摘要:
基于SRP(敏感-恢复-压力)模型,选取18个指标构建2010—2020年河南省生态脆弱性评价指标体系,通过主成分分析获得SRP模型各指标权重,并计算河南省生态脆弱性指数,同时对脆弱性进行评价;使用地理探测器分析模型各要素之间交互影响q值并进行要素驱动力分析.结果表明:河南省整体生态脆弱性呈西部、北部高,东部、南部低的分布特征;中部地区以郑州市为中心辐射的区域整体脆弱性等级明显高于其他区域;研究区域2010、2015、2020年3期生态脆弱性综合指数EVSI分别为2.47、2.56、2.36,整体生态环境呈转好趋势.对2010—2020年河南省生态脆弱性时空变化分析,豫西丘陵地区、伏牛山区、桐柏大别山区生态脆弱性有明显改善,而中部、北部和南阳盆地生态脆弱性升高;2015—2020年向好生态等级转移量大于2010—2015年,生态脆弱性等级向好区域主要分布于豫西地区、豫北地区、伏牛山区、桐柏山区,生态脆弱等级负向转变主要集中于郑州-洛阳-焦作城市群、黄淮海平原东部.对研究区域生态脆弱性变化时空演变的要素驱动力分析,造成河南省生态脆弱性变化空间分异的主要指标为夜间灯光、GDP、人口密度、植被覆盖度、植被净初生产力、土壤类型、水网密度和年降水,反映出诱发河南省生态环境发生变化主要因素为人类活动扰动、土壤、植被及水资源等因素,所以做好水土保持、森林植被抚育涵养水源、提升水资源保障能力、优化城市发展和布局、建立生态型城市仍是河南省生态保护工作持续努力的方向.开展河南省生态系统脆弱性研究,对中原地区环境承载能力评估、生态安全保障具有重要意义,以期为生态脆弱区生态恢复政策的制定提供科学理论依据.

摘要:
目的探究雷公藤红素(celastrol,CEL)通过靶向抑制STAT3发挥抗结肠癌作用。方法体外培养HCT-116细胞、HepG2细胞、A549细胞,经不同浓度的CEL处理,CCK-8检测体外抗增殖活性;选择HCT-116细胞,利用Western blot检测给药前后STAT3及其上下游蛋白(JAK2、Survivin、MCL^(-1))的表达;应用表面等离子共振(SPR)以及分子对接技术分析CEL与人源STAT3重组蛋白(rhSTAT3)的结合;采用流式细胞仪检测HCT-116细胞凋亡与细胞周期阻滞情况;建立4例患者来源的结直肠肿瘤类器官(colorectal cancer organoid,CCO)模型(CCO-1、CCO-2、CCO-3、CCO-4)和1例正常结直肠组织类器官模型(colorectal normal organoid,CNO),评价化合物对CCOs和CNO的抑制作用。结果CEL对3种肿瘤细胞的半数抑制浓度分别为(A549:IC_(50)=2.37±0.02μmol·L^(-1)、HCT-116:IC_(50)=1.40±0.21μmol·L^(-1)、HepG2:IC_(50)=2.52±0.02μmol·L^(-1));HCT-116细胞经IL-6诱导后各蛋白水平均有明显的升高,差异具有统计学意义(P0.05);CEL能够阻滞HCT-116细胞周期并诱导细胞凋亡;SPR分析CEL与rhSTAT3蛋白的平衡解离常数K D=6.038×10^(-5) mol·L^(-1),具有良好的结合力,分子对接预测其结合位点位于STAT3的SH2结构域;CEL抗结直肠癌活性在CCOs上得到进一步验证,且作用强于阳性药奥沙利铂(L-OHP)。结论CEL具有显著的抗肿瘤活性,其可能是通过直接靶向抑制STAT3,进而诱导细胞凋亡、阻滞细胞周期来发挥抗结直肠癌作用。

文章以两层钢铝SPR接头为研究对象,制作不同上层材料的SPR接头,通过剖面测量和强度测试,获得接头自锁量、 "十"字拉伸强度、剪切强度与上层板厚度、抗拉强度的关系,为车身SPR连接设计提供试验依据.

