The present invention provides a preparation method for semaglutide, and an intermediate. The preparation method comprises the steps of: (1) reacting compound SEM110 and compound SEM120 in a solvent under an alkaline condition to obtain compound SEM130; and (2) deprotecting the compound SEM130 to form semaglutide, the reaction formula being as follows. The preparation method of the present invention is simple to operate, has mild reaction conditions, is easy to purify, has high product purity and low cost, and is suitable for industrial production.
