    • 摘要: 目的:建立利用咪喹莫特诱导的银屑病样小鼠模型并结合传统小鼠银屑病样模型,探讨不同浓度的T-5224对银屑病的治疗效果.方法:将48只健康雌性BALB/C小鼠分为空白对照组、模型组、阳性对照药阿维A组、阴性对照组、实验药物低剂量组、高剂量组,共6组,每组8只,利用咪喹莫特诱导的银屑病样小鼠模型,监测各组小鼠PASI评分的动态变化、皮损厚度、并采用酶联免疫吸附法(ELISA)测定小鼠血清中相关细胞因子的含量变化,再借助传统的小鼠银屑病模型,通过苏木精-伊红染色切片观察分析,进一步探讨不同浓度T-5224对银屑病的作用效果.结果:研究发现30 mg/kg T-5224组小鼠皮损情况PASI评分明显降低;病理切片显示30 mg/kg T-5224组小鼠皮肤厚度、Baker评分(3.40±0.66)pg/mL与模型组(6.12±0.53)pg/mL相比明显好转(P<0.05),30 mg/kg T-5224组小鼠血清中银屑病相关细胞因子肿瘤坏死因子(TNF-α)含量(185.25±16.18)pg/mL明显上升(P<0.05),白介素10(IL-10)含量(88.25±6.95)pg/mL明显降低(P<0.05).30 mg/kg T-5224可有效抑制小鼠阴道上皮细胞有丝分裂情并促进小鼠尾鳞片颗粒层的形成.结论:T-5224在给药剂量30 mg/kg时,对利用咪喹莫特诱导产生的小鼠银屑病样皮损具有较好的治疗效果,且对部分反映银屑病症状的传统天然小鼠模型也有一定的治疗作用.该研究为T-5224用于银屑病的治疗提供了理论基础.

    • 摘要: 目的:探讨预先鞘内注射曲马多对福尔马林炎性疼痛大鼠脊髓c—fosmRNA及蛋白质表达的影响。方法:选取鞘内置管成功的32只雄性大鼠随机分成四组,每组8只,分别注射生理盐水(NS组),12.5ug/h曲马多(T1组),25ug/h曲马多(T2组),50ug/h曲马多(T3组)。制备福尔马林炎性疼痛大鼠模型,取模型大鼠脊髓L1~L6阶段脊髓做c—fosmRNA的半定量RT—PCR和Westernblotting检测,观察大鼠脊髓c—fos的mRNA及蛋白质表达量的变化。结果:INS组中的c—fosmRNA表达水平1.34-±0.40明显高于T2组0.69±0.40(P0.01),高于T1组1.12±0.50,但比较差异无统计意义(P0.O5);T3组0.41±0.13显著低于T2组,T2组显著低于T1组,其差异均有统计学意义(P0.01)。Ns组中的c—f0s蛋白表达水平1.20±0.50明显高于T2组0.64±0.40(P0.01),高于T1组0.98±0.38,但比较差异无统计意义(P0.05);T3组0.27±0.09显著低于T2组,T2组显著低于T1组,其差异均有统计学意义(P0.01)。结论:鞘内注射曲马多均能抑制大鼠脊髓背角c-fos的mRNA和蛋白质的表达量;不同剂量的曲马多对cfos表达量的影响不同,但剂量低时对c-fos表达无影响。

