    • 摘要: Objective:Lower grade gliomas(LGGs),classified as World Health Organization(WHO)grade II and grade III gliomas,comprise a heterogeneous group with a median survival time ranging from 4–13 years.Accurate prediction of the survival times of LGGs remains a major challenge in clinical practice.Methods:We reviewed the expression data of 865 LGG patients from 5 transcriptomics cohorts.The comparative profile of immune genes was analyzed for signature identification and validation.In-house RNAseq and microarray data from the Chinese Glioma Genome Atlas(CGGA)dataset were used as training and internal validation cohorts,respectively.The samples from The Cancer Genome Atlas(TCGA)and GSE16011 cohorts were used as external validation cohorts,and the real-time PCR of frozen LGG tissue samples(n=36)were used for clinical validation.Results:A total of 2,214 immune genes were subjected to pairwise comparison to generate 2,449,791 immune-related gene pairs(IGPs).A total of 402 IGPs were identified with prognostic values for LGGs.The HOXA9-related and CRH-related scores facilitated identification of patients with different prognoses.An immune signature based on 10 IGPs was constructed to stratify patients into low and high risk groups,exhibiting different clinical outcomes.A nomogram,combining immune signature,1p/19q status,and tumor grade,was able to predict the overall survival(OS)with c-indices of 0.85,0.80,0.80,0.79,and 0.75 in the training,internal validation,external validation,and tissue sample cohorts,respectively.Conclusions:This study was the first to report a comparative profiling of immune genes in large LGG cohorts.A promising individualized immune signature was developed to estimate the survival time for LGG patients.

    • 摘要: Esophageal cancer(EC)was an aggressive malignant neoplasm characterized by high morbidity and poor prognosis.Identifying the changes in DNA damage repair genes helps to better understand the mechanisms of carcinoma progression.In this study,by comparing EC samples and normal samples,we found a total of 132 DDR expression with a significant difference.Moreover,we revealed higher expression of POLN,PALB2,ATM,PER1,TOP3B and lower expression of HMGB1,UBE2B were correlated to longer OS in EC.In addition,a prognostic risk score based on 7 DDR gene expression(POLN,HMGB1,TOP3B,PER1,UBE2B,ATM,PALB2)was constructed for the prognosis of EC.Meanwhile,EC cancer samples were divided into 3 subtypes based on 132 DDR genes expressions.Clinical profile analysis showed cluster C1 and C2 showed a similar frequency of T2,which was remarked higher than that in cluster 3.Moreover,we found the immune cell inflation levels were significantly changed in different subtypes of EC.The infiltration levels of T cell CD8+,B cell and NK cells were greatly higher in cluster 2 than that in cluster 1 and cluster 3.The results showed T cell CD4+infiltration levels were dramatically higher in cluster 1 than that in cluster 2 and cluster 3.Finally,we perform bioinformatics analysis of DEGs among 3 subtypes of EC and found DDR genes may be related to multiple signaling,such as Base excision repair,Cell cycle,Hedgehog signaling pathway,and Glycolysis/Gluconeogenesis.These results showed DDR genes may serve as new target for the prognosis of EC and prediction of the potential response of immune therapy in EC.

    • 摘要: Traceability system is one of the popular applications of graphic blockchains.However,there are centralisation problems and a long time for final consistency confirmation in the graphic blockchain.In addition,the blockchain system in the traceability application scenario has the problem of insufficient supervision.Therefore,a witness-based graphic blockchain consensus mechanism is proposed.In the consensus mechanism,a verifiable random function is used to screen the publishers of the unit;an SM2 threshold signature is used to sign the unit information to improve the non-repudiation of the traceability information uploaders to the unit information under the supervision of the witness.The improved consistency algorithm cancels the process of finding a stable main chain and makes relatively many nodes to participate in the consensus process.The experimental results show that the graphic blockchain using the improved witness mechanism can reduce the degree of centralisation,shorten the time for new units to reach consensus,and greatly ensure the security and scalability of the blockchain system.

    • 摘要: 系统signature是分析系统可靠性的一个强有力的工具.本文研究了多状态系统的动态signature.Signature概念被拓展到多维的情形,并将其应用于两状态元件下多态关联系统的分析,得到系统在每一状态下的联合生存函数的表达式,探究了系统在时刻的动态signature,为处于工作状态的多状态系统在时刻t接受检查,发现其处于l(t)状态并且恰好已发生了q(t)次元件失效,分别给出了两状态元件多态关联系统动态signature及其剩余寿命的表达式.
