摘要:
目的:利用胶原诱导性关节炎(CIA)模型小鼠,探讨去甲肾上腺素(NE)及其α1-肾上腺素受体(AR)对CIA小鼠Treg细胞的作用.方法:雄性DBA/1小鼠32只,随机分为对照组(n=8)和CIA模型组(n=24).Ⅱ型胶原(CⅡ)乳剂100μl尾根部注射DBA/1小鼠制备CIA小鼠模型,在初次免疫后第41日,用免疫荧光法检测小鼠脾脏中CD4+T与α1-AR的共定位情况;用Western blot法检测小鼠踝关节和脾脏中α1-AR的蛋白表达.分离纯化CIA小鼠脾脏中CD4+T细胞,用抗CD3和抗CD28的单克隆抗体刺激CD4+T细胞,进行细胞培养,分为未加药组和加药组,加药组用NE或α1-AR激动剂苯肾上腺素(phenylephrine)处理细胞,用流式细胞术检测CIA小鼠CD4+T细胞中Treg的细胞数;用Western blot法检测CIA小鼠CD4+T中转化生长因子-β(TGF-β)和IL-10的蛋白表达.结果:CD4+T细胞能够表达α1-AR;与对照组相比,CIA小鼠踝关节和脾脏中α1-AR的蛋白表达显著降低(P<0.01);与未加药的CIA小鼠的CD4+T细胞相比,NE加入后的CIA小鼠CD4+T细胞中Treg细胞的功能显著增强(P<0.01);α1-AR激动剂phenylephrine加入后的CIA小鼠CD4+T细胞中Treg细胞的功能显著增强(P<0.01).结论:激活CIA小鼠CD4+T细胞上的α1-AR可增强Treg细胞的功能,促进CD4+T细胞向Treg细胞方向分化,发挥抗炎作用.

目的 了解前列腺无炎症增生组织和合并炎症增生组织α1肾上腺素受体(α1-AR)的分布情况.方法 选取30例良性前列腺增生(BPH)患者,收集经尿道前列腺等离子电切术后前列腺增生组织,采用双免疫组织化学法,观察前列腺增生组织中α1-AR 3种亚型的染色光密度(OD)和切片组织炎症细胞的浸润情况.结果 12例前列腺标本切片未发现炎症细胞浸润,18例前列腺标本切片有炎症细胞浸润.前列腺无炎症增生组织α受体亚型α1A-AR、α1B-AR、α1D-AR的OD值与合并炎症增生组织比较,差异无统计学意义(P>0.05).结论 前列腺无炎症增生组织与合并炎症增生组织的α受体亚型α1A-AR、α1B-AR、α1D-AR含量无差异.%Objective To investigate the distribution of alpha 1 adrenergic receptor (α1-AR) in benign prostatic hyperplasia tissues with or without inflammation after transurethral plasmakinetic resection of prostate (PKRP).Methods The prostatic tissues of benign prostatic hyperplasia were collected from 30 patients after PKRP.The distribution of α1-AR was measured by immunohistochemistry using specific monoclonal antibodies of α1-AR subtypes.The inflammatory cell infiltration of prostatic tissues was observed at the same time.Results There were no inflammatory cells in 12 prostatic tissues.There were inflammatory cells in 18 prostatic tissues.The mean optical density (OD) of α1A-AR,α1B-AR and α1D-AR in the benign prostatic hyperplasia tissues was not statistically different between the non-inflammation group and the inflammation group (P > 0.05).Conclusions The content of α1A-AR,α1B-AR and α1D-AR in the benign prostatic hyperplasia tissues with inflammation is not significantly different from that in the benign prostatic hyperplasia tissues without inflammation.