    • 摘要: 目的探讨蜂胶黄酮短叶松素-3-乙酸酯(pinobanksin-3-acetate,PB3A)处理SW480细胞后CXCL1和FOS基因表达的改变及其与PB3A抑制人大肠癌细胞增殖的关系。方法采用实时荧光定量RT-PCR和Western blot法检测PB3A(100 mg/L)处理24小时后SW480细胞的CXCL1和FOS基因转录和蛋白质表达水平。结果药物干预诱导CXCL1基因的mRNA转录水平下调,差异倍数为0.03(ΔCT比较,P<0.01),蛋白质表达强度也降低(0.24±0.03)与非干预的对照组(0.52±0.04)比较表达水平差异有统计学意义(P<0.01);药物干预诱导FOS基因的mRNA转录水平上调,差异倍数为13.64(ΔCT比较,P<0.01),蛋白质表达水平也升高(1.43±0.16),与非干预的对照组(0.58±0.07)比较表达水平差异有统计学意义(P<0.01)。结论蜂胶黄酮PB3A抗人大肠癌细胞的作用可能与其下调CXCL1基因和上调FOS基因的表达有关。

    • 摘要: 目的 检测毫米波急性辐照后SD大鼠脊髓P物质(SP)和c-fos表达的变化,探索毫米波痛觉效应的机制.方法 采用毫米波(35GHz,40W/cm2)一次性辐照SD大鼠局部皮肤0s(对照组)、30s、1min、3min,采用Real-time RT-PCR和Western blotting检测辐照后0min、5min、10min、1h、3h大鼠对应节段脊髓SP、c-fos mRNA、c-fos蛋白的表达水平.结果 毫米波辐照30s后各时间点辐照部位相应节段脊髓SP和c-os mRNA表达与对照组比较无明显差异.毫米波辐照1min后第10min辐照部位相应节段脊髓SP和c-fos mRNA表达显著上调(P<0.05,P<0.01),此后下降至对照组水平.毫米波辐照3min后第5min、10min和1h辐照部位相应节段脊髓SP和c-fos mRNA表达显著上调(P<0.01,P<0.05),于3h时间点下降至对照组水平.毫米波辐照30s及1min后各时间点辐照部位相应节段脊髓c-fos蛋白表达与对照组比较无明显差异.毫米波辐照3min后第5min和10min辐照部位相应节段脊髓c-fos蛋白表达水平显著上调(p<0.01),此后下降至对照组水平.结论 毫米波急性辐照后皮肤SP含量增加可能与毫米波辐照导致的热疼痛刺激诱导对应节段脊髓SP和c-fos的表达增加有关.%Objective To observe the expression changes in substance P (SP) and c-fos in rat spinal cord after acute millimeter-wave (MMW) exposure, and explore the mechanism of thermal hyperalgesia at the spinal level. Methods The back skin of SD rats was exposed to 35 GHz MMW (40W/cm2) for Os (control group), 30s, 1 min, or 3min. The corresponding segment of the spinal cord was taken at 0min, 5min, 10min, 1h and 3h after MMW irradiation for total RNA and protein extraction. The expressions of SP and c-fos mRNA were measured by real-time RT-PCR, and the expression of c-fos protein was detected by Western blotting. Results No significant difference was found between the control group and irradiation groups in SP and c-fos mRNA expression in the corresponding segment of spinal cord after MMW irradiation for 30s. After MMW irradiation for 1min, the SP and c-fos mRNA expressions in the corresponding segment of spinal cord increased significantly at lOmin time point, and then decreased to the level of control group. After MMW irradiation for 3min, the SP and c-fos mRNA expression in the corresponding segment of spinal cord increased significantly at 5min, 10min and 1h time points, and decreased to the level of control group at 3h. No significant change was found in c-fos protein expression in the corresponding segment of spinal cord after MMW irradiation for 30s and lmin. After MMW irradiation for 3min, the c-fos protein expression in the corresponding segment of spinal cord increased significantly at 5min and 10min time point, and then decreased to the level of control group. Conclusion The increase of SP expression in rat skin after MMW irradiation may be related to the increase of SP and c-fos expressions in the corresponding segment of the spinal cord induced by thermal pain stimulation.