摘要:
近年来,肿瘤免疫疗法取得了巨大的成功,美国FDA批准了多种PD-1/PD-L1 (programmed death-1/programmed death-ligand 1)抗体药物用于晚期恶性肿瘤治疗.然而,抗体药物作为生物大分子具有成本高、给药方式单一和较强的免疫不良反应等缺点,因此具有较低免疫风险和更好修饰性的小分子抑制剂成为值得研究的方向.天然产物中蕴涵有丰富的小分子化合物,而目前关于天然产物中PD-1小分子抑制剂的研究却鲜有报道.因此本研究以中药材人参为研究对象,通过液质联用的方法从中鉴定了9个皂苷类成分,并使用表面等离子共振(SPR)技术从中筛选出了3个具有PD-1结合活性的人参皂苷类化合物.最后使用细胞药理学方法对其中代表性化合物人参皂苷Rg1进行了PD-1/PD-L1抑制活性验证.本研究为天然产物中免疫信号通路抑制剂的研究提供了参考.

摘要:
针对某车型采用HC340LA镀锌板和6061-T6铝合金制备防撞梁总成连接强度达不到设计标准的问题,通过SPR工艺搭接了2种材料,系统研究了板材厚度、组合方式、铆钉开槽等工艺参数对接头拉剪载荷的影响.试验结果表明,接头拉剪载荷和失效位移受板材厚度影响显著且呈正相关关系,以铝板为下板时具有更佳的力学性能;对铆钉开槽处理,开槽后接头背面"铆扣"外观呈现与槽口对应的塌陷区,拉剪载荷随槽口深度增加而逐渐提高,并在槽口深度为1.2 mm时达到最大值5045 N,较未开槽试样提高11%,但槽口深度进一步增加到1.6 mm时拉剪载荷急剧降低,显示与原始试样同等级力学性能.

摘要:
为探究墨蝶呤还原酶(sepiapterin reductase,SPR)基因在草金鱼(Carassius auratus)体色形成中的作用,本研究利用半定量反转录聚合酶链式反应(Semi-QRT-PCR)技术,对spr基因在不同体色草金鱼鳞片的mRNA表达水平进行了研究.结果 显示,spr在草金鱼不同颜色鳞片中表达程度不同,且在黄色、黑色鳞片中表达量较高.本实验初步探究了spr与草金鱼体色形成的关系,旨在为进一步探究spr在体色形成中的作用提供参考资料.

摘要:
本文探索了使用局域表面等离子激元共振(localized surface plasmon resonance,SPR)对Au25(SG)18纳米团簇的荧光发射光谱进行增强的方法和技术.通过适当改进合成方法,成功制备了Au25(SG)18结构,分别使用介孔二氧化硅修饰的金纳米棒(AuNR@mSiO2)及金纳米粒子(AuNS@SiO2)复合材料和对其进行荧光增强研究.结果表明,在两个增强体系中,Au25(SG)18的荧光性质均表现为猝灭(quenching),并初步认定发生荧光猝灭现象是由于纳米团簇所处的环境以及表面配体状态的改变所导致的.

摘要:
In this paper, a high sensitive photonic crystal fiber (PCF) based surface plasmon resonance (SPR) biosensor is numerically studied. In this structure, as a plasmonic material, gold (Au) is used because of its chemical activeness. And a layer of sensing medium is used outside of the fiber to make the structure effective. Any unknown biomolecular analyte can be detected by placing or flowing it on the metal surface. Guiding properties and results are investigated using Finite element method (FEM). Results show that maximum sensitivity is 4000 nm/RIU, as well as resolution, is 2.5 × 10−5 RIU of the proposed sensor.

The thickness of each film in the multilaver films, the deposition effect of Agnanoparticles, and the processing of DNA adsorption are characterized by SPR. AFM characterization shows that DNA/3(PDDA/Ag)/4-ATP composite is uniformly and firmly distributed on the surface of gold films Compared with other sensors, gentamicin could be highly sensitively measured by DNA/3(PDDA/Ag)/4-ATP/Au sensor. There is a good linear relationship in the concentration range of 5 X 10-8~1 X 10-4 mol/L. The linear equation is found to be △OSPR-1.3521X10-SC+0.08641 Cthe correlation coefficient is 0.9983)with detection limit of 1 X 10-9mo1/L .Since such LBL assembly film is simple to prepare, the work described here provides an effective method for studying small molecule drugs on SPR.
