摘要:
目的 通过观察针刺对慢性束缚应激抑郁模型大鼠海马和额叶皮层γ-氨基丁酸A受体和B受体(γ-aminobuty ric acid A receptor and B receptor,GABAAR、GABABR)表达的影响,旨在深入揭示针刺干预抑郁状态可能存在的特殊机制和作用途径,为针刺抗抑郁之机制研究作出补充,为临床针刺治疗抑郁症提供实验依据.方法 将24只SD大鼠采用随机数表法分为空白组、模型组、模型+针刺组、模型+氟西汀组,通过慢性束缚应激结合孤养的方式建立心理应激模型,针刺干预选取"百会"(GV20)、"印堂"(GV29)、双侧"三阴交"穴(SP6),平刺进针,深度0.5~1 cm,每次20分钟,每日1次,持续28天.采用体重和糖水实验评价模型大鼠行为学改变情况,运用蛋白质印迹法(Western Blot)检测海马和额叶皮层组织GABAAR和GABABR的蛋白表达水平.结果 (1)与空白组相比,模型组大鼠体重增长值以及糖水偏爱明显降低(P<0.01);模型组大鼠海马和额叶皮层中GABAAR和GABABR蛋白含量明显降低(P<0.01);(2)与模型组相比,针刺组大鼠体重增长值和糖水偏爱均明显升高(P<0.01),同时上调海马中GABAAR蛋白含量及额叶皮层GABAAR和GABABR蛋白含量均明显升高(P<0.05,P<0.01,P<0.01).结论 针刺可提高大鼠海马GABAAR和额叶皮层GABAAR,GABABR含量,逆转慢性束缚应激大鼠抑郁状态,提示这可能是针刺抗抑郁作用机制.%Objective To observe the effect of acupuncture on the expression of GABAAR and GABABR in frontal lobe and hippocampus of rats with chronic restraint stress, and to reveal the possible mechanism and possible signaling pathway of acupuncture intervention in depressive disorder. Methods Twenty-four male SD rats were randomly divided into blank group, model group, acupuncture group and fluoxetine group,6 rats in each group. The depression rat model was established by chronic restraint stress. Rats in the acupuncture group were acupunctured at"Baihui"(GV20),"Yintang"(GV29)and"Sanyin-jiao"(SP6)at double sided,20 minutes in each time and once a day for 28 days.The body weight and su-crose preference test were used to evaluate the behavioral changes. The expression of GABAAR and GAB-ABR in hippocampus and frontal cortex was detected by western blot analysis. Results (1)Compared with normal group, body weight and sugar consumption in the model group was decreased significantly (P<0.01);and the expression of GABAAR,GABABR of hippocampus and frontal lobe in the model group was significantly decreased(P<0.01).(2)Compared with the model group, body weight and sugar con-sumption was increased significantly in group(P<0.01). The expression of GABAAR expression of hippo-campus and GABAAR,GABABR expression of hippocampus and frontal lobe in the acupuncture group was significantly increased(P<0.05;P<0.01;P<0.01). Conclusion Acupuncture can regulate the expression of GABAAR in the hippocampus and GABABR in the hippocampus and frontal lobe,can alleviate the de-pression state of rats induced by chronic restraint stress,which can suggest that this may be the mechanism of antidepressant action of acupuncture.
