A tumor microenvironment (TME) responsive expression vector comprises a nucleic acid sequence encoding a synthetic promoter comprising one or more promoter responsive elements and a nucleic acid sequence encoding an immune effector gene such as a chimeric antigen receptor.The TME response vector is designed to induce expression of immune effector genes in TME and to prevent expression in normal healthy tissues.This reduces the risk of an on-target off tumor, focusing on immune activity, increasing safety and targeting normal tissue.Diagram
