 1h and 4h, the body temperature of the rest mice in each group was increased (P<0.01 or P<0.05). Conclusion CJJY shows obvious anti-depression effect on all kinds of depression mouse models.%目的:初步探讨复方柴金解郁片的抗抑郁作用。方法建立两种小鼠抑郁模型,即小鼠强迫游泳实验和悬尾实验复制应激行为绝望抑郁模型,五羟色胺酸和利血平诱导药物所致抑郁模型;检测小鼠强迫游泳实验和悬尾实验不动时间,五羟色胺酸诱导的抑郁小鼠甩头次数,利血平诱导的抑郁小鼠体温、眼睑闭合、运动不能差异程度。结果小鼠强迫游泳实验和悬尾实验中,与空白对照组比较,复方柴金解郁片2倍或(1倍)剂量组不动时间明显减少(P<0.05)。在五羟色胺酸诱导的小鼠甩头实验中,与空白对照组比较,复方柴金解郁片2倍剂量组甩头次数明显增加(P<0.05或P<0.01)。在利血平诱导的小鼠抑郁实验中,与空白对照组比较,模型组小鼠眼睑不能睁开1/2和运动不能动物数明显增加,1 h后及4 h后其余各组小鼠体温均有所下降(P<0.01或P<0.05);与模型组比较,复方柴金解郁片2倍或1倍剂量组小鼠眼睑不能睁开1/2和运动不能动物数明显减少,药后1 h及4 h小鼠体温明显上升(P<0.01或P<0.05)。结论复方柴金解郁片对各种小鼠抑郁模型都有明显的抗抑郁作用。
