The present disclosure provides a human umbilical cord mesenchymal stem cell sheet comprising one or more layers of confluent human umbilical cord mesenchymal stem cells (hUC-MSC). The present disclosure is also a method of producing an hUC-MSC sheet, which is a step of culturing hUC-MSC in a culture medium on a temperature responsive polymer coated on the substrate surface of a cell culture support. The temperature-responsive polymer has a step with a lower critical solution temperature in water at 0-80 ° C; adjusts the temperature of the culture to below the lower critical solution temperature, thereby making the substrate surface hydrophilic.