目的 观察α1-肾上腺素受体在豚鼠前列腺Cajal间质细胞(ICCs)上的表达,并探讨其生理意义.方法取豚鼠前列腺组织冰冻切片以及体外培养的豚鼠前列腺细胞,使用c-Kit标记ICCs,α肌动蛋白(α-actin)标记平滑肌细胞,分别进行c-Kit与α1-肾上腺素受体、α-actin与α1-肾上腺素受体配对的免疫荧光双标记染色.结果 豚鼠前列腺组织及分离培养的c-Kit阳性ICCs细胞膜和细胞浆中均表达α1肾上腺素受体,同时α-actin阳性平滑肌细胞上也有α1-肾上腺素受体表达,但在组织切片中ICCs上α1肾上腺素受体的表达显著强于平滑肌细胞.结论 表达α1-肾上腺素受体的前列腺ICCs作为交感神经的功能靶细胞在前列腺平滑肌活动调控中可能发挥重要的作用.%Objective To observe the expression ofα1-adrenoceptor in the interstitial cells of Cajal in guinea pig pros-tate, and explore it,s possible physiological function. Methods The guinea pig prostate slices and cells were cultured. c-Kit was used to distinguish ICCs,andα-actin was used to label smooth muscle cells. c-kit andα-actin were respectively dou-ble-labeld withα1-adrenoceptor immuno histochemistry. Results Immunohistochemistry showed that c-kit andα1-adre-noceptor were coexpressed in ICCs of c-kit positive guinea pig prostate slices. Andα1-adrenoceptor was expressed inα-ac-tin positive smooth muscle cells. Theα1-adrenoceptor was more highly expressed in ICCs than that around smooth muscle cells in guinea pig prostate slices. Conclusion The ICCs that expressα1-adrenoceptor are the target cells of sympathetic nerve and might play an important role in adjusting the activity of smooth muscle cells in guinea pig prostate.

文章文围绕α1-肾上腺素受体的纯化及制备展开研究,将细胞经复苏后,以含有抗生素200μg/mLG418和10% FBS的DMEM溶液为培养液,在37°C和5% CO2的培养箱培养,获得大量α1-AR高表达细胞,并获得该受体,与文献通过亲和色谱法得到的该受体相比,具有纯度高、省时、省力的特点,能为后续的受体生物色谱模型的建立提供有利的保障,并且为进一步采用亲和色谱法进行受体纯化提供了足量的细胞.

镁在心血管疾病中具有保护作用,给予镁剂治疗可减少心律失常的发生.但镁不同于其他抗心律失常药,镁通过复合于其他离子通道,改变其方式而发挥作用,且在免疫缺陷中起到第二信使的作用.而α1肾上腺素受体可刺激心肌细胞释放镁离子,介导镁离子转运,但其在心律失常发生、发展中的具体机制尚不清楚.明确镁离子相关发病机制,指导临床治疗甚为重要.

目的:探讨高血压患者血清中抗α1肾上腺素受体自身抗体(autoantibodies against α1-adrenergic re-ceptor,α1-AAs)拟去甲肾上腺素的激动样效应及胞内信号分子活化机制。方法:培养大鼠主动脉平滑肌细胞。将收集的原发性高血压病人血清进行免疫亲和层析法纯化,ELISA检测其滴度后以1∶80刺激细胞,并设立不同的处理组。将α1-AAs刺激大鼠主动脉平滑肌细胞后,经Western blotting及免疫荧光方法分析胞内核因子κB (nuclear factor κB, NF-κB)信号分子激活及表达状况。结果: Western bltting 显示α1-AAs 刺激组中胞内核因子NF-κB (p50)表达较空白对照组明显增强,且可被α1受体阻滞剂哌唑嗪和NF-κB拮抗剂PDTC明显抑制(P<0.05)。免疫荧光实验中,α1-AAs组荧光强度显著高于空白对照组和α1-AAs+哌唑嗪组(P<0.01)。结论:高血压患者血清中α1-AAs能够致大鼠主动脉平滑肌细胞NF-κB活化。%AIM:To study the effects of autoantibodies against α1-adrenergic receptor (α1-AAs) isolated from the hypertensive patients, which showed the agonist-like activity similar to norepinephrine , on the signal mechanism of vas-cular smooth muscle cells (VSMCs) isolated from rat thoracic aorta.METHODS:Rat VSMCs were cultured and identi-fied.The serum of hypertensive patients was purified by immunoaffinity chromatography .The autoantibodies were detected by ELISA and used to activate the cells with the titer of 1∶80.The total protein was extracted and the expression of NF-κB in different treatment groups was detected by Western blotting .Meanwhile, the activation of NF-κB in the nucleus was ana-lyzed by immunofluorescence method .RESULTS: The expression of NF-κB in VSMCs was obviously higher in α1-AAs group than that in control group .Meanwhile, the expression of NF-κB was inhibited by prasozin and PDTC.The autoanti-bodies caused a significant increase in NF-κB expression in the nucleus .The fluorescence intensity in α1-AAs group was high than that in control group and α1-AAs+prasozin group (P<0.01).CONCLUSION:Theα1-AAs from hypertensive patients increase NF-κB expression in rat VSMCs.
