The present invention relates to a composition for promoting osteodifferentiation of stem cells comprising Cbfβ protein and a method for promoting osteodifferentiation using the same. In the present invention, human mesenchymal stem cells (MSCs) are transformed and recombinant Cbfβ protein is provided to trigger osteoblast-related processes. Efficient delivery of Cbfβ was achieved by fusing the 30Kc19 protein, a cell-penetrating protein derived from silkworm. As a result, intracellular delivery of Cbfβ-30Kc19 protein altered the transcriptional regulatory network involved in osteoblast-adipocyte lineage processes and showed enhanced osteoblast differentiation in MSCs while suppressing adipogenesis. Cbfβ-30Kc19 according to the present invention can be utilized for the treatment of bone-related diseases such as osteoporosis by regulating transcriptional regulation in MSCs.