    • 摘要: 背景:国外已经有学者使用斑马鱼胚胎开始进行缺氧再灌注的研究,但还没有关于c-fos基因在斑马鱼脑缺氧再灌注过程中的表达及其作用机制的报道。目的:观察缺氧再灌注后斑马鱼胚胎脑部细胞凋亡及脑组织中c-fos基因的表达情况。方法:取48hpf的斑马鱼胚胎进行缺氧实验,模拟新生儿缺氧再灌注损伤环境,通过向水中通入99.999%高纯氮气制造缺氧环境,分别经过6,12,24h的缺氧处理后,在正常氧体积分数下进行6h恢复。对照组为正常通气组(溶解氧浓度在7.0mg/L左右)。采用吖啶橙染色方法,观察不同缺氧时间对斑马鱼神经细胞凋亡的影响,同时采用实时荧光定量核酸扩增检测系统(qPCR),对c-fos基因表达情况进行定量分析,比较缺氧再灌注前后c-fos基因表达水平的变化。结果与结论:对照组脑部能检测到微量细胞凋亡,c-los基因呈低水平表达;实验组经过6,12,24h缺氧后,脑部凋亡细胞逐渐增多,缺氧24h组凋亡细胞增幅最大(尸〈0.05),c-los基因表达有不同程度升高(P〈0.05),尤其是缺氧6h后,该基因的表达上调幅度最高。结果表明缺氧会导致斑马鱼脑细胞内c-los基因表达上调,可能是导致缺氧后期脑细胞凋亡激增的机制之一。

    • 摘要: Objective To observe the effects of hypothermia on cerebral e-fos mRNA and nerve growth factor (NGF) expression following traumatic brain injury. Methods 336 adult SD rats were randomly separated to four groups. Three groups were subjected to traumatic brain injury by the Feeney's method except for sham operation group. Every group was then separated to 7 subgroups according to different decapitated time. C-fos mRNA and NGF expression was then determined respectively. Results C-fos mRNA expression in systemic hypothermia group and focal hypothermia group was significantly higher than TBI group at 4 h, 8 h, 12 h and 24 h after traumatic brain injury( P 0.05). Conclusion Both systemic hypothermia and focal hypothermia could up-regulate the expression of c-fos mRNA and NGF for the traumatic brain injury.%目的 通过对大鼠重型脑挫裂伤后体温的干预,观察不同降温方式对脑外伤后伤灶区c-fos mRNA和神经生长因子(nerve growth factor,NGF)表达的影响,了解其表达强度是否与伤后不同降温方式对脑损伤的保护机制有关. 方法 健康成年SD大鼠336只,随机分为四组(每组84只),除假手术组外各组动物均按自由落体法造成重度脑挫裂伤;每组又按处死时间分为七个亚组(每亚组12只),分别检测c-fos mRNA(6只)与NGF(6只)的表达. 结果 ①伤后4h、8h、12 h与24h,全身亚低温组与局部亚低温组c-fos mRNA表达明显高于TBI组(P<0.01),其他各时点无差异;TBI组在伤后4h、8h、12 h、24h与3d,c-fos mRNA表达均明显高于假手术组(P<0.01).②伤后4h、8h,全身亚低温组与局部亚低温组NGF表达与TBI组无明显差异(P>0.05);而在伤后12 h、24h、3d、5d与7d,两亚低温组NGF表达明显高于TBI组((P<0.05或P<0.01).③全身亚低温组与局部亚低温组在伤后各时间点c-fos mRNA和NGF表达无显著性差异(P>0.05). 结论 脑外伤后亚低温可促进c-fos mRNA和NGF表达上调,全身亚低温与局部亚低温两种不同降温方式无明显差异.

