目的:探讨补阳还五汤对局灶性脑缺血大鼠神经干细胞移植后凋亡相关基因Bag-1的影响.方法:SD大鼠80只,雌雄各半,复制局灶性脑缺血大鼠模型,造模后按神经功能缺损评分进行分级,随机分为5组,每组15只,分别于移植后7 d、14 d、28 d观察神经功能评分,采用补阳还五汤超微饮片与传统饮片予以干预,于相应时间点处死大鼠后采用免疫组化法检测凋亡基因Bag-1.结果:单纯移植组、移植加中药组大鼠神经功能得到明显恢复,Bag-1蛋白表达上升显著,在各时间点(7 d、14 d、28 d)与模型组比较差异有统计学意义(P<0.05或P<0.01),模型加移植加补阳还五汤超微饮片组促进神经功能积分的恢复及上调Bag-1蛋白表达作用优于模型加移植加补阳还五汤传统饮片组,差异有统计学意义(P<0.05或P<0.01).结论:补阳还五汤可促进神经功能的恢复,上调Bag-1蛋白表达,且超微饮片组的作用较传统饮片组更加显著.%Objective: To investigate the effects of Buyang Huanwu decoction on apoptosis gene Bag - 1 after transplantation of neural stem cells in rats with focal cerebral ischemia. Methods: 80 SD rats were made into the models of focal cerebral ischemia, assessed by Neurological Function Defect Scale (NFDS), and the rats were randomly divided into five groups after modeling, with 15 rats in each group. After the transplantation of7 d, 14 d, and 28 d, interventions of Buyang Huanwu decoction were applied with respective ultramicro herbal pieces and traditional ones. The neurological deficit scores were determined and expression of apoptosis gene Bag -1 was detected by immunohistochemical staining. Results: For the rats in the simple transplantation and the transplantation group intervened by Chinese medications, their neurological function was obviously improved and the expression of apoptosis gene Bag -1 was raised significantly after the transplantation of 7 d, 14 d, and 28 d, compared to those in the model group(P <0.05, P<0.01); of which, Buyang Huanwu decoction with ultramicro herbal pieces was superior to the traditional ones(P <0.05, P <0.01). Conclusion: Buyang Huanwu decoction can promote the recovery of neurological function, up - regulate the expression of Bag -1 protein, and the ultramicro herbal pieces were better than the traditional ones.
