Objective:To explore the effect of soyasaponins on IL-6 of blood,urine and kidney of diabetic rats.Methods:Wistar male rats,the experimental animal were randomly divided into three groups,control group,diabetic model group and SS treatment group,the rats in three groups were given daily saline,daily saline,SS(10 mg/kg) orally three month,the day before the animal were killed with metabolic cages to collect 24 h urine,and then the next day were sacrificed blood and kidney tissue.ELISA assay blood,urine and kidneys of IL-6 protein levels; using real-time PCR method to detect IL-6 mRNA levels of kidney; HE staining and immunohistochemical staining detected renal tissue pathological changes and the expression of IL-6.Results:The model rats blood,ufine and kidney protein levels of IL-6,mRNA levels and immunohistochemical staining intensity was significantly higher than that in the control group and the SS administration group(P < 0.01),SS administration group was slightly higher than in the control group,but no statistically significant(P > 0.01) ; HE staining showed a clear structure of nomal rat kidney glomerular and tubular shape rules; clear of the SS dose group rat kidney glomerular and tubular shape more rules ; model of kidney tissue structure was not clear glomerular and tubular shape irregular.Conclusion:Soybean saponin to reduce diabetic rats blood,urine and kidney of IL-6 levels,suggesting that soy saponins a protective effect on the kidneys of diabetic rats,IL-6 may be involved in the pathogenesis of diabetic nephropathy and pathological changes of the process.%目的:探讨大豆皂甙对糖尿病大鼠血、尿及肾脏IL-6的影响.方法:以Wistar雄性大鼠为研究对象,将实验动物分为三组即对照组、糖尿病模型组和大豆皂甙给药组,三组动物分别每日给予生理盐水、生理盐水、大豆皂甙(SS,10 mg/kg)灌胃3个月,动物处死前一天用代谢笼收集24小时尿液,然后第二天处死取血液及肾组织.采用ELISA法检测血、尿和肾脏IL-6的蛋白水平;采用实时荧光定量PCR法检测肾脏IL-6 mRNA水平;采用HE染色和免疫组织化学染色检测肾组织的病理改变及IL-6的表达.结果:模型组大鼠血、尿和肾脏IL-6的蛋白水平、mRNA水平及免疫组化阳性强度明显高于对照组和SS给药组(P<0.01),SS给药组略高于对照组但无统计学意义(P>0.01);HE染色显示正常组大鼠肾脏结构清晰,肾小球、肾小管形状规则;SS给药组大鼠肾脏结构清晰,肾小球、肾小管形状较为规则;模型组大鼠肾组织结构不清晰,肾小球、肾小管形状较规则.结论:大豆皂甙能降低糖尿病大鼠血、尿和肾脏IL-6的水平,提示大豆皂甙对糖尿病大鼠的肾脏有一定的保护作用,IL-6可能参与了糖尿病肾病的发病及病理变化过程.