    • 摘要: 目的 探讨异丙酚对内脏痛大鼠海马c-fosmRNA表达的影响.方法 健康清洁级雄性SD大鼠30只,体重180~ 240 g,采用随机数字表法,将大鼠随机分为3组(n=10):内脏痛组(VP组)经尾静脉脉注射0.9%生理盐水;异丙酚7.5 mg/kg组(P1组)和异丙酚75.0 mg/kg组(P2组)分别经尾静脉注射异丙酚7.5和75.0mg/kg.采用直肠扩张法制备内脏痛模型.于给药前后测定痛阈,将引起大鼠腹壁明显收缩变平的最小扩张气囊内压作为痛阈;痛阈测定结束处死大鼠,提取海马组织,采用RT-PCR法测定c-fos mRNA的表达水平.结果 与VP组比较,P1组和P2组痛阈升高,海马c-fos mR-NA表达下调(P<0.05);与P1组比较,P2组痛阈升高,海马c-fos mRNA表达下调(P<0.05).结论 异丙酚可下调大鼠海马c-fos mRNA的表达从而减轻内脏痛.%Objective To investigate the effect of propofol on the expression of c-fos mRNA in hippocampus in rats with visceral pain (VP).Methods Thirty male Sprague-Dawley rats,weighing 180-240 g,were randomly divided into 3 groups (n =10 each):VP group,propofol 7.5 mg/kg group (group P1) and propofol 75.0 mg/kg group (group P2).0.9% normal saline was injected intravenously via the caudal vein in group VP.Propofol 7.5 and 75.0 mg/kg were injected intravenously via the caudal vein in groups P1 and P2,respectively.VP was produced by colorectal distension in anesthetized rats.The threshold of VP was assessed by the intra-balloon pressure which was limited to 100 mm Hg to avoid damage to intestine before and after administration.The rats were sacrificed after measurement of the pain threshold,their brains were removed and hippocampi were isolated for determination of the expression of c-fos mRNA by RT-PCR.Results Compared with group VP,the threshold of VP was significantly increased and the expression of c-fos mRNA in hippocampus was down-regulated in groups P1 and P2 (P < 0.05).The threshold of VP was significantly higher and the expression of c-fos mRNA in hippocampus was lower in group P2 than in group P1 (P < 0.05).Conclusion Propofol can reduce VP through down-regulating the expression of c-fos mRNA in hippocampus in rats.

    • 摘要: 目的 探讨大鼠严重烫伤延迟复苏后肺组织中c-fos 的表达变化及其与肺损伤的相关性.方法 雄性SD 大鼠96 只,随机分为延迟液体复苏组(DF,n =40)、即时液体复苏组(IF,n =48)和正常对照组(NC,n =8),建立总体表面积30%的Ⅲ度烫伤模型,分别于伤后1 h、6 h、12 h、24 h、72 h和168 h 取材.采用组织病理学、免疫组织化学染色与图像分析技术,观察肺组织病理改变与c-fos的表达.结果 伤后6 ~24 h DF 组大鼠肺组织病变严重,IF 组相对较轻.c-fos 的阳性表达位于肺泡上皮细胞核或细胞质,其表达强度DF 组与IF 组分别高于NC 组,且DF 组高于IF 组(P <0.05).结论 c-fos表达强度的变化与严重烫伤延迟复苏大鼠肺组织损伤相关.%Objective To explore the changes and significance of c-fos after severe burn with delayed fluid resuscitation. Methods A total of 96 male SD rats were employed as the experimental models ( TBSA 30% , grade Ⅲ ) . and they were randomly divided into 3 groups : delayed fluid resuscitation group ( DF) ,immediate fluid resuscitation group (IF) and normal control group ( NC). The lung samples were harvested at 1 ,6 ,12,24 ,72 and 168 h after bum respectively. Pathological changes of lung tissue and c -fos expression were detected hy using histopathology , immunohistochemistry and image analysis techniques .Results Posthurn 6-24 h, the pathological changes of lung tissue were more severe in DF group than those in IF group. c-fos were much higher in DF group and IF group than those in NC group (P < 0. 05 ). The increase of c-fos was observed in nucleus and cytoplasm . Conclusions The result indicates that c-fos may contribute significantly to lung injury following severe hum with delayed fluid resuscitation in rats .
